Surfactant protein-A (SP-A) belongs to a family of collagen-containing C-type lectins called collectins. SP-A is expressed by renal tubule epithelial cells. We investigated the distribution of SP-A in renal cell carcinomas (RCC) using immunohistochemical techniques and western blotting. We used 35 formalin fixed, paraffin embedded (FFPE) RCC tissue samples. We compared results with clinico-pathological parameters of RCC including age, sex, Fuhrman grade, tumor volume, tumor node metastasis (TNM) and clinical stage. SP-A was localized in the glomerulus and renal tubule epithelium in nontumor tissue and strong SP-A immunoreactivity was observed in tumor tissue. SP-A was expressed in the RCC tumor cells (64%) and nontumor cells (34%) in males and RCC tumor cells (90%) and nontumor cells (30%) in females. There was a significant correlation between SP-A immunoreactivity in tumor cells and gender, age, tumor diameter, Fuhrman grade and tumor diameter. Western blot analysis supported the immunohistochemical findings. We present evidence for involvement of SP-A in RCC and suggest that increased SP-A expression in RCC is associated with favorable prognosis. 相似文献

In a three-hour bioassay, we tested the palatability and feeding preferences of Uresiphita maorialis (kōwhai moth) for Sophora tetraptera, Sophora microphylla and Sophora prostrata. Palatability tests showed no differences among the Sophora species. Feeding preferences, on the other hand, showed that S. tetraptera and S. microphylla leaves are preferred over S. prostrata leaves. Our results support our field observations in Wellington city parks and gardens showing that S. tetraptera and S. microphylla plants frequently have higher densities of larvae than S. prostrata. 相似文献

Aims
Weed control in rice is challenging, particularly in light of increased resistance to herbicides in weed populations including Echinochloa crus-galli (L.) Beauv. Indica rice cultivars can produce high yields and suppress barnyardgrass, but have not been commercially acceptable in the U.S. due to inferior agronomic traits and grain quality. Our objectives were to combine high yield and weed-suppressive characteristics from indica cultivars with commercially acceptable grain quality and plant types from long-grain cultivars grown in the southern U.S.Methods
Crosses between indica and commercial tropical japonica (cv. Katy, and cv. Drew) rice were evaluated for weed suppression and agronomic traits in a breeding program.Results
In some tests, the selection STG06L-35-061 was nearly as weed suppressive as PI 312777, the suppressive parent, and more suppressive than its tropical japonica parents. Its main crop yield is commercially acceptable, and intermediate between PI 312777 and Katy. Its milling quality and cooking quality are similar to long-grain commercial cultivars, and it has resistance to rice blast disease. Marker analyses identified introgressions from the indica parents on chromosomes 1 and 3 of STG06L-35-061 that require further analysis as possible sources of weed suppressive traits.Conclusions
STG06L-35-061 might be suitable for organic rice or reduced input conventional systems. 相似文献4.

In vitro alkaline elution is a sensitive and specific short term assay which measures DNA strand breakage in a mammalian test system (primary rat hepatocytes). This lab has previously demonstrated the performance of the assay with known genotoxic and non-genotoxic compounds. The methodology employed has relatively low sample throughput and is labor-intensive, requiring a great deal of manual processing of samples in a format that is not amenable to automation. Here, we present an automated version of the assay. This high-throughput alkaline elution assay (HT-AE) was made possible through 3 key developments: (1) DNA quantitation using PicoGreen and OliGreen fluorescent DNA binding dyes; (2) design and implementation of a custom automation system; and (3) reducing the assay to a 96-well plate format. The assay can now be run with 5-50mg of test compound. HT-AE was validated in a similar manner as the original assay, including assessment of non-genotoxic and non-carcinogenic compounds and evaluation of cytotoxicity to avoid confounding effects of toxicity-associated DNA degradation. The validation test results from compounds of known genotoxic potential were used to set appropriate criteria to classify alkaline elution results for genotoxicity. 相似文献

Keratan sulphate (KS) proteoglycans (PGs) are key molecules in the connective tissue matrix of the cornea of the eye, where
they are believed to have functional roles in tissue organisation and transparency. Keratocan, is one of the three KS PGs
expressed in cornea, and is the only one that is primarily cornea-specific. Work with the developing chick has shown that
mRNA for keratocan is present in early corneal embryogenesis, but there is no evidence of protein synthesis and matrix deposition.
Here, we investigate the tissue distribution of keratocan in the developing chick cornea as it becomes compacted and transparent
in the later stages of development. Indirect immunofluorescence using a new monoclonal antibody (KER-1) which recognises a
protein epitope on the keratocan core protein demonstrated that keratocan was present at all stages investigated (E10–E18),
with distinct differences in localisation and organisation observed between early and later stages. Until E13, keratocan appeared
both cell-associated and in the stromal extracellular matrix, and was particularly concentrated in superficial tissue regions.
By E14 when the cornea begins to become transparent, keratocan was located in elongate arrays, presumably associated along
collagen fibrils in the stroma. This fibrillar label was still concentrated in the anterior stroma, and persisted through
E15–E18. Presumptive Bowman’s layer was evident as an unlabelled subepithelial zone at all stages. Thus, in embryonic chick
cornea, keratocan, in common with sulphated KS chains in the E12–E14 developmental period, exhibits a preferential distribution
in the anterior stroma. It undergoes a striking reorganisation of structure and distribution consistent with a role in relation
to stromal compaction and corneal transparency.

