Abstract. A village-scale field trial of pyrethroid-impregnated mosquito nets was undertaken in The Gambia, West Africa, in the Mandinka village of Saruja (13o13'N, 14o55'W) during July-November 1989. Nearly all the villagers possessed and used their own bednets. Anopheles gambiae is the main vector of human malaria in the area.
An experimental wash-resistant formulation of permethrin was compared with standard emulsifiable concentrate (EC) formulations of permethrin and lambda-cyhalothrin, versus placebo-treated bednets. Target concentrations of pyrethroids on bednets were permethrin 500mg/m2 and lambda-cyhalothrin 25 mg/m2. The experimental design involved random allocation of a treatment to one net per family. Whereas 68% of people questioned said they washed their nets fortnightly, observations during the 16-week trial period showed that only 4/130 (3%) of nets involved in the trial had been washed as frequently as once per month.
Early morning searches for mosquitoes under bednets (1 day/week for 16 weeks) found significantly more mosquitoes (60% An. gambiae ) in placebo-treated nets than in pyrethroid-treated nets. The numbers found with each of the three pyrethroid treatments did not differ significantly from each other. Insecticidal efficacy of the treatments was tested by bioassays using wild-caught unfed mosquitoes exposed to netting for 3min. Linear regression analysis of bioassay mortality against number of times that a net had been washed by villagers showed that nets impregnated with the wash-resistant permethrin retained their insecticidal properties better than nets impregnated with lambda-cyhalothrin or with the standard permethrin formulation.  相似文献

ABSTRACT. The cicadelloid leaf-hoppers, Eurymela distincta (Signoret) and Eurymela fenestrata (Le Pelletier & Serville), feed on the phloem sap of Eucalyptus spp. The osmotic pressure profile of haemolymph and gut fluids is consistent with a filtration mechanism based on passive osmosis. This is responsible for shunting water rapidly from foregut to hindgut in the relatively simple filter chamber. The osmotic gradient which drives the system is probably produced by active secretion of sodium and potassium into the Malpighian tubules and posterior midgut. Although the osmotic pressures in all parts of the system are higher, the mechanism is probably similar to that previously described for xylem-feeding cicadas.
The major haemolymph cations are sodium and magnesium and the concentrations of sodium and magnesium in the urine are higher than potassium. A variety of storage granules in the midgut contain calcium, phosphorus (as phosphate), ferric iron, zinc, magnesium, manganese and copper.  相似文献

Detection of aphid remains in predatory insects and spiders by ELISA   总被引:7,自引:0,他引:7  
An ELISA was developed which would detect and quantify ingested aphids in predators found in and around cereal crops. The detection limit of the assay was less than one hundredth of an homogenised adult aphid. Tests with 13 species of aphid showed that those which had been used as the principal immunogens reacted most strongly in the assay. Nearly a hundred species of invertebrates, both predators and alternative prey, have been tested in the assay and no evidence of significant cross-reaction was found with any of these species or with a number of samples of plant material on which aphids may be found. Aphid material could still be detected in predators which had been stored for up to 7 days in 4% formalin or 70% ethanol.  相似文献

Permethrin-impregnated bednets reduce nuisance arthropods in Gambian houses   总被引:1,自引:0,他引:1  
The prevalence of bedbugs (Cimex hemipterus L.), chicken ticks (Argas persicus Oken) and headlice (Pediculus capitis De Geer) was surveyed in a rural area of The Gambia. At the beginning of the study 37.5% of children's beds were infested with bedbugs and 3.9% with chicken ticks, whilst the prevalence rate of pediculosis in children under 10 years old was 28.8%. Both bedbugs and headlice were clustered within compounds. Headlice prevalence increased with hair length and they were more common on girls than boys. Following this cross-sectional survey all bednets in the sixteen hamlets were either dipped in permethrin or a placebo. About 4 months later it was found that bedbugs and chicken ticks had disappeared from homes in which the bednets had been impregnated with permethrin. There was no reduction in hamlets with placebo-treated bednets. The rate of acquiring headlice between the two surveys was reduced by 91.1% in children who slept under insecticide-treated bednets compared with children with placebo-treated bednets. There were also significantly fewer day-flying and crawling insects, except earwigs, in homes of children who slept under insecticide-treated bednets compared with those with placebo-treated nets. These additional benefits of permethrin-treated bednets should contribute to their widespread acceptance and utilization by the community for personal protection.  相似文献

