SYNOPSIS. A current model concerning the process of limb regenerationin vertebrates is examined. According to this model (Bryantet al, 1981), new positional values in the proximal-distal limbaxis are laid down as a result of local interactions betweencells in the limb circumference. Cells with disparate circumferentialpositional values come together at the site of future outgrowthand intercalation between them generates more distal levelsof the pattern. The results of a number of experiments on surgicallycreated symmetrical limb stumps are discussed in relation tothis model. In addition, an extension of this model to accountfor digit formation is presented, and the implications of thisformulation for limb evolution are discussed.  相似文献

Many of the factors and mechanisms guiding the migration/differentiation of neural crest cells that give rise to a number of distinguishable cell types, including all dermal and epidermal pigment cells, remain unknown. The axolotl possesses three pigment cell types that differentiate according to specific developmentally programmed sequences and contribute to pigment pattern in the adult. A single lineage of the crest that becomes restricted to one of three pigment cell types gives us the opportunity to examine the existence of a neural crest stem cell population and the potential for transdifferentiation events. Interpretations of experiments involving drug-treated and mutant axolotls implicate cellular plasticity leading to observed phenotypes. We present results from recent in vitro studies designed to identify parameters influencing differentiation events of individual neural crest-derived pigment cell lineages. We demonstrate that the differentiation of xanthophores is enhanced, while that of the melanophores are inhibited in guanosine-supplemented neural crest cell cultures. Data suggest that the increase in one pigment cell population is at the expense of another, indicative of cellular plasticity. Videomicroscopy used in this study agrees with an abundance of correlative evidence supporting the hypothesis of transdifferentiation events among neural crest-derived pigment cell populations. The embryonic neural crest-derived pigment cell system is an ideal model to study differentiation of multipotential stem cells that play critical roles in patterning.  相似文献

We have isolated 10 polymorphic microsatellite markers for the black‐bellied seedcracker (Pyrenestes ostrinus) from genomic libraries enriched either for (AAGG)n or (ATCT)n repetitive elements and characterized them in 39 individuals. The number of alleles ranged from two to 27 per locus with the observed heterozygosity ranging from 0.38 to 0.94. These markers will be useful for analysis of questions concerning parentage and population genetic structure.  相似文献

The effects of ambient and elevated ozone levels on growth and photosynthesis of beech ( Fagus sylvatica ) were studied by exposing seedlings in open-top chambers for one growing season to three treatments: charcoal-filtered (CF), non-filtered (NF) and non-filtered air with addition of ozone (30 ppb ozone) on clear days for 8–10 h d−1 (NF +). Ambient levels were relatively low and accumulated to an AOT40 (accumulated exposure over a threshold of 40 ppb) of 4055 ppb h (for the period 23 Apr–30 Sept). The NF + chambers received an AOT40 of 8880 ppb h. Throughout the growing season we measured growth and photosynthetic properties. The treatments did not cause strong effects: measurements of gas exchange (light-saturated assimilation rate, CO2 and light-response curves) and chlorophyll fluorescence showed slight and mostly non-significant reductions of several parameters. No significant differences were found for growth, though in the NF + treatment (AOT40 8880 ppb h) the relative growth rate for diameter increment was at times reduced by 12% compared with the control treatment.  相似文献

The incidence of severe neurologic complications from spinal anesthesia in San Diego County in a series of 32,882 cases in a five-year period was 0.012 per cent. Morbidity of 1 in 10,000 cases compares favorably with morbidity associated with general anesthesia. Meticulous technique and abandonment of continuous spinal anesthesia can further reduce the incidence of complications. More emphasis should be placed on the elimination of preexisting neurologic disease as a contraindication to spinal anesthesia.  相似文献

ABSTRACT Until large numbers of bat fatalities began to be reported at certain North American wind energy facilities, wildlife concerns regarding wind energy focused primarily on bird fatalities. Due in part to mitigation to reduce bird fatalities, bat fatalities now outnumber those of birds. To test one mitigation option aimed at reducing bat fatalities at wind energy facilities, we altered the operational parameters of 21 turbines at a site with high bat fatalities in southwestern Alberta, Canada, during the peak fatality period. By altering when turbine rotors begin turning in low winds, either by changing the wind-speed trigger at which the turbine rotors are allowed to begin turning or by altering blade angles to reduce rotor speed, blades were near motionless in low wind speeds, which resulted in a significant reduction in bat fatalities (by 60.0% or 57.5%, respectively). Although these are promising mitigation techniques, further experiments are needed to assess costs and benefits at other locations.  相似文献

The neural crest of vertebrate embryos has been used to elucidate steps involved in early embryonic cellular processes such as differentiation and migration. Neural crest cells form a ridge along the dorsal midline and subsequently they migrate throughout the embryo and differentiate into a wide variety of cell types. Intrinsic factors and environmental cues distributed along the neural tube, along the migratory pathways, and/or at the location of arrest influence the fate of neural crest cells. Although premigratory cells of the cranial and trunk neural crest exhibit differences in their differentiation potentials, premigratory trunk neural crest cells are generally assumed to have equivalent developmental potentials. Axolotl neural crest cells from different regions of origin, different stages of development, and challenged with different culture media have been analyzed for differentiation preferences pertaining to the pigment cell lineages. We report region-dependent differentiation of chromatophores from trunk neural crest at two developmental stages. Also, dosage with guanosine produces region-specific influences on the production of xanthophores from wild-type embryos. Our results support the hypothesis that spatial and temporal differences among premigratory trunk neural crest cells found along the anteroposterior axis influence developmental potentials and diminish the equivalency of axolotl neural crest cells.  相似文献

The phylogenetic position of the trichomonad, Histomonas meleagridis was determined by analysis of small subunit rRNAs. Molecular trees including all identified parabasalid sequences available in data bases were inferred by distance, parsimony, and likelihood methods. All reveal a close relationship between H. meleagridis, and Dientamoeba fragilis. Moreover, small subunit rRNAs of both amoeboid species have a reduced G + C content and increased chain length relative to other parabasalids. Finally, the rRNA genes from H. meleagridis and D. fragilis share a recent common ancestor with Tritrichomonasfoetus, which exhibits a more developed cytoskeleton. This indicates that Histomonas and Dientamoeba secondarily lost most of the typical trichomonad cytoskeletal structures and hence, do not represent primitive morphologies. A global phylogeny of parabasalids revealed significant discrepancies with morphology-based classifications, such as the polyphyly of most of the parabasalid families and classes included in our study.  相似文献
