We raised monoclonal antibodies against senile plaque (SP) amyloid and obtained a clone 9D2, which labeled amyloid fibrils in SPs and reacted with approximately 50/100 kDa polypeptides in Alzheimer's disease (AD) brains. We purified the 9D2 antigens and cloned a cDNA encoding its precursor, which was a novel type II transmembrane protein specifically expressed in neurons. This precursor harbored three collagen-like Gly-X-Y repeat motifs and was partially homologous to collagen type XIII. Thus, we named the 9D2 antigen as CLAC (collagen-like Alzheimer amyloid plaque component), and its precursor as CLAC-P/collagen type XXV. The extracellular domain of CLAC-P/collagen type XXV was secreted by furin convertase, and the N-terminus of CLAC deposited in AD brains was pyroglutamate modified. Both secreted and membrane-tethered forms of CLAC-P/collagen type XXV specifically bound to fibrillized Abeta, implicating these proteins in beta-amyloidogenesis and neuronal degeneration in AD.  相似文献

Phase-separated two-stage anaerobic process was examined and evaluated using artificial organic solid waste in laboratory scale. Acidogenic process, which was combined with subsequent methanogenic process using packed-bed reactor, was operated emphasizing on either hydrogen production, or solublizing efficiency of solid materials. In either effluent from hydrogenogenic, or solublizing operation, maximum allowable OLR achieved at methanogenesis was higher than the single methanogenic process. Hydrogenogenic operation was more suitable to combine methanogenic process than solublizing operation, since retention time of hydrogenogenic operation was much shorter than the solublizing operation, obtaining almost the same levels of overall removal efficiency in both COD and VSS. The combination of hydrogenogenic operation in acidogenic process and methanogenic process produced approximately 442mmoll-reactor(1)days(-1) of methane and 199mmoll-reactor(1)days(-1) of hydrogen at 25h of total retention time indicating 82% of COD removal with 96% of VSS decomposition.  相似文献

The green lacewing, Chrysoperla carnea (Stephens), is an important natural enemy of various crop pests, especially aphids. In the Japanese fauna, there are two types of larval forms, A and B, characterized by different head capsule markings. The Type A form is distributed throughout Japan, but the Type B form has a more limited distribution. Adults use abdominal vibration as a communication signal (courtship song) during mating. We analyzed oscillograms of these songs among several Japanese populations of C. carnea. The courtship songs of types A and B are distinctly different from one another. We then performed crossing tests between the two types. Copulation between same-type pairings was much more likely than between different-type pairings. We also analyzed courtship songs of European C. carnea sensu stricti, introduced to Japan as a natural enemy of crop pests. The song of these introduced green lacewings appeared to differ from either type of Japanese C. carnea. The two types of C. carnea are likely to be different species, and also distinct from C. carnea sensu stricti of Europe.  相似文献

Expansion of polyglutamine (pQ) chain by expanded CAG repeat causes dominantly inherited neurodegeneration such as Huntington disease, dentatorubral-pallidoluysian atrophy (DRPLA), and numbers of other spinocerebellar ataxias. Expanded pQ disrupts the stability of the pQ-harboring protein and increases its susceptibility to aggregation. Aggregated pQ protein is recognized by the ubiquitin proteasome system, and the enzyme ubiquitin ligase covalently attaches ubiquitin, which serves as a degradation signal by the proteasome. However, accumulation of the aggregated proteins in the diseased brain suggests insufficient degradation machinery. Ubiquitin has several functionally related proteins that are similarly attached to target proteins through its C terminus glycine residue. They are called ubiquitin-like molecules, and some of them are similarly related to the protein degradation pathway. One of the ubiquitin-like molecules, FAT10, is known to accelerate protein degradation through a ubiquitin-independent manner, but its role in pQ aggregate degradation is completely unknown. Thus we investigated its role in a Huntington disease cellular model and found that FAT10 molecules were covalently attached to huntingtin through their C terminus glycine. FAT10 binds preferably to huntingtin with a short pQ chain and completely aggregated huntingtin was FAT10-negative. In addition, ataxin-1,3 and DRPLA proteins were both positive for FAT10, and aggregation enhancement was observed upon FAT10 knockdown. These findings were similar to those for huntingtin. Our new finding will provide a new role for FAT10 in the pathogenesis of polyglutamine diseases.  相似文献

