Dithionite causes the depletion of dioxygen from suspensions of erythrocytes by reduction of the external dioxygen and not by diffusion into the cell. The molar enthalpy for the reduction shows a small difference with respect to the values found for free hemoglobin; and the normal stoichiometry of 2 moles dithionite/mole dioxygen found there is not observed with erythrocytes. At low hematocrit, the stoichiometry is 2.6:1 and decreases to 1.5:1 at high hematocrit. The change is not due to differences in the hemoglobin saturation or to an inability of dithionite to reduce all dioxygen present at the higher hematocrit. Neither catalase nor peroxidase added to the extracellular volume significantly alters the stoichiometry or the enthalpy of dioxygen reduction by dithionite. Addition of superoxide dismutase, however, restores the normal stoichiometry at high hematocrit and further increases the stoichiometry at low hematocrit. The calorimetrical signal of hydrogen peroxide, clearly seen with free dioxygen, is not present with erythrocytes. In all these cases the total heat evolved is the same.  相似文献

Calorimetric studies of the effect of superoxide dismutase and/or catalase on the reduction of dioxygen into water by dithionite in oxyhemoglobin have been carried out and the results compared with those in red cell hemolysates. In the absence of the enzymes the stoichiometry (moles dithionite/mole dioxygen) is less than the value of 2:1 which was found previously in red cell hemolysates [Forlani et al., J. Inorg. Biochem. 20, 147-155 (1984)]. In the presence of either superoxide dismutase or catalase alone the stoichiometry increases but is still less than 2:1. In the presence of both enzymes the stoichiometry and the shape of the thermogram is that previously observed for hemolysates, suggesting the presence of a hemoglobin-catalase-superoxide dismutase integrated system. The absence of a calorimetric signal for hydrogen peroxide in the reduction of oxyhemoglobin in the presence of superoxide dismutase suggests a wider biological role of superoxide dismutase than previously thought.  相似文献

Purpose

The population living in urban areas of the world continues to grow rapidly. It is, thus, a great priority for the planning practice to embed sustainability concept in their urban development endeavors. Currently, development and expansion of urban systems stress the need to control consumption of resources, especially non-renewable ones. There is also a need to reduce related environmental impacts, while stimulating a sustainable pathway for the population and urban growth.

Methods

Strategic environmental assessment (SEA) is useful for policy design to build an integrated method for supporting the development of a sustainable society. It undertakes territorial assessments and describes urban flows and impacts related to them by using a variety of tools, including material flow accounting (MFA). This study employs MFA, as it fits well within the scope of SEA and supports the growing environmental attention in the urban metabolism approach. Although helpful, MFA has not been systematically applied in the urban development context; for this reason, this paper proposes the integration of SEA and MFA.

Results and discussion

Integration of SEA and MFA generates a new framework for sustainable development planning. The framework is structured in phases oriented to the continual improvement based on the Deming cycle (i.e., plan, do, check, act), a key management approach mainly used in businesses for improving the effectiveness of an organization. It can also be implemented at the urban system level. In order to maintain normative compliance, each process (urban planning, strategic environmental assessment with urban metabolism approach, participatory processes) is standardized in line with a common and mandatory approach. While the processes are integrated among them, highlighting the reciprocal contact points, the results are combined in a holistic perspective. The framework, hence, transforms the voluntary MFA tool into a mandatory process.

Conclusions

The proposed SEA-MFA framework has the potential to unify and standardize the processes of categorizing and quantifying data in order to improve the understanding of urban metabolic principles and scale effects. It also supports management and policy development and meets the requirements of different stakeholders. The framework, thus, generated a novel approach for sustainable urban development planning by providing solutions for specific policy problems and ensuring urban ecological balance and sustainable urban futures.

Optical modes of dielectric micro-cavities have received significant attention in recent years for their potential in a broad range of applications. The optical modes are frequently referred to as "whispering gallery modes" (WGM) or "morphology dependent resonances" (MDR) and exhibit high optical quality factors. Some proposed applications of micro-cavity optical resonators are in spectroscopy1, micro-cavity laser technology2, optical communications3-6 as well as sensor technology. The WGM-based sensor applications include those in biology7, trace gas detection8, and impurity detection in liquids9. Mechanical sensors based on microsphere resonators have also been proposed, including those for force10,11, pressure12, acceleration13 and wall shear stress14. In the present, we demonstrate a WGM-based electric field sensor, which builds on our previous studies15,16. A candidate application of this sensor is in the detection of neuronal action potential.The electric field sensor is based on polymeric multi-layered dielectric microspheres. The external electric field induces surface and body forces on the spheres (electrostriction effect) leading to elastic deformation. This change in the morphology of the spheres, leads to shifts in the WGM. The electric field-induced WGM shifts are interrogated by exciting the optical modes of the spheres by laser light. Light from a distributed feedback (DFB) laser (nominal wavelength of ~ 1.3 μm) is side-coupled into the microspheres using a tapered section of a single mode optical fiber. The base material of the spheres is polydimethylsiloxane (PDMS). Three microsphere geometries are used: (1) PDMS sphere with a 60:1 volumetric ratio of base-to-curing agent mixture, (2) multi layer sphere with 60:1 PDMS core, in order to increase the dielectric constant of the sphere, a middle layer of 60:1 PDMS that is mixed with varying amounts (2% to 10% by volume) of barium titanate and an outer layer of 60:1 PDMS and (3) solid silica sphere coated with a thin layer of uncured PDMS base. In each type of sensor, laser light from the tapered fiber is coupled into the outermost layer that provides high optical quality factor WGM (Q ~ 106). The microspheres are poled for several hours at electric fields of ~ 1 MV/m to increase their sensitivity to electric field.  相似文献

Thyroglobulin (Tg), isolated from soluble iodoproteins by ammonium sulphate fractionation, was enzymatically deglycosylated in vitro and analyzed by polyacrylamide gel electrophoresis, double immunodiffusion and non-commercial RIA. Carbohydrate and iodine content was chemically determined. By PAAGE deglycosylated Tg (dTg) showed the appearance of a major band in the 12S region and three slower migrating bands corresponding to higher aggregates than 19S Tg. In immunodiffusion by testing native and deglycosylated Tg against anti-native Tg antiserum it was shown the appearance of a spur of native on deglycosylated Tg. By RIA of native and deglycosylated Tg against anti-deglycosylated Tg antiserum it was shown a minor binding capacity of the anti-deglycosylated antibody against native Tg at high dilutions. The results demonstrate that the enzymatic deglycosylation release almost all the carbohydrates of goiter Tg and that the removal of the carbohydrates of Tg produces a loss of antigenic determinants of the molecule.  相似文献

Diet restriction, prolonging the lifespan of rodents, represents an interesting model for gerontological studies. We analyzed the activity of antioxidant enzymes, Superoxide Dismutase, Catalase and Glutathione Peroxidase in erythrocytes from young, old and old food restricted Wistar rats. Diet restriction was applied feeding the animals on every-other-day schedule starting from the age of 3.5 months. The age-dependent decrease of Catalase and Glutathione Peroxidase activities was prevented by food restriction, whereas Superoxide Dismutase activity was not influenced either by aging and dietary intervention. Present results support the hypothesis that diet restriction increases the protection of cell structure against the peroxidative damage, preserving the activity of antioxidant enzymes.  相似文献
