High-sensitivity recording techniques demonstrate a continuousrelation between the onset and magnitude ot tension and themembrane depolarization that is induced by increasing K in thebathing medium or by intracellularly applied outward currents.This finding is not consistent with the mechanism of signallinge-c coupling by electrotonic spread of a "critical" depolarizationinward along the membrane of the transverse tubular system.It is in accord, however, with the channelled current mechanismthat is based on the known anion-permselectivity of the membranein the terminals of the TTS. The channelled-current model alsopredicts a direct role of Cl and a possible interaction betweenCa and CI in e-c coupling. The initiation and maintenance oftension as well as its magnitude, are in fact dependent uponthe concentrations of Ca and Cl in the medium. Thus, both thesignalling to, and the activation of, the contractile systemappear to be performed by a flow of current in the loop: cellmembrane – cell interior – TTS membrane –TTS channels – exterior, as is envisaged in the channelled-currentmodel of e-c coupling.  相似文献

1. The major aim of this study was to test the hypothesis that nutrient enrichment and the introduction of the Nile tilapia (Oreochromis niloticus), an exotic omnivorous filter‐feeding fish, operate interdependently to regulate plankton communities and water transparency of a tropical reservoir in the semi‐arid northeastern Brazil. 2. A field experiment was performed for 5 weeks in 20 enclosures (9.8 m3) to which four treatments were randomly allocated: tilapia addition (F), nutrient addition (N), tilapia and nutrient addition (F + N) and a control treatment with no tilapia or nutrient addition (C). A two‐way repeated measures anova was undertaken to test for time, tilapia and nutrient effects and their interactions on water transparency, total phosphorus and total nitrogen concentrations, phytoplankton biovolume and zooplankton biomass. 3. Nutrient addition had no effect except on rotifer biomass, but there were significant fish effects on the biomass of total zooplankton, copepod nauplii, rotifers, cladocerans and calanoid copepods and on the biovolume of total phytoplankton, large algae (GALD ≥ 50 μm), Bacillariophyta and Zygnemaphyceae and on Secchi depth. In addition, we found significant interaction effects between tilapia and nutrients on Secchi depth and rotifers. Overall, tilapia decreased the biomass of most zooplankton taxa and large algae (diatoms) and decreased water transparency, while nutrient enrichment increased the biomass of rotifers, but only in the absence of tilapia. 4. In conclusion, the influence of fish on the reservoir plankton community and water transparency was significant and even greater than that of nutrient loading. This suggests that biomanipulation of filter‐feeding tilapias may be of importance for water quality management of eutrophic reservoirs in tropical semi‐arid regions.  相似文献

1. The freshwater calanoid copepod Boeckella gibbosa is typical of high elevation lakes and ponds in Patagonia (Argentina). Previous studies have shown that this species is highly tolerant to short-term exposure to natural and artificial UVB radiation, and that its tolerance is due to photoreactivation by longer wavelength radiation. In this study, we investigate the potential sublethal effects of solar radiation after prolonged exposure.
2. We incubated B. gibbosa at 1 m depth in oligotrophic Lake Toncek for 24 days. The incubation chambers were 1.2 l acrylic cylinders covered with appropriate filters in order to obtain three radiation treatments: visible radiation only, visible radiation + UVA and visible radiation + UVA + UVB.
3. The three treatments did not differ significantly in variables considered as indicators of survival (number of individuals), reproduction (proportion of ovigerous females, clutch size) and development (instar composition). Although resistance to solar UVB radiation is certainly a requisite to live in transparent high elevation habitats, the fact of being effectively exposed to natural levels of UVB radiation does not seem to have measurable consequences on an already adapted species, such as B. gibbosa  相似文献

We developed six new microsatellite loci for the black mangrove (Avicennia germinans), an important member of wetland communities worldwide. Loci showed moderate to high polymorphism and a survey of four locations [Puerto Rico (Jobos Bay and Luquillo), Mexico, French Guyana] revealed clear regional (and local) population structure. All populations were genetically distinct and the two continental populations showed much higher diversity than the two insular Puerto Rican locations. These loci complement those recently published by Nettel et al. (2005 ) and promise to be valuable for characterizing local and regional population dynamics in the black mangrove.  相似文献

