Freshwater mussels (Unionidae) are among North America's most imperilled organisms. Mussels produce small larvae (glochidia) that parasitize aquatic vertebrates. We modified the Epicentre QuickExtract protocol to extract DNA from a single glochidium, collected directly from the marsupium of a female mussel, to use as template in polymerase chain reactions (PCRs). Yield per glochidium in a 40 µL extraction volume provided enough DNA for ≥ 15 PCRs per individual. We were successful in using this DNA for microsatellite analysis of up to three loci per individual. Offspring from one female showed evidence for multiple paternity within her brood. Our results are the first documentation of this phenomenon in freshwater mussels.  相似文献

We assessed the potential impacts of land-use changes resulting from a change in the current biofuel policy on biodiversity in Europe. We evaluated the possible impact of both arable and woody biofuel crops on changes in distribution of 313 species pertaining to different taxonomic groups. Using species-specific information on habitat suitability as well as land use simulations for three different biofuel policy options, we downscaled available species distribution data from the original resolution of 50 to 1 km. The downscaled maps were then applied to analyse potential changes in habitat size and species composition at different spatial levels. Our results indicate that more species might suffer from habitat losses rather than benefit from a doubled biofuel target, while abolishing the biofuel target would mainly have positive effects. However, the possible impacts vary spatially and depend on the biofuel crop choice, with woody crops being less detrimental than arable crops. Our results give an indication for policy and decision makers of what might happen to biodiversity under a changed biofuel policy in the European Union. The presented approach is considered to be innovative as to date no comparable policy impact assessment has been applied to such a large set of key species at the European scale.  相似文献

Abstract.  Teneral reserve components (soluble sugars, glycogen, lipids), as well as proteins, of laboratory-reared yellow dung flies ( Scathophaga stercoraria ) are shown to increase hypo-allometrically (slope < 1) with body size. This is also true for sugars and glycogen of field-caught reproductive flies, the lipid content of which however, increases hyper-allometrically (slope > 1), probably related to foraging or reproductive activity. The longevity, or starvation resistance, of adults subsisting exclusively on their teneral reserves equally increases with body size, and this positive relationship remains when flies have access to sugar, pollen or prey for 24 h after emergence. Energy use, by contrast, is generally not body size dependent, except for lipid (and possibly glycogen) utilization when flies were completely starved. All the results obtained are independent of sex after controlling for sexual size dimorphism. These net results clearly support the greater energy efficiency of larger flies (Kleiber's law), whereas only weak evidence is obtained in support of the contrasting hypothesis of greater absolute energy demands of larger individuals, possibly because such effects are likely masked by stronger size-dependent anabolic effects.  相似文献

Sisyrinchium micranthum is native to southern Brazil and shows a wide range of morphological variation in this region. This study was performed to compare individuals of this species in terms of traits such as morphology, chromosome number, meiotic behaviour, pollen viability and genetic diversity using inter simple sequence repeat markers. First, general morphological aspects were evaluated and plants were classified into three morphological categories (CI, CII and CIII) to analyse the cytogenetic and genetic data. Three chromosome numbers corresponding to three ploidies were found: 2n = 2x = 16, 2n = 4x = 32 and 2n = 6x = 48. All morphological types showed regular meiotic behaviour and high meiotic index values and pollen viability. CII is the most frequent in southern Brazil. The most common haploid chromosome number, n = 8, has not been reported previously in this species. The percentage of polymorphic loci ranged from 6.45 to 61.29% for each accession. Molecular and cytogenetic analyses indicate that some S. micranthum accessions may have more than one ploidy.
