Different mulberry genotypes show great variation in their resistance to the powdery mildew Phyllactinia corylea. Conidial germination and hyphal growth of P. corylea on the leaf surface of two susceptible mulberry genotypes, viz., Kanva 2 (K2) and Victory 1 (V1) varieties of Morus indica, and on two resistant species, viz., M. laevigata and M. serrata were studied by scanning electron microscopy. Conidial germination and growth of germ tubes were normal on all the leaves. The hyphae of P. coryleaidentify stomata on host leaves by their topographical features to produce the stomatopodia precisely over them. The holes and/or the grooves of stomata appear to provide the signals for the initiation of stomatopodia and similar structures are erratically developed over many local depressions or grooves on leaf surface. The abaxial surface of K2 leaf is smooth without prominent undulations of epidermal cell surface, and the stomata are flush with the leaf surface. Although successful penetration is also achieved on V1 leaf, its slightly undulated surface occasionally provides inaccurate tropic signals to the hyphae, inducing the development of stomatopodia away from the stomata. The leaf surfaces of M. laevigata and M. serrata are very rough with highly sculptured cuticle and abundant epidermal outgrowths. Stomata mostly remain sunken or hidden amidst the cuticular ornamentations and the hyphae fail to recognise the precise signals from them. As the surface architecture of the leaves provides many immense sources of tropic signals, stomatopodia are often produced over local depressions or grooves. In these cases the fungus fails to penetrate the leaf, does not develop beyond 24 h and penetration is rarely achieved on the leaves of the resistant plants. The study indicates that the stimulatory effect of the leaf surface topography of resistant varieties misleads the pathogen from successful penetration, thus contributing to the plant's resistance.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

Exploding global population, rapid urbanization, salinization of soils, decreasing arable land availability, groundwater resources, and dynamic climatic conditions pose impending damage to our food security by reducing the grain quality and quantity. This issue is further compounded in arid and semi-arid regions due to the shortage of irrigation water and erratic rainfalls. Millets are gluten (a family of proteins)-free and cultivated all over the globe for human consumption, fuel, feed, and fodder. They provide nutritional security for the under- and malnourished. With the deployment of strategies like foliar spray, traditional/marker-assisted breeding, identification of candidate genes for the translocation of important minerals, and genome-editing technologies, it is now tenable to biofortify important millets. Since the bioavailability of iron and zinc has been proven in human trials, the challenge is to make such grains accessible. This review encompasses nutritional benefits, progress made, challenges being encountered, and prospects of enriching millet crops with essential minerals.  相似文献

Withaferin A is an abundant withanolide present in Withania somnifera leaves and to some extent in roots. It has been known for its profound anti-cancer properties, but its role in counteracting the Leishmania donovani infection has to be explored. Pteridine reductase 1 (PTR1) is involved in pteridine salvage and an important enzyme for the parasite growth, which could be targeted for the development of an efficient antileishmanial drug. We employed molecular docking studies to identify the binding mode of withaferin A with PTR1 in silico. We further cloned, expressed, and purified PTR1 of L. donovani and performed the enzyme kinetics using the Michaelis–Menten equation and enzyme inhibition studies with withaferin A by plotting the Lineweaver–Burk graph, which followed an uncompetitive mode of inhibition. We also showed the inhibition of the enzyme in the crude lysate of treated parasites. Thus, our study contributes towards understanding the mode of action of withaferin A against L. donovani parasite.  相似文献

A number of major changes have occurred over the past few years, which give cause for a re-examination of conventional wastewater treatment methods. Among these are growing problems of worldwide energy and food shortages and nutrients not removed by conventional secondary processes causing algal blooms and other problems in the receiving waters. The global increase in wastewater calls for innovative low cost technology approaches to its recycling. Biotreatment systems, utilizing living organisms are receiving growing attention since they are ecologically sound, cheap and applicable in areas without land constraints. Filter feeders (both invertebrates and vertebrates) are promising in this area since they can remove suspended organic matter and bacteria, even in the size range of microns. In the present study biological treatment of municipal wastewater using two invertebrates--Paramecium caudatum, a protozoan and Daphnia magna, a cladoceran was investigated. Analysis at pre-experimental and post-experimental stages revealed the potential of these species in abatement of water pollution. D. magna was more efficient than P. caudatum in laboratory-scale studies.  相似文献

Extirpation of the cerebral neurosecretory cells of the pars intercerebralis of the red cotton bug, Dysdercus cingulatus, does not prevent maturation of the first batch of eggs in the female. However, vitellogenesis is visibly inhibited by the operation. There is considerable individual variation in the degree of inhibition of vitellogenesis in experimental animals. But in these experimental animals ovaries are shorter, their oöcytes smaller, the number of oöcytes undergoing vitellogenesis fewer: also the follicle cells and their nuclei are smaller. On the contrary, the protein concentration in the blood of experimental animals is higher but is subject to considerable variation and it appears to have no significance. It is concluded that the median neurosecretory cells stimulate vitellogenesis in this animal to a considerable extent.  相似文献