1. Mycosporine‐like amino acids (MAAs) are ubiquitous compounds in aquatic organisms that are usually considered sunscreens that protect them from harmful ultraviolet radiation. Given that virtually all animals lack the metabolic pathways to synthesise MAAs de novo, they must acquire them either from their diet or from microorganisms living in close association. In freshwater copepods, accumulation of MAAs is stimulated by exposure to ultraviolet and/or visible radiation. 2. A 2 × 2 factorial experiment was performed to assess the contributions of dietary and microbial sources of MAAs in the freshwater copepod Boeckella antiqua. The treatments consisted of two different diets: an MAA‐free diet, including only Chlamydomonas reinhardtii, and an MAA‐rich diet, including both C. reinhardtii and Peridinium inconspicuum, crossed with two antibiotic treatments, with and without chloramphenicol. Treatment with chloramphenicol was intended to inhibit the development of bacteria associated with the copepods. 3. MAA concentration in B. antiqua was affected by the experimental conditions: (i) exposure to artificial PAR + UVR stimulated the accumulation of several MAAs (up to 62% increase in total MAA concentration with respect to the initial concentration); (ii) the presence of chloramphenicol in the culture media reduced the MAA concentration in copepods fed an MAA‐free diet; (iii) in the absence of chloramphenicol, copepods fed the MAA‐rich diet had significantly higher total MAA concentration than those fed the MAA‐deficient diet; but (iv) dietary supplementation with an MAA‐rich algae in the presence of chloramphenicol failed to significantly increase total MAA concentration. 4. Analysis of profiles from denaturing gradient gel electrophoresis (DGGE) showed that the prokaryotic community associated with the copepods was affected by chloramphenicol. Dendograms constructed from digitalised DGGE images consistently grouped the antibiotics treatments separately from the initial samples and the treatments without antibiotics. Two band positions were exclusive to treatments without antibiotics. 5. We conclude that when offered an MAA‐rich diet, B. antiqua may accumulate a proportion of MAAs from diet. However, we suspect that in the absence of an MAA‐rich dietary source (as in its natural habitat), virtually all MAAs present in B. antiqua are produced by copepod‐associated prokaryotes.  相似文献

Although physical features of the environment are known to influence shape, to date, the effects of many environmental conditions on asymmetry remain unexamined. The present study aimed to examine the relationship between asymmetry and the fluid media (air versus water) that an organism inhabits and, subsequently, to use these findings to test a novel hypothesis that fluid environments can impose selective forces acting to constrain asymmetry to maintain or enhance biomechanical function. We examined carapace asymmetry in 114 species (69 aquatic and 45 terrestrial) from the chelonian superfamily Testudinoidea. The results obtained indicate that environment is correlated with the degree of asymmetry, but in different directions for the two clades that comprise the Testudinoidea. Within the Testuguria, aquatic turtles have lower levels of asymmetry than terrestrial turtles, which is consistent with our proposed biomechanical hypothesis. This pattern was not observed within the Emydidae, possibly due to the much shorter time that terrestrial taxa in this clade have existed. The discovery of cryptic species (i.e. two or more distinct but morphologically undistinguishable species) has grown exponentially in the last two decades, due mainly to the increasing availability of DNA sequences. This suggests that hidden in the known species, many of which have been described based solely on morphological information, there might be a high number of species waiting to be discovered. In this issue Damm et al. (2010) use a combination of genetic, morphological and ecological evidence to identify the first cryptic species complex found within dragonflies (insect order Odonata). Their findings add more evidence for the importance of combining information from different disciplines to new species’ discovery ( DeSalle et al. 2005 ).  相似文献

A survey of protozoa polluting bottled mineral water in Mexico was carried out using samples obtained from the three best-selling brands of bottled mineral water in the country. The organisms were concentrated through filtration procedures and subsequently cultured in sterile media. The cultures were observed over four weeks, with identification to the level of genus and species. Most commonly found were the amoebae Naegleria gruberi, Acanthamoeba astronyxis, and Vahlkampfia vahlkampfi (trophic as well as cystic stages) plus one flagellate, Bodomorpha minima. No ciliates were detected. The public health importance of the findings is obvious, since some strains of Naegleria and Acanthamoeba have the potential to cause human disease that may lead to death.  相似文献
