Haploinsufficiency of the short stature homeobox contaning SHOX gene has been shown to result in a spectrum of phenotypes ranging from Leri–Weill dyschondrosteosis (LWD) at the more severe end to SHOX-related short stature at the milder end of the spectrum. Most alterations are whole gene deletions, point mutations within the coding region, or microdeletions in its flanking sequences. Here, we present the clinical and molecular data as well as the potential molecular mechanism underlying a novel microdeletion, causing a variable SHOX-related haploinsufficiency disorder in a three-generation family. The phenotype resembles that of LWD in females, in males, however, the phenotypic expression is milder. The 15523-bp SHOX intragenic deletion, encompassing exons 3–6, was initially detected by array-CGH, followed by MLPA analysis. Sequencing of the breakpoints indicated an Alu recombination-mediated deletion (ARMD) as the potential causative mechanism.  相似文献

WE have found a glycoprotein in the human brain which is brain specific on immunological criteria1. Ontogenetically, this glycoprotein appears at the stage of premyelination glia. The amount increases in parallel with the development of the human brain and probably remains at a constant level in the adult brain2. We found it in normal and pathological human brain, predominantly in the white matter and in trace quantities in the grey matter (putamen, n. caudatus)2. It is not present in the peripheral nerves, but was found in the n. opticus. This component is almost identical with the brain specific protein of Macacus rhesus and semi-identical with the brain-specific protein of rats2.  相似文献

The pioneering research of Harold Flor on flax and the flax rust fungus culminated in his gene-for-gene hypothesis. It took nearly 50 years before the first fungal avirulence ( Avr ) gene in support of his hypothesis was cloned. Initially, fungal Avr genes were identified by reverse genetics and map-based cloning from model organisms, but, currently, the availability of many sequenced fungal genomes allows their cloning from additional fungi by a combination of comparative and functional genomics. It is believed that most Avr genes encode effectors that facilitate virulence by suppressing pathogen-associated molecular pattern-triggered immunity and induce effector-triggered immunity in plants containing cognate resistance proteins. In resistant plants, effectors are directly or indirectly recognized by cognate resistance proteins that reside either on the plasma membrane or inside the plant cell. Indirect recognition of an effector (also known as the guard model) implies that the virulence target of an effector in the host (the guardee) is guarded by the resistance protein (the guard) that senses manipulation of the guardee, leading to activation of effector-triggered immunity. In this article, we review the literature on fungal effectors and some pathogen-associated molecular patterns, including those of some fungi for which no gene-for-gene relationship has been established.  相似文献

Jerdon's Courser Rhinoptilus bitorquatus is one of the most endangered and least understood birds in the world. It is endemic to scrub habitats in southeast India which have been lost and degraded because of human land use. We used satellite images from 1991 and 2000 and two methods for classifying land cover to quantify loss of Jerdon's Courser habitat. The scrub habitats on which this species depends decreased in area by 11–15% during this short period (9.6 years), predominantly as a result of scrub clearance and conversion to agriculture. The remaining scrub patches were smaller and further from human settlements in 2000 than in 1991, implying that much of the scrub loss had occurred close to human population centres. We discuss the implications of our results for the conservation of Jerdon's Courser and the use of remote sensing methods in conservation.  相似文献

Phylogenetic analysis of 27 behavioural and life history traits for five Mediterranean sand goby species (Perciformes, Gobiidae) produced one tree with a consistency index (excluding uninformative characters) of 0.756. This tree agreed with previous molecular analyses in providing strong support for the monophyly of the sand gobies, indicating that Pomatoschistus and Knipowitschia are paraphyletic and helping to resolve the ambiguous position of Economidichthys pygmaeus, placing it as the basal member of the reduced data set. Although the tree was completely resolved, the branches above E. pygmaeus were only moderately supported in the bootstrap analysis. Overall, the behavioural data provide information that may eventually help clarify the speciation bursts within the Mediterranean sand goby clade as much as is possible. © 2012 The Linnean Society of London, Zoological Journal of the Linnean Society, 2012, 165 , 916–924.  相似文献

