The changes in germination, peroxidase activity and isoperoxidase spectrum have been studied in apple embryos at 5°C (stratification) and at 20°C in the presence or absence of seed coats. The embryo dormancy is progressively released at 5°C, but not at 20°C. The peroxidase activity in embryos covered with seed coats is very low at 5°C as well as at 20°C which corresponds to a restricted number of isoenzymes. In isolated embryos the peroxidase activity increases significantly. This is due to an increase in both the number and the activity of the isoperoxidases and it is more pronounced at 20°C than at 5°C. The obtained results suggest that the soluble peroxidases are not involved in the process of the release of embryo dormancy. The variations observed are attributed to the growth process following germination, which can occur even at low temperature.  相似文献

The content and pattern of soluble isoperoxidases were determined in epidermal explants taken from different internodes of tobacco plants in the vegetative and floral states. There were qualitative and quantitative differences in the isoperoxidases, with a decrease in content and fewer bands being observed acropetally, i.e., in going from the base of the stem towards the apex. Epidermal explants from floral branches were grown in in vitro culture, with various media moditications, to form de novo floral or vegetative buds, roots or callus. Changes in soluble isoperoxidases were followed electrophoretically in relation to these varying morphogenetic pathways. In each of them, the number of bands increased on both the anodic and cathodic sides with time in culture. Compared to each other these four morphogenetic programmes were different in their peroxidase zymograms, mainly through varying kinetics in the development of activity of the isoenzymes. The changes observed during root and vegetative bud formation agree with previously published data, and the changes during floral bud formation agree with those observed in vivo.  相似文献

Abstract The abscission of citrus leaf explants demonstrates the well-known enhancing effect of ethylene and the delaying one of auxin when treatment is started at excision time. Total peroxidase activity increases differently in tissues of the blade, abscission zone, and petiole. The highest activity at zero time is recovered in abscission zone in which also the response to the abscission regulators is the most visible. Isoperoxidase profiles are modified in opposite directions by ethylene and auxin respectively. Both regulators affect the activity of the same cathodic and anodic isoperoxidases without any qualitative changes. By the same time, auxin-like compounds increase in isolated abscission zones at 24 h from excision and decrease at 48 h. The level of one inhibitor complex undergoes an inverse variation. It is suggested that the increase in auxin during the first stage of abscission is necessary for influencing the growth of cells which is required to cause abscission.  相似文献

The effects of acetylcholine (Ach) on growth, the total peroxidase activity and the isoperoxidase spectrum of the roots ofLens culinaris were studied and compared with actual and earlier results obtained with an auxin (IAA) treatment. The purple dye murex, Bolinus brandaris (Linnaeus, 1758), is a muricid gastropod common throughout the Mediterranean and along the Moroccan and Portuguese Atlantic coasts. In the present study, we confirmed the diploid chromosome number of 2 n  = 70 for this species, and established for the first time the karyotype, which comprised 12 metacentric, 15 submetacentric and eight subtelocentric chromosome pairs. To facilitate cytotaxonomic comparisons, we carried out a comparative karyological analysis through multidimensional scaling between B. brandaris and three other 2 n  = 70 muricid species ( Hexaplex trunculus , Ocenebra erinaceus , and Stramonita haemastoma ) for which chromosomal measurements have been previously published. The interpretation of the ideograms and the statistical analysis highlighted the closest similarity of B. brandaris and H. trunculus compared to S. haemastoma and O. erinaceus . Indeed, B. brandaris and H. trunculus showed the smallest dissimilarities both for relative length and arm ratio, with O. erinaceus presenting intermediate values, whereas the highest dissimilarities were found between H. trunculus and S. haemastoma for both data. The karyotypes of B. brandaris and H. trunculus (subfamily Muricinae) presented the highest proportions of metacentric chromosomes compared to the other two muricids analysed, suggesting that those karyotypes could be considered primitive within the 2 n  = 70 Muricidae studied so far.  © 2009 The Linnean Society of London, Biological Journal of the Linnean Society , 2009, 96 , 185–193.  相似文献

From 2 per cent to 5 per cent of all indirect inguinal hernias are of the sliding variety. (Sliding hernias are those in which part of the wall of the sac is formed by a viscus.) The proportion of sliding hernias is even higher in the aged. Hernias of this kind are found almost exclusively in males and usually on the left side. Preoperative diagnosis is not essential if the surgeon can recognize the lesion at operation and knows how to repair it properly. The LaRoque technique in which the peritoneal cavity is entered above the internal ring allows accurate definition of the pathological anatomy and effective repair of the hernia. It should be used in all true sliding indirect inguinal hernias.  相似文献
