In a randomised, double blind, long term, crossover study 1 g twice daily of methenamine hippurate was compared with placebo for its preventive effect on recurrent attacks of acute cystitis. Methenamine hippurate and placebo were interchanged every six months for two years. During one of the years patients took 250 ml extra fluid every morning and evening. Out of 21 enrolled patients, 14 completed the first year and 13 both years of treatment, which permitted the evaluation of 27 patient years. There were 52 episodes of acute cystitis caused by reinfection: 41 occurred during placebo treatment and only 11 during the methenamine hippurate regimen (p less than 0.01). Extra fluid intake did not reduce the incidence of acute cystitis, nor did it reduce the effect of methenamine hippurate. Methenamine hippurate is an effective prophylactic agent against recurrent acute cystitis and has the advantage of not inducing cross resistance to conventional antibiotics.  相似文献

In the purification of proline hydroxylase by affinity chromatography on poly(L-proline)-Sepharose it was found earlier that two other components, profilin and the complex profilin-actin, also bind with high affinity to this matrix. We have exploited this observation to develop a rapid procedure for the isolation of profilin and profilin-actin complexes in high yields directly from high-speed supernatants of crude tissue-extracts. Through an extensive search for elution conditions, avoiding poly(L-proline) as the desorbant, we have found that active proteins can be recovered from the affinity column with a buffer containing 30% dimethyl sulphoxide. Subsequent chromatography on hydroxylapatite separates free profilin and the two isoforms of profilactin, profilin-actinβ and profilin-actinγ. The profilin-actin complexes produced this way have high specific activities in the DNAase-inhibition assay, give rise to filaments on addition of Mg2+, and can be crystallized. From the isolated profilin-actin complexes the β- and γ-actin isoforms of non-muscle cells can easily be prepared in a polymerization competent form. Pure profilin is either obtained from an excess pool present in some extracts or by dissociation of profilin-actin complexes and removal of the actin.  相似文献

Water level regulation causes large-scale ecological changes in the littoral areas of lakes in northern Finland. If the summertime water level is raised, intensive erosion processes begin, causing a sudden decline in shore vegetation. The need for shore protection is obvious in areas of high recreational value. At lake Ontojärvi, planting experiments with littoral helophytes and bushes were carried out during the years 1990–92. All the experiments were carried out in the eroded sandy areas, which were partly protected by mechanical barriers. Several plant species were planted on the shore which had been treated with different peat mixtures, etc. The frequencies of the different species were followed monthly. After the first summer, the average survival rates were about 45 % due to the drying of seedlings. A rapid decrease in the survival rates took place during the high water level period in 1991 at which point only 20% of the planted individuals were alive. The best results were obtained for the helophyte Carex rostrata Stokes, of which 30% had survived erosion. Tall willows (Salix phylicifolia L.) were also erosion-resistant with a survival rate of 80%.  相似文献

Skeletal muscle transport of lactate and pyruvate was studied in primary cultures of rat myotubes, applying the pH-sensitive fluorescent indicator 2', 7'-bis(carboxyethyl)-5(6)-carboxyfluorescein. The initial rate of decrease in intracellular pH (pHi) upon lactate or pyruvate incubation was used to determine total transport (carrier mediated and diffusion). Both lactate and pyruvate transport could be inhibited by a combination of 0.5 mM 4,4'-diisothiocyanostilbene-2, 2'-disulfonic acid, 5 mM mersalyl and 10 mM alpha-cyano-4-hydroxycinnamate. The kinetic parameters, Km and Vmax, for carrier-mediated transport of lactate were 9.9+/-1.1 mM and 0. 69+/-0.02 mmol l-1 s-1, respectively. For pyruvate, Km and Vmax were 4.4+/-1.3 mM and 0.30+/-0.05 mmol l-1 s-1, respectively. The diffusion component of the total transport was 0.0040+/-0.0005[S] (n=4) and 0.0048+/-0.0003[S] (n=4) for lactate and pyruvate, respectively. Furthermore, it was observed that the two monocarboxylate transporter isoforms present in mature skeletal muscles, MCT1 and MCT4 (formerly called MCT3 (M.C. Wilson, V.N. Jackson, C. Heddle, N.T. Price, H. Pilegaard, C. Juel, A. Bonen, I. Montgomery, O.F. Hutter, A.P. Halestrap, Lactic acid efflux from white skeletal muscle is catalyzed by the monocarboxylate transporter isoform MCT3, J. Biol. Chem. 273 (1998) 15920-15926)), were also expressed in primary culture of myotubes.  相似文献

