Summary Cell lines susceptible or resistant to the active antitumor sulfonylurea [N-(4-methylphenylsulfonyl)-N-(4-chlorophenyl)-urea] (LY 181984) were treated with 100 M sulfonylurea for 1 or 3 h followed by monensin for 1 h. With cell lines where growth was inhibited by the active sulfonylurea, swollen Golgi apparatus cisternae following treatment were fewer and smaller than in untreated cells. Overall the volume of monensin-responsive trans cisternae was reduced by about 50% to 75% in cells lines where the antitumor sulfonylurea was growth inhibitory. The swelling response was unaffected by sulfonylurea in sulfonylurea-unresponsive cells. The antitumor-inactive sulfonylurea [N-(4-methylphenylsulfonyl)-N-(phenyl)urea] (LY 181985) was without effect on cisternal swelling with both susceptible and resistant cell lines. The results suggest a response of the trans Golgi apparatus to the active antitumor sulfonylurea that resulted in reduced acidification of the trans Golgi apparatus cisternae. This response appears to be restricted to susceptible cell lines where growth was inhibited by the active antitumor sulfonylurea but not in resistant cell lines where growth was unaffected by the active antitumor sulfonylurea.  相似文献

The mechanism of the inhibition of phosphatidylcholine biosynthesis by the phospholipid analogue, hexadecylphosphocholine, was investigated in Madin-Darby canine kidney cells. In the presence of 50 mumol/liter hexadecylphosphocholine, there was a translocation of CTP:choline-phosphate cytidylyltransferase (EC 22.7.7.15) activity from the membranes to the cytosol of the cells. Since we recently demonstrated that hexadecylphosphocholine also inhibits protein kinase C in vitro, [methyl-3H]choline labeling experiments were repeated with phorbol ester-desensitized cells. In these cells the same inhibitory effect of hexadecylphosphocholine was measured. As a consequence of inhibition, the [methyl-3H]choline incorporation into the phosphocholine pool was increased time-dependently. In addition, there was no evidence for a difference between the choline uptake of control and hexadecylphosphocholine-treated cells. Likewise, the amount of diacylglycerol, a known activator of the translocation process, was not reduced. Finally, we showed that the inhibitory effect of hexadecylphosphocholine on CTP:choline-phosphate cytidylyltransferase translocation cannot be explained by the detergent properties of this phospholipid analogue. Therefore, we suggest a direct inhibitory effect of hexadecylphosphocholine on the translocation of CTP:choline-phosphate cytidylyltransferase.  相似文献

Sphingomyelin hydrolysis seems to be a ubiquitous pathway generating ceramide, an important cell response modifier. Upon agonist-stimulation this pathway is linked to biological responses as inhibition of proliferation, promotion of differentiation and induction of apoptosis. One of the agonists described is 1alpha,25-dihydroxyvitamin D3. Recently, we could demonstrate the existence of sphingomyelin hydrolysis in human primary keratinocytes as well as in the human keratinocyte cell line HaCaT after treatment with 1alpha,25-dihydroxyvitamin D3. In the present study we tested four vitamin D analogues on HaCaT keratinocytes for their ability to inhibit cell proliferation and to induce sphingomyelin hydrolysis. These analogues, calcipotriol, EB 1213, GS 1500 and tacalcitol inhibit cell growth after 48 hrs. of incubation and trigger the hydrolysis of sphingomyelin. Moreover, all analogues tested induce apoptotic cell death in HaCaT keratinocytes after 24 hrs. of incubation. This study indicates that sphingomyelin hydrolysis, subsequently leading to the elevation of cellular ceramide levels, may represent an important signal transduction pathway for 1alpha,25-dihydroxyvitamin D3 and its analogues in human keratinocytes. Possible differences of the mechanism underlying vitamin D-induced sphingomyelin hydrolysis has to be studied in more detail and may contribute to the antipsoriatic action of these analogues.  相似文献