The antimicrobial activity of the azole fungicides cyproconazole and propiconazole as single active ingredients and in mixtures with the ATP-Binding Cassette (ABC) transporter modulators rhodamine 6G, quercetin, quinidine, and verapamil and the strobilurin kresoxim-methyl was assessed against the wheat pathogen Mycosphaerella graminicola . Interactions amongst these compounds were evaluated on germination and germ tube growth of pycnidiospores using the Colby and Wadley method. Water agar proved to be the best test medium since all pycnidiospores germinated within 24 h of incubation and apical germ tube growth dominated over bud formation by intermediate cells. Analysis with the Colby method revealed that interactions between the compounds in all mixtures tested on germination of pycnidiospores were additive. With regard to germ tube growth, mixtures of cyproconazole and verapamil or kresoxim-methyl displayed a synergistic interaction. Analysis of mixtures of cyproconazole and kresoxim-methyl with the Wadley method revealed that the interaction between the two compounds was purely additive. These results indicate that the Colby method overestimated the interaction between these two compounds in a mixture.  相似文献

The anion-exchange capacities (AEC) of roots of intact plants of two plum clones [Marianna 2624 (Prunus cerasifera × P. munsoniana) and Myrobalan 3-J (P. cerasifera)] were assessed with an anionic dye, eosin Y. The positively charged eosin-specific adsorption sites were metabolically dependent and also affected by nutrient status. Myrobalan 3-J roots adsorbed twice as much exchangeable NO3 as did Marianna 2624. Since we previously showed that net nitrate uptake by Myrobalan 3-J persisted at half the ambient nitrate concentration as that characteristic of Marianna 2624, the data provide circumstantial support for a functional role of labile anion binding in active uptake at dilute concentrations of ambient nitrate.  相似文献

Diversity in leaf morphological traits was assessed in 38 Greek Fagus sylvatica populations, covering the distribution of the species in the country. The relationship between the post‐glacial origin of these populations and leaf morphology was investigated. The results showed a complex and geographically continuous morphological diversity pattern, influenced mainly by traits expressing leaf size, leaf shape and petiole length. Two simultaneous trends appeared to be responsible for the existing diversity pattern. One was geographical, with leaf types resembling F. sylvatica subsp. sylvatica occurring in the western part of the distribution of beech in Greece and types resembling F. sylvatica subsp. orientalis being dominant in the eastern part. A second trend seemed to be connected with the post‐glacial origin of the populations, as described by previous plastid DNA haplotype studies of the same trees. The genetic background and the possible adaptation of beech populations to different environmental conditions have resulted in a complex morphological pattern, especially in areas in which different post‐glacial lineages appear to meet. © 2011 The Linnean Society of London, Botanical Journal of the Linnean Society, 2011, 165 , 422–436.  相似文献

Comparative phylogeographical studies in island archipelagos can reveal lineage-specific differential responses to the geological and climatic history. We analysed patterns of genetic diversity in six codistributed lineages of darkling beetles (Tenebrionidae) in the central Aegean archipelago which differ in wing development and habitat preferences. A total of 600 specimens from 30 islands and eight adjacent mainland regions were sequenced for mitochondrial cytochrome oxidase I and nuclear Muscular protein 20. Individual gene genealogies were assessed for the presence of groups that obey an independent coalescent process using a mixed Yule coalescent model. The six focal taxa differed greatly in the number of coalescent groups and depth of lineage subdivision, which was closely mirrored by the degree of geographical structuring. The most severe subdivision at both mitochondrial DNA and nuclear DNA level was found in flightless lineages associated with presumed stable compact-soil habitats (phrygana, maquis), in contrast to sand-obligate lineages inhabiting ephemeral coastal areas that displayed greater homogeneity across the archipelago. A winged lineage, although associated with stable habitats, showed no significant phylogenetic or geographical structuring. Patterns of nucleotide diversity and local genetic differentiation, as measured using ΦST and hierarchical amova , were consistent with high levels of ongoing gene flow in the winged taxon; frequent local extinction and island recolonisation for flightless sand-obligate taxa; and very low gene flow and geographical structure largely defined by the palaeogeographical history of the region in flightless compact-soil taxa. These results show that differences in dispersal rate, mediated by habitat persistence, greatly influence the levels of phylogeographical subdivision in lineages that are otherwise subjected to the same geological events and palaeoclimatic changes.  相似文献