Are Roots a Source of Abscisic Acid for the Shoots of Flooded Pea Plants?   总被引:4,自引:1,他引:3  
Flooding the soil for 2–5 d decreased stomatal conductancesof pea plants (Pisum sativum L., cv. Sprite) with six or sevenleaves. This coincided with slower transpiration, increasedleaf water potentials and increased concentrations of abscisicacid (ABA) in the leaves. No increase in ABA was found in theterminal 20 mm of roots of flooded plants over the same timeperiod. Small stomatal conductances associated with increases in foliarABA were also found in plants grown in nutrient solution whenaeration was halted, causing the equilibrium partial pressuresof dissolved oxygen to fall below 05 It Pa. No increase in ABAconcentration in young secondary roots of the non-aerated plantswas detected after 24, 48 or 72 h, even when the shoot, thepresumed site of deposition for any ABA from the roots, wasremoved 5–6 h before analysis. Similarly, ABA concentrations in roots were not increased whenthe nutrient solution was de-oxygenated by continuous purgingwith nitrogen gas. The abscisic acid concentration in leaf epidermis,the tissue most likely to be the recipient of any ABA movingin the transpiration stream from oxygen-deficient roots, waslower than in the remaining parts of the leaf when examinedin the mutant Argenteum which possesses easily removable epidermallayers. It is concluded that the leaves of plants subjectedto flooding of the soil or oxygen shortage in the root environmentare not enriched substantially with ABA from the roots. The development of Caryospora bigenetica in vitro is described by light microscopy. Sporozoites from snake-derived oocysts were purified and inoculated onto cultures of primary testicle cells of the cotton rat, cotton rat kidney cells, and human fetal lung cells. Intracellular sporozoites were observed one and two days postinoculation (DPI). Motile, extracellular first-generation merozoites were present 3 DPI, and second-generation merozoites were present 5 DPI. Mature gamonts were observed 9 DPI and developed into unsporulated oocysts by 10 DPI. Oocysts sporulated in vitro, and excystation was observed. Cells that were penetrated by in vitro-produced sporozoites formed caryocysts by 16 DPI. To test infectivity of in vitro-derived stages, merozoites were removed from cultured cells 5 DPI and inoculated intraperitoneally into a mouse; infection resulted. Sporulated oocysts removed from cell cultures 12 DPI produced facial swelling in an orally inoculated cotton rat.  相似文献

Release of sporozoites from the oocysts of Cryptosporidium baileyi is described from Nomarski interference-contrast microscopy. Just prior to excystation, the four sporozoites became motile and rearranged themselves within the oocyst. The sporozoites were then rapidly expelled through an opening that formed in the oocyst wall, and the residuum was either released or retained within the oocyst. Excysted sporozoites were crescent shaped and measured 5.0–9.0 μm × 1.0–1.6 μm (x?= 6.8 × 1.1 μm). Excystation occurred when sodium taurocholate or a mixture of trypsin and sodium taurocholate was present in the incubation medium. High levels of excystation occurred at 37° or 40°C, but excystation did not occur at 4°C. The ability of biles from two avian and two mammalian hosts to produce excystation of C. baileyi was also studied. After a 2-h incubation at 40°C, the percentages of excystation were 69.5% in goat bile, 45.0% in pig bile, 33.0% in chicken bile, and 34.5% in turkey bile.  相似文献

