The cytoskeletal events that assist restitution of the native intestinal epithelium are poorly understood. To enhance our understanding of repair mechanisms in the native intestinal epithelium we assessed the functional role of actin and the temporal and spatial alterations in actin and villin that occur in native enterocytes migrating in response to injury. Using a well-characterizedin vitroUssing chamber model of native intestine epithelial restitution, the actin inhibitor cytochalasin D (CD) was applied to determine the functional importance of actin to restitution as assessed by sensitive electrophysiological means and structural techniques. Additionally we used phalloidin and indirect immunohistochemistry to localize and semi-quantitate F-actin and villin in migrating cells during restitution. We report new data that shows that when cytoskeletal changes were impaired with CD, the epithelial monolayer was re-established in fewer than 20% of CD-treated villi, cells bordering the epithelial defect did not assume the characteristic phenotype associated with migrating cells, and transepithelial resistance did not return to pre-injury levels. F-actin and villin were present at the leading edge of the migrating cells, basolateral F-actin was decreased, and cytoplasmic villin was increased as determined by phalloidin and immunohistochemical methods. We conclude thatin vitrorepair of the native intestinal epithelium is functionally and structurally dependent on major changes in the cytoskeleton of cells involved in re-establishing the epithelial monolayer over a complex extracellular matrix.  相似文献

Summary

Two neuropeptides (NPs), hydra head activator (HHA), and the luteinizing hormone releasing hormone (LHRH), were used to study their effects on the development of the larvae of the grain beetle Tenebrio molitor (mealworms). The larvae received the NPs in the course of development at the beginning or at the end of instar. The LHRH given at the beginning of the instar stimulated molting and pupation in larvae; it did not affect molting in the late period of the instar. The C-end fragment of LHRH(9–10) produced fewer molts and increased the duration of the instar period. The effect of this fragment was weak when it was injected at the initial period of the instar; it was considerably stronger in the end of the instar. It inhibited development when administered during the last instar. On the contrary, the N-end fragment of the LHRH(1–2) stimulated growth and development of larvae. The action of these NPs can be connected to the function of the hormones of metamorphosis. The HHA administrated to the larvae at different days of the instar stimulated molting but in the last instar it delayed pupation. The C-end of the HHA(7–11) stimulated molting when injected during the whole period of the instar. The other C-end fragments, the HHA(8–11) and HHA(9–11) had inhibitory effects on the molting. The injection of the N-end fragment of the HHA(1–5) had no effect. The action of HHA and its fragments did not correlate with the function of the hormones of metamorphosis.  相似文献

SYNOPSIS. The cytotoxicity of 97 antioxidants of various structural types was determined for Tetrahymena pyriformis grown in a peptone medium. Cytotoxicity fo Tetrahymena, generally, was positively associated with acute oral tomicoiciity to rats but not with antioxidant pdency as measured by the Tetrahymena photodynamic assay for antioxidants. Some commonly used hindered-phenol, lipidsoluble anttioxidants, e.g., 2,6-di-tert.-butyl-P-cresol (BHT), had low toxicity (to Tetrahymena but poor antioxidant activity by the photodynamic test. Nmdihydroguahetic acid, with low toxicity to the rat and high Coxicity (to Tetrahymena, had very high activity in the photodynamic test.  相似文献

Head lice, Pediculus capitis , were collected from children aged 3–12 years in Maale Adumin, a town near Jerusalem, after reports of control failure with the pyrethroid insecticide permethrin. A total of 1516 children were examined: living lice and eggs were found on 12.1% of the children; or another 22.8% of the children only nits were found. Twice as many girls as boys (8.1% v 4%) were infested with lice and or nits. Head lice collected from infested children were exposed to permethrin impregnated filter-papers. Log time probit mortality (ltp) regression lines were calculated for mortality data and compared to ltp lines for a similar collection of head lice made in 1989. The regression lines for the two years were significantly different, with a 4-fold decrease in susceptibility at the LT50 level between 1989 and 1994. The slopes of the lines also suggested that the 1994 population was more heterogenous in its response to permethrin than the 1989 population. In contrast, a laboratory population of body lice (Pediculus humanus) tested with the same batch of permethrin-impregnated papers showed a slight but non-significant increase in susceptibility between 1989 and 1994. The results suggest that resistance to pyrethroids has developed rapidly among head lice since permethrin was introduced in 1991 as a pediculicide in Israel.  相似文献

The incidence of abnormally developing embryos in brood pouchesof females was studied in five populations of Littorina saxatilisliving under different salinity conditions. The proportion ofabnormals decreased during embryonic development. Age of femaleand infection with the ciliate Protophrya ovi-cola had no effecton the frequency of abnormals. The proportion of abnormal eggs(PAE) increased at the end of the reproductive season; a similartrend was found for the proportion of abnormal veligers (PAV)although it was not always significant. The proportion of abnormaljuveniles (PAJ) showed no seasonality. The increase of PAE coincidedwith decrease of water temperature in September. Environmentalsalinity was significantly correlated with frequency of abnormalsin L. saxatilis populations: the highest values of PAV and PAJwere observed in estuarine population (salinity range 5–8%),intermediate in populations living under conditions of 10–20%osalinityand lowest in control populations (23–24%). Itis suggested that critical salinity (sensu Khle-bovich, 1974)(approximately 8–10%) may also be a crucial point forstability of embryonic development in the White Sea populationsof L. saxatilis. This article reviews the contribution made by functional electronmicroscopy towards identifying and understanding the reactionsof plant roots and shoots to anaerobic stress. Topics examinedinclude: (1) unexpected hypersensitivity, rather than hyper-resistance,to anoxia of root tips of flooding-tolerant plants; (2) protective,rather than damaging, effects of a stimulated energy metabolism(glycolysis and fermentation) under anaerobic conditions; (3)the concept of two main strategies of plant adaptation to anaerobicenvironments, namely avoidance of anaerobiosis on the wholeplant level, termed ‘apparent’ tolerance, and metabolicadaptation at the cellular and molecular levels, termed ‘true’tolerance; (4) the importance of protein synthesis during hypoxiaand anoxia for enhanced energy production and metabolic adaptation;(5) a general adaptive syndrome in plants to stress at the ultrastructurallevel and a possible molecular mechanism for its realizationunder anoxia; (6) the physiological role of anaerobically synthesizedlipids and nitrate as alternative electron acceptors in an oxygen-freemedium; and (7) the selection of cell lines derived from calluscultures that possess enhanced tolerance to anoxia and can regeneratewhole plants with improved tolerance of soil waterlogging.  相似文献
