From the soluble and membrane fractions of rat brain homogenate, two enzymes that liberate dipeptides of the type Xaa-Pro from chromogenic substrates were purified to homogeneity. The two isolated dipeptidyl peptidases had similar molecular and catalytic properties: For the native proteins, molecular weights of 110,000 were estimated; for the denatured proteins, the estimate was 52,500. Whereas the soluble peptidase yielded one band of pI 4.2 after analytical isoelectric focusing, two additional enzymatic active bands were detected between pI 4.2 and 4.3 for the membrane-associated form. As judged from identical patterns after neuraminidase treatment, both peptidases contained no sialic acid. A pH optimum of 5.5 was estimated for the hydrolysis of Gly-Pro- and Arg-Pro-nitroanilide. Substrates with alanine instead of proline in the penultimate position were hydrolyzed at comparable rates. Acidic amino acids in the ultimate N-terminal position of the substrates reduced the activities of the peptidases 100-fold as compared with corresponding substrates with unblocked neutral or, especially, basic termini. The action of the dipeptidyl peptidase on several peptides with N-terminal Xaa-Pro sequences was investigated. Tripeptides were rapidly hydrolyzed, but the activities considerably decreased with increasing chain length of the peptides. Although the tetrapeptide substance P 1-4 was still a good substrate, the activities detected for the sequential liberation of Xaa-Pro dipeptides from substance P itself or casomorphin were considerably lower. Longer peptides were not cleaved. The peptidases hydrolyzed Pro-Pro bonds, e.g., in bradykinin 1-3 or 1-5 fragments, but bradykinin itself was resistant. The enzymes were inhibited by serine protease inhibitors, like diisopropyl fluorophosphate or phenylmethylsulfonyl fluoride, and by high salt concentrations but not by the aminopeptidase inhibitors bacitracin and bestatin. Based on the molecular and catalytic properties, both enzymes can be classified as species of dipeptidyl peptidase II (EC 3.4.14.2) rather than IV (EC 3.4.14.5). However, some catalytic properties differentiate the brain enzyme from forms of dipeptidyl peptidase II of other sources.  相似文献

The genus Roccellinastrum Follm. emend. Henssen & Vobis is placed in the Lecideaceae. Two new species are described, R. epiphyllum Henssen & Vobis, from Chile and R. neglectum Henssen & Vobis, from New Zealand and Tasmania. The new combination R. candidum (Mull. Arg.) Henssen is proposed. Roccellinastrum is characterized by apothecia of diverse shape without a proper margin, and a developmental morphology corresponding to that of the Lecanorales, by small amyloid asci, branched paraphyses and excipular hyphae, a byssoid–spon–giose thallus composed of thick–walled hyphae, and members of the Chlorococcales as phycobionts. The principal chemical constituent is protocetraric acid. The species have different spore septation and chemistry. Summary in Spanish.  相似文献

The aim of the present study was to evaluate phytohormone production by symbiotic and saprophytic actinomycetes isolated from the actinorhizal plant Ochetophila trinervis which had previously proved to stimulate nodulation by Frankia. Three saprophytic strains out of 122, isolated from the rhizosphere of this plant with multiple enzymatic activities were selected for plant growth experiments in pots: Streptomyces sp. (BCRU-MM40), Actinoplanes sp. (BCRU-ME3) and Micromonospora sp. (BCRU-MM18). For experiments, the symbiotic N2-fixing strain Frankia (BCU110501), isolated from nodules of the same actinorhizal plant was used. Phytohormone production was evaluated in supernatant of non-inoculated and inoculated culture media in exponential growth phase. Indole 3-acetic acid (IAA) and gibberellic acid (GA3) were analyzed by gas chromatography-mass spectrometry (GC–MS), while zeatine (Z) production was determined by gas chromatography-flame ionization detector and high performance liquid chromatography (HPLC fluorescent and UV). The levels of the three phytohormones produced by the saprophytic rhizoactinomycetes were higher than that produced by the symbiotic Frankia strain. Zeatine biosynthesis was higher (μg ml−1) than IAA and GA3 (ng ml−1), and Micromonospora strain produced the highest levels of these phytohormones. Although O. trinervis has been shown to be intercellularly infected by Frankia without mediation of root hair deformation, when plants were co-inoculated with actinomycetes’ culture, some root hair deformation was observed. This is the first report on identification of IAA, GA3 and Z in saprophytic actinomycetes and their potential role in plant–microbe interaction.  相似文献

