A theoretical relationship has been developed to allow the effect of free ligand concentration on the capacity of an affinity Chromatography matrix to be determined where the protein adsorbed shows co-operative binding. Computer simulations using literature values for association constants show that under optimal conditions resin capacity can be increased significantly in the presence of a small but finite concentration of free ligand. The model also allows prediction of the soluble ligand concentration required for biospecific elution. The results obtained suggest the possibility of a new elution technique, "reverse biospecific elution," that reduces the amount of free ligand required to effect elution.  相似文献

A simple thermal monitoring technique has been successfully applied to an adsorption system using a novel ion exchanger with a large internal void volume (voidage) which can be operated at high superficial velocity (SV). Temperature changes resulting from heats of adsorption could be followed effectively using semi-conductor thermistor devices inserted into the resin through the column wall. Results show that, despite the high feed rates adopted, the thermal signals generated can be consistently related to the position of the breakthrough front within the bed.  相似文献

To identify genes influencing age at onset (AAO) in two common neurodegenerative diseases, a genomic screen was performed for AAO in families with Alzheimer disease (AD; n=449) and Parkinson disease (PD; n=174). Heritabilities between 40%–60% were found in both the AD and PD data sets. For PD, significant evidence for linkage to AAO was found on chromosome 1p (LOD = 3.41). For AD, the AAO effect of APOE (LOD = 3.28) was confirmed. In addition, evidence for AAO linkage on chromosomes 6 and 10 was identified independently in both the AD and PD data sets. Subsequent unified analyses of these regions identified a single peak on chromosome 10q between D10S1239 and D10S1237, with a maximum LOD score of 2.62. These data suggest that a common gene affects AAO in these two common complex neurodegenerative diseases.  相似文献

Background

Children with antisocial behaviour show deficits in the perception of emotional expressions in others that may contribute to the development and persistence of antisocial and aggressive behaviour. Current treatments for antisocial youngsters are limited in effectiveness. It has been argued that more attention should be devoted to interventions that target neuropsychological correlates of antisocial behaviour. This study examined the effect of emotion recognition training on criminal behaviour.

Methods

Emotion recognition and crime levels were studied in 50 juvenile offenders. Whilst all young offenders received their statutory interventions as the study was conducted, a subgroup of twenty-four offenders also took part in a facial affect training aimed at improving emotion recognition. Offenders in the training and control groups were matched for age, SES, IQ and lifetime crime level. All offenders were tested twice for emotion recognition performance, and recent crime data were collected after the testing had been completed.

Results

Before the training there were no differences between the groups in emotion recognition, with both groups displaying poor fear, sadness and anger recognition. After the training fear, sadness and anger recognition improved significantly in juvenile offenders in the training group. Although crime rates dropped in all offenders in the 6 months following emotion testing, only the group of offenders who had received the emotion training showed a significant reduction in the severity of the crimes they committed.

Conclusions

The study indicates that emotion recognition can be relatively easily improved in youths who engage in serious antisocial and criminal behavior. The results suggest that improved emotion recognition has the potential to reduce the severity of reoffending.  相似文献

Riverside vegetation is a significant factor influencing the occurrence and progress of streambed and riverbank erosion. Recent riparian management practice in Australia has focussed on re-establishing or maintaining native riparian vegetation in order to control or prevent erosion as well as regenerate or preserve the complex variety of in-stream and riverside habitats. This work presents an integrated review of field and experimental studies conducted in eastern Australia that evaluate native vegetation's role in mass failure of riverbanks. Several results of these studies have general application and include the following: (1) The presence of riparian forest on riverbanks significantly reduces the likelihood of erosion by mass failure due to reinforcement of riverbank soils by tree roots and this reduced likelihood of mass failure enables a narrower channel cross-section than would otherwise be the case for many Australian coastal streams. (2) A number of Australian tree species have apparently evolved roots that seek the permanent, summer water table in order to survive prolonged dry spells, these root systems are particularly effective in mass failure mitigation due to rooting depths that are commonly greater than 5 m and are sometimes well in excess of 20 m. (3) The so-called “Root-Area-Ratio method” of calculating the shear strength of root-reinforced soil using root tensile strength data and Waldron's [Waldron, L.J., 1977. The shear resistance of root-permeated homogenous and stratified soil. J. Soil Sci. Soc. Am. 41, 843–849] and Wu et al.’s [Wu, T.H., McKinnell, W.P., Swanston, D.N., 1979. Strength of tree roots and landslides on Prince of Wales Island, Alaska. Can. Geotech. J. 16, 19–33] simple root model leads to significant overestimation of the actual root-reinforcement due to (a) breakage or pull-out of roots that taper and narrow beneath the shear plane such that individual roots do not achieve the tensile strength calculated on the basis of root diameter at the shear plane; and/or (b) the fact that the soil mass fails progressively along the length of potential shear plane rather instantaneously across the entire shear plane.  相似文献
