Electrophoretic separation of the protein extracts of the whole accessory reproductive gland (ARG) reveals a progressive increase in number of fractions from day 0 to day 12 of adult life. The number of fractions secreted by the different types of tubules, viz., white tubules, hyaline tubules and seminal vesicles, varies. The protein concentration of the ARG of 12-day-old insects is four times higher than that of newly emerged adult insects. Three protein fractions of the ARG extract are immunologically precipitatale by antiserum to whole accessory gland complex. Immunoelectrophoresis of the oocyte extract from mated females and total protein extract of empty spermatophores revealed that one fraction of the oocyte protein and two of the spermatophores have their immunological counterparts in the whole ARG extract.  相似文献

Although it is well established that the root growth in manyspecies is very sensitive to mechanical impedance or to confinementin small volumes, little is known about the consequent effectson growth of the whole plant and the mechanisms involved. Thiswork investigated the effects of root confinement on the waterrelations, growth and assimilate partitioning of tomato (Lycopersiconesculentum Mill) grown in solution culture. Six-week old plants were transferred to either 4500 ml or 75ml containers filled with nutrient solution, and allowed togrow for 14 d. Transpiration, leaf-air temperature differences,and leaf diffusive resistances were measured frequently. Leaf,stem and shoot dry masses, leaf area and root length, were estimatedwhen the treatments were imposed and at the end of the experiment.After 14 d growth the root and shoot hydraulic resistances wereestimated from measurements of leaf water potential and transpirationrate, using a steady-state technique. Confining root growth to the small containers substantiallyreduced shoot and root growth and increased the proportion oftotal dry matter present in the stems. These effects were dueto drought stress. The hydraulic resistance of the root systemwas greatest in the confined plants. This led to more negativeleaf water potentials, increased leaf diffusive resistance,and reduced the net assimilation rate by a factor of 2.5. Transpirationper unit leaf area was less affected. However, cumulative transpirationwas also reduced by a factor of 2.5. mostly because of the smallerleaf area on the confined plants. Root hydraulic resistivitywas measured at 3.1 x 1012s m–1 in the control treatment,but increased to 3.9 x 1012 s m–1 for roots in the smallcontainer. The mechanisms by which root confinement caused drought stressand disrupted the pattern of assimilate partitioning are discussedin detail. Susceptibility of field populations of Helicoverpa armigera (Lepidoptera: Noctuidae) from Punjab, Pakistan to selected insecticides was evaluated using the leaf dip method. The resistance factors varied between populations. The general trends were low to moderate resistance to endosulfan; very low to low resistance to chlorpyrifos and profenofos; very low resistance to spinosad, indoxacarb and thiodicarb; moderate to high resistance to bifenthrin; high to very high resistance to cypermethrin; and very high resistance to lambda-cyhalothrin. Integrated pest management strategies aimed at reducing pesticide application, such as rotating the conventional insecticides having low resistance with newly developed synthetic insecticides and conserving natural enemies, are recommended.  相似文献

A stock culture of cotton leaf curl virus from Pakistan (CLCuV-PK), was transmitted by whiteflies (Bemisia tabaci) to seven plant species, including French bean, okra, tobacco and tomato, and caused vein thickening and leaf curl symptoms. It was readily detected in triple antibody sandwich ELISA (TAS-ELIS A) by 11 out of 31 monoclonal antibodies raised against the particles of three other geminiviruses: African cassava mosaic, Indian cassava mosaic and okra leaf curl viruses. Reaction strength was enhanced when the tissue extraction fluid contained sodium sulphite. Minor variations in epitope profile were found among virus isolates from cotton (Gossypium hirsutum) collected from different districts in Pakistan over a 5-year period. These epitope profiles were distinguishable from that of cotton leaf curl virus from G. barbadense in southern India but indistinguishable from the profiles of viruses causing yellow vein disease of okra in India or Pakistan, or leaf curl of okra {Abelmoschus esculentus), Hibiscus tiliaceus, radish or sunflower in Pakistan, suggesting that these plants are putative natural hosts of CLCuV-PK. The viruses in cotton, and in okra with leaf curl or yellow vein symptoms, were also detected by PCR with three pairs of CLCuV-PK-specific primers. Five additional whitefly-transmitted geminiviruses were found among isolates from 11 other naturally-infected species in Pakistan, and were distinguished by their epitope profiles. These viruses were associated, respectively, with tobacco leaf curl, squash yellow blotch, tomato yellow leaf curl, watermelon leaf crinkle and soybean yellow mosaic diseases. The first four of these viruses were detected readily by PCR with geminivirus general primers but only weakly, if at all, with two pairs of CLCuV-PK-specific primers. Pakistani crops are infected with a range of distinguishable but relatively closely related whitefly-transmitted geminiviruses, some of which resemble those found in India.  相似文献