In the present study, metagenomic library of Western Ghats soil sample was constructed in a fosmid vector (pCC1FOS) and screened for biocatalytic properties. The clones showed amylolytic activity on Luria-Bertani starch agar plates and one of them was studied in detail. The enzyme exhibited stability at elevated temperature with 60°C being the optimal temperature. The enzyme retained more than 30% activity after 60 min incubation at 80°C. It also showed more than 70% activity retention in 1.5 M NaCl solution. The pH optimum of the enzyme was at pH = 5.0. The enzyme possesses good activity in the presence of chelating and strong reducing agents with activity enhancements or retention being observed at 5 mM β-mercaptoethanol, dithiothreitol and N-bromosuccinimide. However, almost complete loss of activity was observed with 5 mM EDTA, while activity enhancement was observed upon incubation with Ca2+ suggesting it to be a Ca2+-dependent α-amylase, which was further confirmed by a thin-layer chromatography (TLC). The TLC run revealed that digestion pattern was similar to commercial α-amylase. The 16S rRNA gene sequence (GenBank accession number HQ680979) BLAST showed 95% similarities with Exiguobacterium sp. AFB-11 and AFB 18, with query sequence coverage of 99%.  相似文献

A phytase gene from Aspergillus niger was isolated and two Escherichia coli expression systems, based on T7 RNA polymerase promoter and tac promoter, were used for its recombinant expression. Co-expression of molecular chaperone, GroES/EL, aided functional cytosolic expression of the phytase in E. coli BL21 (DE3). Untagged and maltose-binding protein-tagged recombinant phytase showed an activity band of ~49 and 92 kDa, respectively, on a zymogram. Heterologously-expressed phytase was fractionated from endogenous E. coli phytase by (NH4)2SO4 precipitation. The enzyme had optimum activity at 50 °C and pH 6.5.  相似文献

Both cDNA including 5′UTR and 3′UTR and genomic clones of cinnamyl alcohol dehydrogenase (CAD) were isolated and characterized from a pulp-yielding leguminous tree Leucaena leucocephala (LlCAD1). The deduced amino acid sequence shared high identity with orthologous sequences of Acacia mangium?×?Acacia auriculiformis (83%), Medicago sativa (83%), Nicotiana tabaccum (83%) and Aralia cordata (81%). Full length cDNA contained 78 bases of 5′UTR and 283 bases of 3′UTR, while the genomic clone contained 5 exons and 4 introns. Western blot analysis revealed elevated expression of LlCAD1 in seedling roots and shoots compared to leaves. Sense and antisense CAD tobacco transgenics showed increased and reduced CAD activity accompanied by a change in monomeric lignin composition. Histochemical staining of lignin in down-regulated plants suggested an increase in aldehyde units and a decrease in S/G ratio. Down-regulation of CAD resulted in accumulation of syringic, ferulic, p-coumaric and sinapic acids compared to untransformed controls. These observations were validated by anatomical studies of down-regulated transgenic stems which showed thin walled, elongated phloem and xylem fibres, accompanied by a reduction in the density of vessel elements and amount of secondary xylem when compared to untransformed plants. Furthermore, Klason lignin analysis of CAD antisense transgenics showed 7–32% reduced lignin and normal phenotype as compared to untransformed plants. Such a reduction was not noticed in up-regulated transgenics. These results demonstrate a unique opportunity to explore the significant role that down-regulation of CAD gene plays in reducing lignin content thereby offering potential benefits to the pulp and paper industry.  相似文献

Healthy mulberry roots were inoculated with second stage juveniles of the root-knot nematode, Meloidogyne incognita and the sequential anatomical changes in the root concurrent with the development of the nematode were studied with light and scanning electron microscopes. Visible anatomical changes occurred four weeks post-inoculation when the pear-shaped adult female nematodes appeared in the tissues. The infected roots lost their circular outline and increased in thickness as traumatic parenchyma was induced in the root. The stele was disorganised with reduced and deformed vascular elements. Many giant cells developed in the traumatic tissue, adjacent to the adult females. Eight weeks after the inoculation, cavities developed around the adult females by tissue disintegration, and the galls became discernable on the roots. The number and size of thegalls increased in the next three weeks, corresponding to the development of more traumatic tissues and the enlargement of the adult female nematodes. The majority of the nematodes were settled towards the gall periphery, with their posterior oriented outwards for the release of their eggs into soil by the disintegration of the outer root layers. But many were settled deep inside the gall tissue without a channel to release their eggs to soil. The inner cavities enlarged in large proportions, sometimes occupying the major volume of the gall. The conductive tissues of the root were totally disorganised or were nonexistent.  相似文献