SYNOPSIS. The fine structure of 2 isolates of Trypanosoma congolense maintained in laboratory rodents has been studied from thin sections of osmium- and aldehyde-fixed flagellates. The pellicular complex, nucleus, and flagellar apparatus are all similar to those of other African trypanosomes. Aberrant intracellular differentiation of the flagellum is occasionally found. As in bloodstream forms of other salivarian trypanosomes the single mitochondrion forms an irregular canal running from one end of the body to the other, with a shallow bowl-shaped expansion forming a capsule for the fibrous kinetoplast (mitochondrial DNA). A connexion between the mitochondrial envelope of the kinetoplast and the basal body of the flagellum is not evident, and sometimes the flagellum base is not even apposed to the kinetoplast but lies behind it. Tubular cristae are present in the mitochondrial canal and, by light microscopy, this structure gives a positive reaction for NAD diaphorase suggesting at least some activity in electron transport, even tho at this stage in its life cycle respiration is doubtfully sensitive to cyanide and cytochrome pigments are in all probability absent. The region of the cytoplasm between the nucleus and the flagellar pocket has all the trappings associated with secretory cells in higher animals, or with the secretion of surface structures in phytoflagellates. just behind the nucleus a limb of granular reticulum subtends a Colgi stack of flattened saccules with attendant vesicles. Close to the distal pole of the Golgi complex is a network of smooth-membraned cisternae, termed here the agranular or secretory reticulum, which undergoes localized swelling with the accumulation of a secretory product to form large spherical sacs or vacuoles. These network-linked vacuoles probably correspond to the post nuclear vacuole complex visible by light microscopy. From its apparent secretory function this complex is regarded here as being possibly an extension or derivative of the Golgi complex, the smooth-membraned tubules lying alongside the 2 structures possibly representing a link between them. By analogy with phytoflagellates and the secretory cells of higher animals, it is suggested that the secretion is transported for discharge into the flagellar pocket by way of multivesicular bodies and smooth-walled tubules or vesicles. Spiny pits in the wall of the flagellar pocket, and similar-sized vesicles in the nearby cytoplasm, could be stages in either exocytosis of secretion or endocytosis (pinocytosis). It is tentatively suggested that the secretion may be the material from which the surface coat is formed. Neither a cytostome nor a contractile vacuole has been observed in T. congolense.  相似文献

Traditionally, compared with the tropics, temperate systems are believed to: (i) have environments which are less favourable (i.e. harsher, more variable and less predictable); and (ii) support communities which are less diverse. Explanations for differences between temperate and tropical communities, including differences in diversity, generally rely on the former notion. The evidence for these ideas is, at best, equivocal. Organisms are subjected to physical stresses and disturbances on both temperate and tropical reefs. Communities on temperate reefs are not invariably less diverse than those in the tropics, at least at small spatial scales. Finally, there is as yet little evidence of genuine differences between the ecology of temperate and tropical communities. There is, however, much small-scale, spatial patchiness in the structure of reef communities and their physical environment. This patchiness in structure may result from patchiness in biological factors (e.g. recruitment) or in the physical environment. This small-scale variation in environmental factors may prove to be a more important determinant of community structure than the large-scale, latitudinal trends some ecologists have been obsessed with.  相似文献

SYNOPSIS. Cryptobia vaginalis (Hesse 1910) occurs as long thin and short broad forms in the vagina of the gnathobdelliform leeches Haemopis sanguisuga (Linnaeus) and Hirudo medicinalis Linnaeus. Cytochemical staining for DNA and transmission electron microscopy of sectioned material indicate that in the thin forms the kinetoplast DNA (kDNA) is dispersed irregularly through the mitochondrial network ( pankinetoplastic condition) rather than concentrated in the adbasal region of the mitochondrion ( eukinetoplastic condition) as in trypanosomatids and most other kinetoplastid flagellates. Light-microscopic studies on the rare broad forms, however, suggest that these have conventional adbasal location of the kinetoplast. Binary fission appears to occur in the thin forms, suggesting that the dispersed kinetoplast is either highly polyenergid or lacks a genetic function. In other features of its microanatomy, C. vaginalis is a conventional kinetoplastid. The flagellate has an incomplete corset of pellicular microtubules which may have a role in the cortical contractility characteristic of the genus Cryptobia . Feeding is by pinocytosis of vaginal colloids through a microtubule-lined cytopharynx, possibly after binding to a prominent filament-coated preoral ridge. A pulsatile (contractile) vacuole is present and appears to be responsible for defecation as well as osmoregulation. Some individuals have elongate bacterial epibionts attached to the body in parallel with the cortical microtubules. All individuals have 2–8 spheroplast-like endobiotic bacteria in the prenuclear cytoplasm.  相似文献
