The maize b-32 protein is a functional ribosome-inactivating protein (RIP), inhibiting in vitro translation in the cell-free reticulocyte-derived system and having specific N-glycosidase activity on 28S rRNA. Previous results indicated that opaque-2 (o2) mutant kernels, lacking b-32, show an increased susceptibility to fungal attack and insect feeding and that ectopic expression in plants of a barley and a pokeweed RIP leads to increased tolerance to fungal and viral infection. This prompted us to test whether b-32 might functi on as a protectant against pathogens. The b32.66 cDNA clone under the control of the potato wun1 gene promoter was introduced into tobacco by Agrobacterium tumefaciens-mediated transformation. Out of 23 kanamycin resistant regenerated shoots, 16 contained a PCR fragment of the corrrect size spanning the boundary between the promoter used and the coding region of the b-32 gene. Eight independently transformed tobacco lines were randomly chosen for protein analysis: all of them expressed b-32 protein. The data presented indicate that transgenic tobacco plants expressing b-32 show an increased tolerance against infection by the soil-borne fungal pathogen Rhizoctonia solani Kuhn  相似文献

Volcanic eruptions impact the global and the hemispheric climate, but it is still unknown how and to what degree they force the climate system and in particular the global carbon cycle. In this paper, the relationships between individual eruptions (reconstructed for the past using written records), tree primary productivity (estimated using ring widths), photosynthetic rate and stomatal conductance (assessed by carbon and oxygen isotope data) are investigated, to understand the impact of volcanic eruptions on net primary production. Data from a mixed stand of Fagus sylvatica L. and Acer pseudoplatanus L. located in the area of the Vesuvio volcanic complex (Southern Italy) showed a significant decrease in ring width following each eruption. Isotope analyses indicate a change in climatic conditions after such events. Specifically, the lower oxygen isotope ratio in the tree‐ring cellulose strongly suggests an increase in relative humidity and a decrease in temperature, with the latter resulting in a strong limitation to tree‐ring growth. The carbon isotope ratio was only moderately but not significantly reduced in the years of volcanic eruption, suggesting no major changes in C fixation rates. This work is a case study on the effects of volcanic eruptions resulting in strong climatic changes on the local scale. This is an opportunity to explore the process and causal relationships between climatic changes and the response of the vegetation. Thus, we propose here a realistic model scenario, from which we can extrapolate to global scales and improve our interpretations of results of global studies.  相似文献

ABSTRACT Rose-ringed parakeets (Psittacula krameri) have become widely established outside their native range through accidental or deliberate release. Potential economic impacts on agriculture, conservation concerns, and mixed public opinion regarding the species have highlighted the need to develop effective but humane management options. Fertility control might provide such a solution if a safe and environmentally benign contraceptive was available. The chemical 20,25-diazacholesterol dihydrochloride (diazacon) has previously been used to reduce reproductive output in avian species through reduction of blood cholesterol and cholesterol-dependent reproductive hormones. We orally dosed captive rose-ringed parakeets with a solution of either 9 mg/kg or 18 mg/kg of diazacon for up to 10 days and found that a dose of 18 mg/kg for 10 days temporarily reduced blood cholesterol levels with no adverse side effects. We evaluated this dose level in a captive population in semi-natural conditions during the 2008 breeding season and found a significant decrease in fertility. We concluded that diazacon has potential for fertility control in this species if a suitable formulation and delivery system is developed for free-living populations.  相似文献

ABSTRACT Bait-delivered pharmaceuticals, increasingly used to manage populations of wild boar (Sus scrofa) and feral pigs, may be ingested by nontarget species. Species-specificity could be achieved through a delivery system. We designed the BOSTM (Boar-Operated-System) as a device to deliver baits to wild pigs. The BOSTM consists of a metal pole onto which a round perforated base is attached. A metal cone with a wide rim slides up and down the pole and fully encloses the base onto which the baits are placed. We conducted a pilot, captive trial and found that captive wild boar fed from the BOSTM either directly, by lifting the cone, or indirectly, by feeding once another animal had lifted the cone. Thus, we tested whether free-living wild boar fed from the BOSTM and whether the BOSTM could prevent bait uptake by nontarget species. We observed that free-living wild boar fed regularly from the BOSTM and that the device successfully prevented bait uptake by nontarget species. The BOSTM should be trialed more extensively to confirm its effectiveness and species-specificity to distribute pharmaceuticals to wild suids. If successful, the BOSTM could be used to deliver vaccines in disease control programs as well as contraceptives to manage overabundant populations of wild suids.  相似文献

