Summary A major pathogen of potato plants (Solanum tuberosum) is the potato cyst nematode (Globodera spp.), which induces localized redifferentiation of a limited number of host cells to form a specialized feeding-site termed the syncytium. A novel strategy utilizing the polymerase chain reaction (PCR) was employed to construct a cDNA library from dissected potato roots highly enriched in syncytial material. The library was differentially screened with cDNA probes derived from the infected root tissue from a compatible interaction and from healthy root tissue. Characterization of one gene identified by the library screen indicated an expression pattern that correlated with events in the immediate vicinity of the pathogen after syncytial establishment. The strategy for library construction and screening could be applicable to the study of gene expression in any plant-pathogen interaction in which the limited supply of cells at the interface of the two organisms precludes a more traditional approach.  相似文献

Summary The induction and decay of ornithine decarboxylase (ODC) by insulin and asparagine in cultures of H4-II-EC3 (H35) hepatoma cells was studied in a modified Waymouth medium in the presence of fetal bovine serum (FBS) and in serum-free media. The insulin response was enhanced by the presence of asparagine although the effect of asparagine was not so much on the initial increase as it was on a slowing of the decline after the maximum was reached at 6 to 8 h after the supplements were added together with fresh medium. In all cases the initial ODC activity was zero at zero time for addition of media and supplements, and, after reaching the maximum, activity declined to near zero by 24 h. Fetal bovine serum gave induction that followed a similar time course but was inferior to the combintion, of insulin plus asparagine and, in fact, FBS inhibited the latter response. Putrescine (the product formed from ornithine by ODC), at 10−5 M, markedly inhibited the induction of ODC by insulin or FBS, but the inhibition was less when asparagine was present. 5'AMP-activated protein kinase (AMPK) activation occurs under a variety of stress conditions but the role of this enzyme in the promotion or inhibition of stress-induced cell death is unclear. To address this issue, we transformed two different cell lines with shRNA-expressing plasmids, targeting the alpha subunit of AMPK, and verified AMPKalpha downregulation. The cell lines were then stressed by exposure to medium without glucose (PC12 cells) or with the viral thymidine kinase-specific DNA replication inhibitors: acyclovir, penciclovir and ganciclovir (herpes simplex virus thymidine kinase-expressing Baby Hamster Kidney cells). In non-AMPK-downregulated cells, these stress treatments induced AMPK upregulation and phosphorylation, leaving open the question whether the association of AMPK activation with stress-induced cell death reflects a successful death-promoting or an ineffective death-inhibiting activity. In AMPKalpha-deficient cells (expressing AMPKalpha-specific shRNAs or treated with Compound C) exposure to low glucose medium or DNA replication inhibitors led to an enhancement of cell death, indicating that, under the conditions examined, the role of activated AMPK is not to promote, but to protect from or delay stress-induced cell death.  相似文献

Many insects have coevolved with certain angiosperm taxa to act as pollinators. However, the nectar and pollen from such flowers is also widely fed upon by other insects, including entomophagous species. Conservation biological control seeks to maximise the impact of these natural enemies on crop pests by enhancing availability of nectar and pollen-rich plants in agroecosystems. A risk with this approach is that pests may also benefit from the food resource. We show that the flowers of some plants (viz., buckwheat, Fagopyron esculentum Moench and dill, Anethum graveolens L.), and the extrafloral nectaries of faba bean (Vicia faba L.) benefit both Copidosoma koehleri Blanchard (Hymenoptera: Encyrtidae) and its host, the potato pest, Phthorimaea operculella Zeller (Lepidoptera: Gelechiidae). In contrast, phacelia (Phacelia tanacetifolia Benth) and nasturtium (Tropaeoleum majus L.) benefited only the parasitoid. When adult moths of P. operculella were caged with flowers of phacelia or nasturtium, longevity of males and females, egg laying life, fecundity, average oviposition rate, and number of eggs in ovaries at death were no greater than in the control treatment with access to shoots without flowers plus water. All the foregoing measures were increased compared to the control when the moths were allowed access to dill, buckwheat or faba bean extrafloral nectaries. Such selectivity has the potential to make the use of floral resources in conservation biological control more strategic. We present morphometric and observational evidence to illustrate how such mechanisms may operate.  相似文献

Arsenic can either enhance or reduce nitric oxide (NO) production, depending on the type of cell, the species and dose of arsenical tested. The mechanisms of how arsenic increases or decreases NO production remain unclear. Because NO is associated with many pathological conditions, it is conceivable that in those arsenic-target tissues, the NO production may be upregulated by continuous arsenic exposure, and a prolonged over-production of NO may cause inflammation hence a pathological condition. A prolonged interference with the normal physiological level of NO may also play a role in the initiation, promotion, and progression of arsenic-related human cancers. Suppression of NO production has been shown to reduce arsenite-induced oxidative DNA damage, inhibition of pyrimidine dimer excision, and micronuclei. However, a completely reliable story on how NO is involved in arsenic-related human disease is still lacking.  相似文献

Mitogen-activated protein (MAP) kinases represent a group of serine/threonine kinases which play a pivotal role in signal transduction processes in eukaryotic cells. Using degenerate PCR primer design based on published and aligned MAP kinase sequences we have cloned and characterised two MAP kinase genes from the barley powdery mildew fungus, Blumeria graminis. We have utilised 'step down' PCR to attain the full length mildew genomic clones. The single-copy genes, named mpk1 and mpk2, encode putative proteins of 356 and 410 amino acids and carry three and four introns, respectively. Expression studies, using RT-PCR, reveal a differing pattern of tissue gene expression with mpk1 and mpk2 during germling morphogenesis and this is compared with the constitutive expression of the 'control' beta-tubulin gene.  相似文献