The present study investigated the effect of unilateral and bilateral resistance exercise (RE) on maximal voluntary strength, total volume of load lifted (TVLL), rating of perceived exertion (RPE) and blood lactate concentration of resistance-trained males. Twelve healthy men were assessed for the leg extension one-repetition maximum (1RM) strength using bilateral and unilateral contractions. Following this assessment, an RE session (3 sets of repetitions to failure) was conducted with bilateral and unilateral (both limbs) contractions using a load of 50% 1RM. The TVLL was calculated by the product of the number of repetitions and the load lifted per repetition. RPE and blood lactate were measured before, during and after each set. Session RPE was measured 30 minutes after RE sessions. There was a significant difference in the bilateral (120.0±11.9) and unilateral (135.0±20.2 kg) 1RM strength (p < 0.05). The TVLL was similar between both RE sessions. Although the repetitions decreased with each successive set, the total number of repetitions completed in the bilateral protocol (48) was superior to the unilateral (40) protocol (p < 0.05). In both bouts, RPE increased with each subsequent set whilst blood lactate increased after set 1 and thereafter remained stable (p < 0.05). The RPE and lactate responses were not significantly different between both sessions. In conclusion, a bilateral deficit in leg extension strength was confirmed, but the TVLL was similar between both RE sessions when exercising to voluntary fatigue. This outcome could be attributed to the number of repetitions completed in the unilateral RE bout. The equal TVLL would also explain the similar perceptual and metabolic responses across each RE session. 相似文献

In greenhouse and field experiments, an invert emulsion (MSG 8.25) was tested with dried, formulated spores of the bioherbicidal fungus Colletotrichum gloeosporioides f. sp. aeschynomene, a highly virulent pathogen of the leguminous weed Aeschynomene virginica (northern jointvetch), but considered ‘immune’ against another more serious leguminous weed, Sesbania exaltata (hemp sesbania). A 1:1 (v/v) fungus/invert emulsion mixture resulted in 100% infection and mortality of inoculated hemp sesbania seedlings over a 21-day period under greenhouse conditions. In replicated field tests of the fungus/invert formulation conducted in Stuttgart, AR, and Stoneville, MS, hemp sesbania was controlled 85 and 90%, respectively. These results suggest that this invert emulsion expands the host range of C. gloeosporioides f. sp. aeschynomene, with a concomitant improvement of the bioherbicidal potential of this pathogen. 相似文献

Null mutations in DNA mismatch repair (MMR) genes elevate both base substitutions and insertions/deletions in simple sequence repeats. Data suggest that during replication of simple repeat sequences, polymerase slippage can generate single-strand loops on either the primer or template strand that are subsequently processed by the MMR machinery to prevent insertions and deletions, respectively. In the budding yeast Saccharomyces cerevisiae and mammalian cells, MMR appears to be more efficient at repairing mispairs comprised of loops on the template strand compared to loops on the primer strand. We identified two novel yeast pms1 alleles, pms1-G882E and pms1-H888R, which confer a strong defect in the repair of "primer strand" loops, while maintaining efficient repair of "template strand" loops. Furthermore, these alleles appear to affect equally the repair of 1-nucleotide primer strand loops during both leading- and lagging-strand replication. Interestingly, both pms1 mutants are proficient in the repair of 1-nucleotide loop mispairs in heteroduplex DNA generated during meiotic recombination. Our results suggest that the inherent inefficiency of primer strand loop repair is not simply a mismatch recognition problem but also involves Pms1 and other proteins that are presumed to function downstream of mismatch recognition, such as Mlh1. In addition, the findings reinforce the current view that during mutation avoidance, MMR is associated with the replication apparatus. 相似文献

Agricultural production is under increasing pressure by global anthropogenic changes, including rising population, diversion of cereals to biofuels, increased protein demands and climatic extremes. Because of the immediate and dynamic nature of these changes, adaptation measures are urgently needed to ensure both the stability and continued increase of the global food supply. Although potential adaption options often consider regional or sectoral variations of existing risk management (e.g. earlier planting dates, choice of crop), there may be a global-centric strategy for increasing productivity. In spite of the recognition that atmospheric carbon dioxide (CO(2)) is an essential plant resource that has increased globally by approximately 25 per cent since 1959, efforts to increase the biological conversion of atmospheric CO(2) to stimulate seed yield through crop selection is not generally recognized as an effective adaptation measure. In this review, we challenge that viewpoint through an assessment of existing studies on CO(2) and intraspecific variability to illustrate the potential biological basis for differential plant response among crop lines and demonstrate that while technical hurdles remain, active selection and breeding for CO(2) responsiveness among cereal varieties may provide one of the simplest and direct strategies for increasing global yields and maintaining food security with anthropogenic change. 相似文献

目的:研究三氧化二砷(As2O3)对食管癌细胞增殖和热休克蛋白70(HSP70)表达的影响。方法:通过相差显微镜、流式细胞术、免疫细胞化学染色和免疫印迹分析等方法观察As2O3对人食管癌细胞株EC1的作用效果和作用机制。结果:与对照组相比,经2μmol/L和5μmol/Las2O3作用的细胞出现明显的生长抑制,G2/M期细胞比例增加;2μmol/Las2O3作用48h后经Ecl细胞HSP70(heat shock protein70)及HSC70(heat shock cognate protein70)表达均增加。结论:As2O3诱导食管癌细胞G2/M期阻滞抑制细胞增殖和生长;HSP70的升高是细胞对As2O3作用后出现的应激反应,并与细胞周期阻滞相关。 相似文献