Mediterranean shrub species are described as having phenology, habitus , reproductive biology and anatomical alterations in certain tissues, allowing their survival during the dry season and protecting them from herbivory. Anatomical and chemical analyses were conducted in 1-year-old branches of Cistus ladanifer L. in order to investigate the role played by shoot structure in the adaptive strategies of this species in the Mediterranean environment. Results showed that both xylem and pith underwent lignification. Pith parenchyma cells had thickened walls, higher lignin content than xylem and different monomer composition. Xylem presented features aiding safe water transport. A large accumulation of phenolic substances was found in xylem, pith and cortical parenchyma. Observations reported in this paper suggest the occurrence of adaptive strategies in 1-year-old branches of C. ladanifer whose structural features: (1) allow mechanical reinforcement of tissues to withstand drought without suffering permanent damage; (2) favour safety rather than efficiency in water transport; (3) defend the plants from animal predation and pathogens by accumulating phenolics in various tissues, and (4) protect inner tissues against UV-B radiation through deposition of phenolic compounds in cortical layers.  © 2007 The Linnean Society of London, Botanical Journal of the Linnean Society, 2007, 155 , 361–371.  相似文献

Seed germination and seedling development were studied in Myrtus communis L. (Myrtaceae), an evergreen shrub widespread in Mediterranean woodlands, maquis and garrigues, to investigate the presence of morphological and anatomical adaptations that enhance successful seedling establishment under biotic and abiotic stresses. Seeds were germinated under controlled conditions and were observed daily to identify different post-germination stages of development. The anatomy of developing hypocotyls and emerging radicles was studied through light and epi-fluorescence microscopy using both cross and longitudinal sections. Tegument rupture was caused by hypocotyl elongation, which, when water availability in the substrate was limited, was accompanied by a dense ring of hairs in the lower region. Absorption tests showed that the hypocotyl hairs were functional in water uptake. They might also enhance the anchorage of the seedling to the substrate. Further investigations showed the early formation of a suberized layer of cells (exodermis) in the radicle and the occurrence of phenolics in the hypocotyl cells of cortical parenchyma. In conclusion, our results show that seedlings of M. communis have developed a strategy of establishment that: (a) regulates water uptake through hypocotyl hairs and the exodermis; (b) might allow mechanical support through hypocotyl hairs; and (c) accumulates phenolics that might defend the plant against animal predation.  © 2004 The Linnean Society of London, Botanical Journal of the Linnean Society , 2004, 145 , 195–202.  相似文献

Blue mould [ Peronospora hyoscyami f. sp. tabacina (Adam) Skalicky 1964] is one of the most important foliar diseases of tobacco that causes significant losses in the Americas, south-eastern Europe and the Middle East. This review summarizes the current knowledge of the mechanisms employed by this oomycete pathogen to colonize its host, with emphasis on molecular aspects of pathogenicity. In addition, key biochemical and molecular mechanisms involved in tobacco resistance to blue mould are discussed.

Disease symptoms: The pathogen typically causes localized lesions on tobacco leaves that appear as single, or groups of, yellow spots that often coalesce to form light-brown necrotic areas. Some of the leaves exhibit grey to bluish downy mould on their lower surfaces. Diseased leaves can become twisted, such that the lower surfaces turn upwards. In such cases, the bluish colour of the diseased plants becomes quite conspicuous, especially under moist conditions when sporulation is abundant. Hence the name of the disease: tobacco blue mould.
Infection process: The pathogen develops haustoria within plant cells that are thought to establish the transfer of nutrients from the host cell, and may also act in the delivery of effector proteins during infection.
Resistance: Several defence responses have been reported to occur in the Nicotiana tabacum – P. hyoscyami f. sp. tabacina interaction. These include the induction of pathogenesis-related genes, and a correlated increase in the activities of typical pathogenesis-related proteins, such as peroxidases, chitinases, β-1,3-glucanases and lipoxygenases. Systemic acquired resistance is one of the best characterized tobacco defence responses activated on pathogen infection.  相似文献

The reproductive biology of Ruscus aculeatus L. was studied at a mediterranean site during 1996–1999. Field observations showed that reproductive success was very low, because only 3% of the marked flowers developed fruits in 1996 and none in 1997 and 1998. Several experiments were performed to investigate the reasons for reproductive failure in this species. Pollen viability was on average 85%, showing that reproductive failure is not due to poor pollen quality. Two hand-pollination experiments showed that all flowers are potentially able to develop fruits throughout the flowering period, suggesting that no aberration occurs in sporogencsis, fertilization or post-zygote processes in R. aculeatus and that low reproductive success in the field occurs because of lack of pollination. This hypothesis was tested by looking for pollen on the stigmatic surface of randomly sampled flowers, but without success. Moreover, the bodies of insects trapped in the proximity of the female plants were carefully checked for pollen but none was found. The number of pollen grains produced per flower is lower than that reported for wind-pollinated species. Nevertheless, we showed with empirical probes that the possibility of wind pollination cannot be ruled out. The breeding system and reproductive characteristics of R. aculeatus seem to match the traits of the sclerophyllous taxa remnants of the tropical Tertiary flora. Moreover, the climatic fluctuation of the Quaternary and the establishment of the mediterranean climate might have changed the conditions of a good pollination process. At present, the low sexual reproduction of R. aculeatus is due to the lack of pollen transport. Within a population of R. aculeatus , genetic variability is revealed by seeds from the few developed fruits which have been shown to be viable. However, maintenance of the individual plants in the population relies on the resprouting capacity of the species in question.  相似文献