The metabolic role of 5'AMP-activated protein kinase (AMPK) in regulation of skeletal muscle metabolism in humans is unresolved. We measured isoform-specific AMPK activity and beta-acetyl-CoA carboxylase (ACCbeta) Ser(221) phosphorylation and substrate balance in skeletal muscle of eight athletes at rest, during cycling exercise for 1 h at 70% peak oxygen consumption, and 1 h into recovery. The experiment was performed twice, once in a glycogen-loaded (glycogen concentration approximately 900 mmol/kg dry wt) and once in a glycogen-depleted (glycogen concentration approximately 160 mmol/kg dry wt) state. At rest, plasma long-chain fatty acids (FA) were twofold higher in the glycogen-depleted than in the loaded state, and muscle alpha1 AMPK (160%) and alpha2 AMPK (145%) activities and ACCbeta Ser(221) phosphorylation (137%) were also significantly higher in the glycogen-depleted state. During exercise, alpha2 AMPK activity, ACCbeta Ser(221) phosphorylation, plasma catecholamines, and leg glucose and net FA uptake were significantly higher in the glycogen-depleted than in the glycogen-loaded state without apparent differences in muscle high-energy phosphates. Thus exercise in the glycogen-depleted state elicits an enhanced uptake of circulating fuels that might be associated with elevated muscle AMPK activation. It is concluded that muscle AMPK activity and ACCbeta Ser(221) phosphorylation at rest and during exercise are sensitive to the fuel status of the muscle. During exercise, this dependence may in part be mediated by humoral factors.  相似文献

Regulation of water flow constitutes the most important hydromorphological burden to Finnish lakes. The total area of regulated lakes is nearly 11,000 km2, equalling one-third of the total area of Finnish inland waters. Extensive research projects have been carried out since the end of the 1980s to find out opportunities to mitigate harmful effects of the regulation of watercourses. A water-level fluctuation analysis tool, known as Regcel, has been developed to study water level data and to identify the most significant impacts. Results of the Regcel analysis give an overall picture of the impact of lake regulation in northern climate. The model is based on relationships between the water-level fluctuation and factors related to environmental, social and economical effects. Regcel has been used in 12 Lake Regulation Development Projects in Finland. In this article, we show how the Regcel model was applied in two cases. Aquatic macrophytes are one of the biological quality elements in the Water Framework Directive (WFD) for which status assessments must be defined. We tested two methods to classify macrophyte species and their response to eutrophication pressure: one based on percentiles of occurrence along a phosphorous gradient and another based on trophic ranking of species using Canonical Correspondence Analyses in the ranking procedure. The methods were tested at Europe-wide, regional and national scale as well as by alkalinity category, using 1,147 lakes from 12 European states. The grouping of species as sensitive, tolerant or indifferent to eutrophication was evaluated for some taxa, such as the sensitive Chara spp. and the large isoetids, by analysing the (non-linear) response curve along a phosphorous gradient. These thresholds revealed in these response curves can be used to set boundaries among different ecological status classes. In total 48 taxa out of 114 taxa were classified identically regardless of dataset or classification method. These taxa can be considered the most consistent and reliable indicators of sensitivity or tolerance to eutrophication at European scale. Although the general response of well known indicator species seems to hold, there are many species that were evaluated differently according to the database selection and classification methods. This hampers a Europe-wide comparison of classified species lists as used for the status assessment within the WFD implementation process.  相似文献

Historical (1947–1950) and present day (1998–2000) aerial photograph data of aquatic vegetation were combined with soil, land use and habitat morphology variables in two lakes, oligotrophic L. Näsijärvi (256 km2) and eutrophic L. Pyhäjärvi (122 km2), with regulated water levels in southern Finland. Multivariate GLM analyses were used to examine the determining factors of occurrence and horizontal extension of vegetation, occurrence of common reed (Phragmites australis) and water horsetail (Equisetum fluviatile) and the vegetation change in 474 sites. Despite differences in geography and water level regulation between the lakes, congruent trends were identified. The vegetation occurrence in both lakes had a positive response to clay soils and incoming nutrients and a negative response to housing and shore slope. Moraine soils, housing and shore slope had a negative effect on horizontal extension of vegetation in both lakes. The occurrence of P. australis was controlled by a negative response to moraine and the occurrence of E. fluviatile to slope in both lakes. The vegetation change in both lakes was characterized by a negative response to housing and slope.  相似文献