Zusammenfassung 1. Zur Untersuchung kleinerer Wassertiere wurde eine Durchflußapparatur entwickelt, die eine stufenlose Regelung einer gerichteten Strömung sowie die Einstellung einer konstanten Gaskonzentration erlaubt.2. Das rheotaktische Verhalten vonGammarus pulex, G. roeseli, G. fossarum, G. tigrinus undG. salinus wurde bei einer Temperatur von 15° C unter verschiedenen Sauerstoffkonzentrationen getestet.3. Die höchste rheotaktische Aktivität der untersuchten Amphipoden wurde bei geringen Strömungsgeschwindigkeiten (5 cm/sec) konstatiert.4. Diese Aktivität erfuhr mit abnehmendem O2-Gehalt eine Steigerung bis zu einem Aufwanderungsmaximum, das bei einer kritischen O2-Konzentration lag. Noch geringerer Sauerstoffgehalt bewirkte Abwanderung in Strömungsrichtung, rasche Aktivitätsabnahme und führte meist zum Tod der Versuchstiere.5. Die so definierte kritische O2-Konzentration lag fürGammarus pulex bei 2,7 mg O2/l fürG. roeseli bei 3,1 mg O2/l, fürG. fossarum bei 5,3 mg O2/l und für die BrackwasserformG. tigrinus bei 3,5 mg O2/l. Die euryhaline ArtG. salinus hatte keine ausgeprägte Rheotaxis; lediglich die Grenze vom Auf- zum Abwandern konnte definiert werden (2,5 mg O2/l).
Rheotactic behaviour of someGammarus species in different oxygen concentrations of the water
An experimental vessel has been developed which makes it possible to observe the behaviour of gammarids and other aquatic invertebrates in various water current speeds and oxygen concentrations. Studies have been conducted with the following limnic and brackish-water amphipods:Gammarus pulex (L.),G. roeseli Gervais,G. fossarum Koch,G. tigrinus Sexton andG. salinus Spooner. In adequate oxygen concentrations, the gammarids tested show a moderate positive rheotaxis. Lethal and sublethal oxygen concentrations, however, lead to negative rheotaxis. The maximum degree of upstream movement is attained in the region of the critical oxygen concentration. Under the experimental conditions (15° C, water current 5 cm/sec), these maxima vary according to species. The maximum occurred at 2.7 mg O2/l forG. pulex, 3.1 mg O2/l forG. roeseli, 3.5 mg O2/l forG. tigrinus, and 5.3 mg O2/l forG. fossarum. No such maximum was found forG. salinus. Oxygen deficit causesG. salinus to move downstream at 2.5 mg O2/l.

The suppressive function of regulatory T cells (T(reg)) is impaired in multiple sclerosis (MS) patients. The mechanism underlying the T(reg) functional defect is unknown. T(reg) mature in the thymus and the majority of cells circulating in the periphery rapidly adopt a memory phenotype. Because our own previous findings suggest that the thymic output of T cells is impaired in MS, we hypothesized that an altered T(reg) generation may contribute to the suppressive deficiency. We therefore determined the role of T(reg) that enter the circulation as recent thymic emigrants (RTE) and, unlike their CD45RO(+) memory counterparts, express CD31 as typical surface marker. We show that the numbers of CD31(+)-coexpressing CD4(+)CD25(+)CD45RA(+)CD45RO(-)FOXP3(+) T(reg) (RTE-T(reg)) within peripheral blood decline with age and are significantly reduced in MS patients. The reduced de novo generation of RTE-T(reg) is compensated by higher proportions of memory T(reg), resulting in a stable cell count of the total T(reg) population. Depletion of CD31(+) cells from T(reg) diminishes the suppressive capacity of donor but not patient T(reg) and neutralizes the difference in inhibitory potencies between the two groups. Overall, there was a clear correlation between T(reg)-mediated suppression and the prevalence of RTE-T(reg), indicating that CD31-expressing naive T(reg) contribute to the functional properties of the entire T(reg) population. Furthermore, patient-derived T(reg), but not healthy T(reg), exhibit a contracted TCR Vbeta repertoire. These observations suggest that a shift in the homeostatic composition of T(reg) subsets related to a reduced thymic-dependent de novo generation of RTE-T(reg) with a compensatory expansion of memory T(reg) may contribute to the T(reg) defect associated with MS.  相似文献