Synthetic choline-containing phospholipids comprise a new class of compounds with antineoplastic properties. We have investigated the effect of recently synthesized glucose-containing analogs of lysophosphatidylcholine (glyceroglucophospholipid, Glc-PC) and of lysoplatelet activating factor (Glc-PAF) and its C16, C14 and C12 derivatives (ET-16, ET-14, and ET-12) on proliferation of immortalized human keratinocyte (HaCaT) cells. The data were compared to the ability of the compounds to intercalate into phosphatidylserine liposomes and to form lesions in planar bilayer membranes. A correlation between bioactivity and membrane activity was found. The number of molecules that intercalated into phosphatidylserine liposomes depended on the chemical structure of the compounds and was in the order Glc-PAF approximately ET-16 approximately ET-14 > Glc-PC > ET-12. All compounds induced membrane lesions, and the lesion forming activity was in the same order. Similar activity rankings were found for the release of lactate dehydrogenase from HaCaT cells as a measure of lytic activity and for the influence on cell number as a measure of proliferation. In the latter test, however, proliferation was already inhibited at non-toxic concentrations. From these findings, it may be concluded that the intercalation of the compounds at toxic concentrations leads to the formation of membrane lesions and finally results in membrane rupture leading to cell death.  相似文献

Non-bullous congenital ichthyosis erythroderma (NCIE) and lamellar ichthyosis (LI) are characterized by mutations in 12R-lipoxygenase (12R-LOX) and/or epidermal lipoxygenase 3 (eLOX3) enzymes. The eLOX3 lacks oxygenase activity, but is capable of forming hepoxilin-type products from arachidonic acid-derived hydroperoxide from 12R-LOX, termed 12R-hydroperoxyeicosa-5,8,10,14-tetraenoic acid (12R-HpETE). Mutations in either of two enzymes lead to NCIE or LI. Moreover, 12R-LOX-deficient mice exhibit severe phenotypic water barrier dysfunctions. Here, we demonstrate that 12R-HpETE can also be transformed to 8R-HXA(3) by hepoxilin A(3) (HXA(3)) synthase (12-lipoxygenase), which exhibits oxygenase activity. We also presented a novel form of ichthyosis in a patient, termed hepoxilin A(3) synthase-linked ichthyosis (HXALI), whose scales expressed high levels of 12R-LOX, but were deficient of HXA(3) synthase.  相似文献

Summary The stimulation of NADH oxidase activity of plasma membranes of rat liver observed with guanine nucleotides may involve both guanine nucleotide-binding proteins of the plasma membrane and responses not mediated by classic heterotrimeric G proteins. These conclusions are based on findings that detergent treatment and peptide antisera to a consensus guanine nucleotidebinding domain (GAGES) of G subunits of heterotrimeric G proteins reduced but did not eliminate the stimulation of NADH oxidase activity by guanine nucleotides. The proteins immunoprecipitated by the antisera, when added back to plasma membranes, stimulated the NADH oxidase activity. This stimulated rate was further stimulated by the addition of GTP but was not dependent upon guanine nucleotide presence. Additions of cytosol, either fractionated or unfractionated did not appear to stimulate the NADH oxidase activity of rat liver plasma membranes. The activities of the plasma membranes and the activities introduced by the cytosol fractions were nearly, but not entirely, additive. The results are suggestive of a subunit composition of the NADH oxidase but one distinct from that involving solely heterotrimeric G proteins. Also a strong dependence on cytosolic components, as found with the NADPH oxidase complex of neutrophils, is not obvious. In addition, the possibility that the NADH oxidase may exhibit an intrinsic re-sponse to guanine nucleotides, not dependent on accessory proteins, cannot be ruled out. Among the several bands immunoprecipitated with the antisera and reactive with the antisera on Western blots, were peptide bands in the molecular weight range ascribed to the NADH oxidase.  相似文献

The Bcl-2-related proteins Bcl-X(L) and Bcl-X(S) represent alternative splice products and exert opposite activities in the control of apoptosis, but their significance for melanoma is not yet clear. Applying the tetracycline-inducible expression system Tet-On, we found overexpression of Bcl-X(S) by itself sufficient to induce apoptosis in vitro in stably transfected human melanoma cell lines. Combination with proapoptotic agents such as etoposide, pamidronate, and ceramide resulted in additive proapoptotic effects, whereas Bcl-X(L) protected from apoptosis caused via CD95/Fas stimulation. In nude mice growth of melanoma xenotransplants derived from stably transfected cells was significantly reduced after induction of Bcl-X(S) by doxycycline. Our results indicate that Bcl-X proteins are of major importance for control of apoptosis in malignant melanoma.  相似文献