Since the 1960s, Japan has become highly dependent on foreign countries for natural resources, and the amount of managed lands (e.g. coppice, grassland, and agricultural field) has declined. Due to infrequent natural and human disturbance, early-successional species are now declining in Japan. Here we surveyed bees, birds, and plants in four human-disturbed open habitats (pasture, meadow, young planted forest, and abandoned clear-cut) and two forest habitats (mature planted forest and natural old-growth). We extended a recently developed multispecies abundance model to accommodate count data, and used the resulting models to estimate species-, functional group-, and community-level state variables (abundance and species richness) at each site, and compared them among the six habitats. Estimated individual-level detection probability was quite low for bee species (mean across species = 0.003; 0.16 for birds). Thirty-two (95% credible interval: 13–64) and one (0–4) bee and bird species, respectively, were suggested to be undetected by the field survey. Although habitats in which community-level abundance and species richness was highest differed among taxa, species richness and abundance of early-successional species were similar in the four disturbed open habitats across taxa except for plants in the pasture habitat which was a good habitat only for several exotic species. Our results suggest that human disturbance, especially the revival of plantation forestry, may contribute to the restoration of early-successional species in Japan.  相似文献

Mutations in alpha-synuclein gene cause familial form of Parkinson disease, and deposition of wild-type alpha-synuclein as Lewy bodies occurs as a hallmark lesion of sporadic Parkinson disease and dementia with Lewy bodies, implicating alpha-synuclein in the pathogenesis of Parkinson disease and related neurodegenerative diseases. Dopamine neurons in substantia nigra are the major site of neurodegeneration associated with alpha-synuclein deposition in Parkinson disease. Here we establish transgenic Caenorhabditis elegans (TG worms) that overexpresses wild-type or familial Parkinson mutant human alpha-synuclein in dopamine neurons. The TG worms exhibit accumulation of alpha-synuclein in the cell bodies and neurites of dopamine neurons, and EGFP labeling of dendrites is often diminished in TG worms expressing familial Parkinson disease-linked A30P or A53T mutant alpha-synuclein, without overt loss of neuronal cell bodies. Notably, TG worms expressing A30P or A53T mutant alpha-synuclein show failure in modulation of locomotory rate in response to food, which has been attributed to the function of dopamine neurons. This behavioral abnormality was accompanied by a reduction in neuronal dopamine content and was treatable by administration of dopamine. These phenotypes were not seen upon expression of beta-synuclein. The present TG worms exhibit dopamine neuron-specific dysfunction caused by accumulation of alpha-synuclein, which would be relevant to the genetic and compound screenings aiming at the elucidation of pathological cascade and therapeutic strategies for Parkinson disease.  相似文献

Because the effects of land-use change on biodiversity have primarily been examined at or below the regional scale, it remains unclear whether such effects scale up to the macroecological scale (i.e. nationwide or continental scale). In Japan, forests have become more mature since the cessation of most forestry efforts in the 1970s. At a nationwide scale, this forest maturation may lead to reductions in the abundance of species that depend on early successional forests (early successional species) and increases in the abundance of species that depend on mature forests (mature forest species). Japan has met its high demand for wood through imports from South-east Asia, resulting in deforestation there. Therefore, the abundance of mature forest species that migrate long distances to overwinter in South-east Asia may decrease. We examined changes in the range sizes of birds in Japan over the past 20 years using the living planet index (LPI). The LPI indicated that the range sizes of early successional species decreased. For mature forest species, the range sizes of long-distance migrants decreased, whereas those of short-distance migrants and residents increased. Our predictions were generally supported. Our results indicate that the effects of land-use change extend to the macroecological scale and that such changes in one country can affect the biodiversity dynamics in other countries. Forest maturation in Japan and concomitant deforestation in South-east Asia have been caused by internationally coupled socioeconomic processes. Therefore, biodiversity conservation at the macroecological scale must consider the role of land use, and such efforts will require both international and socioeconomic perspectives.  相似文献