Changes in the infectious capacity of soilborne Frankia from the same site may depend on environmental conditions. To test this, we examined the effect of season of sampling, sample storage protocol and storage time. The nodulation capacity of Frankia from rhizospheric soils of Discaria trinervis (Hook et Arn.) Reiche (Rhamnaceae) growing in northwest Patagonia (Argentina) was measured using the most probable number method. Soil samples were collected seasonally and either stored moist at 4°C or air dried at room temperature for few days. Old (air-dried) soil samples were also assayed. All soils nodulated D. trinervis seedlings. Nodulation units (NU) ranged from 44 (spring, moist storage) to about 1 ml−1 of soil (summer moist, and summer and autumn, air-dried storage), with intermediate values in other samples. Soils stored for 12 years, 6 months or 1 week had similar NU. Frankia NU positively correlated with soil water content ( r = 0.6, P < 0.05); therefore, it is likely that soil moisture is a relevant factor regulating soilborne Frankia nodulation ability in Patagonian soils. We suggest that Frankia can remain as spores or grow saprophytically in Patagonian soils.  相似文献

The type strain of Streptomyces torulosus Lyons and Pridham (1971) was studied by scanning- and transmission electron microscope. Spore chains were formed in spirals by aerial mycelium. The spores were connected by nozzles in which small channels could be observed. The knobby ornamentations of the spores arised on a thin fibrous sheath, enveloping the spore chains. These irregular blunt projections, called knobs, had varying diameters of 100 to 250 nm. The base of the knob, consisting of globose to flattened electron dense material, was sitting directly on the sheath. It was covered by several small vesicles of the same material. Each hollow vesicle beared a thin bowlshaped shell of electron transparent material. In general, the cupular bowls and their supporting vesicles became easily depressed on their base, but not detached from the surface of the spores. This type of knobby spore ornamentation was suggested to be designated as a verrucate spore type.  相似文献

The phosphate uptake behaviour of monospecific cultures of green algae in the laboratory and of mixed phytoplankton populations in a mesotrophic lake has been analyzed with the aid of a force-flow relationship. This analysis yields two parameters:
  • 1 A conductivity coefficient, that characterizes the activity of the phosphate uptake system.
  • 2 An external threshold phosphate concentration, below which uptake of phosphate is excluded on energetic grounds.
When the phosphate concentration lies below the threshold value, the algae show an activation of the uptake system, reflected in an increase in the conductivity coefficient. Correspondingly, excess phosphate above the threshold value leads to a diminuation of the conductivity. Using this simple analysis, phosphate discharge into lake water may be readily monitored.  相似文献

Saprophytic rhizoactinomycetes isolated from the root nodule surface of the nitrogen-fixing actinorhizal plant Discaria trinervis, Streptomyces MM40, Actinoplanes ME3, and Micromonospora MM18, previously shown to stimulate nodulation in Frankia-Discaria trinervis symbiosis, were assayed as co-inoculants with Sinorhizobium meliloti 2011 on Medicago sativa. When plants were fertilized with a low level of N (0.07 mM), the inoculation of the actinomycetes alone did not show any effect on plant growth. Meanwhile, when actinomycetes were co-inoculated with S. meliloti, nodulation and plant growth were significantly stimulated compared to plants inoculated with only S. meliloti. The analysis of nodulation kinetics of simultaneously or delayed co-inoculations suggests that the effect of the actinomycetes operates in early infection and nodule development counteracting the autoregulation of nodulation by the plant. Because the actinomycete effect was found in the symbiotic nitrogen-fixing state of the plant, we investigated the effects of the actinomycetes, in single inoculation or co-inoculation with S. meliloti, on plants grown under a high level of N (7 mM) that was inhibitory for nodulation by S. meliloti. The inoculation of the actinomycetes alone did not show any effect on plant growth although high N was available. Unexpectedly, the co-inoculation of actinomycetes with S. meliloti on plants grown with high N (7 mM) significantly stimulates nodulation, clearly counteracting the inhibition of nodulation by high N. These results corroborate that the interaction of rhizoactinomycetes would interfere with the autoregulation of nodulation in alfalfa mediated by high N, opening new research lines of potential agronomical applications.  相似文献
