【背景】色氨酸脱羧酶在自然界有高度特异性,行使催化色氨酸为色胺的功能。一个色氨酸脱羧酶BaTDC参与南海海绵共生菌Bacillus atrophaeus C89次级代谢产物bacillamides的生物合成过程。【目的】探究BaTDC酶学特征和底物谱,建立体外合成色胺衍生物的方法。【方法】通过构建系统发育树揭示BaTDC在进化中的地位。在温度梯度和pH梯度下进行酶反应,利用不同的色氨酸衍生物为底物,通过HPLC和UPLC-MS检测酶反应过程,表征BaTDC活性。【结果】系统发育分析显示BaTDC与肠道菌Ruminococcus gnavus亲缘关系相近。纯化重组BaTDC的最适温度为40?45 °C,最适pH值为8.0。BaTDC可以催化羟代色氨酸和卤代色氨酸包括4-氟色氨酸和5,6,7-氯色氨酸及4-溴色氨酸,得到相应的卤代色胺衍生物和血清素。【结论】本研究分析了BaTDC的特性,发现BaTDC表现出宽泛的底物耐受性,可为前体喂养或定向生物合成新型药用色胺衍生物和下游复杂天然产物奠定基础。 相似文献

Mitochondrial F(1)F(0)-ATPase normally synthesizes ATP in the heart, but under ischemic conditions this enzyme paradoxically causes ATP hydrolysis. Nonselective inhibitors of this enzyme (aurovertin, oligomycin) inhibit ATP synthesis in normal tissue but also inhibit ATP hydrolysis in ischemic myocardium. We characterized the profile of aurovertin and oligomycin in ischemic and nonischemic rat myocardium and compared this with the profile of BMS-199264, which only inhibits F(1)F(0)-ATP hydrolase activity. In isolated rat hearts, aurovertin (1-10 microM) and oligomycin (10 microM), at concentrations inhibiting ATPase activity, reduced ATP concentration and contractile function in the nonischemic heart but significantly reduced the rate of ATP depletion during ischemia. They also inhibited recovery of reperfusion ATP and contractile function, consistent with nonselective F(1)F(0)-ATPase inhibitory activity, which suggests that upon reperfusion, the hydrolase activity switches back to ATP synthesis. BMS-199264 inhibits F(1)F(0) hydrolase activity in submitochondrial particles with no effect on ATP synthase activity. BMS-199264 (1-10 microM) conserved ATP in rat hearts during ischemia while having no effect on preischemic contractile function or ATP concentration. Reperfusion ATP levels were replenished faster and necrosis was reduced by BMS-199264. ATP hydrolase activity ex vivo was selectively inhibited by BMS-199264. Therefore, excessive ATP hydrolysis by F(1)F(0)-ATPase contributes to the decline in cardiac energy reserve during ischemia and selective inhibition of ATP hydrolase activity can protect ischemic myocardium. 相似文献

【背景】磷酸泛酰巯基乙胺基转移酶(PPTase)催化非核糖体肽合成酶(NRPS)中肽酰载体蛋白(PCP)从无活性的脱辅基形态转化为有活性的全辅基形态,从而启动非核糖体肽类化合物的生物合成。【目的】鉴定贪婪倔海绵共生萎缩芽孢杆菌C89中Sfp型PPTase Bap,验证Bap激活NRPS中PCP的能力。【方法】通过BLAST和氨基酸多序列比对鉴定萎缩芽孢杆菌C89中Sfp型PPTase Bap。将bap基因在sfp基因突变株枯草芽孢杆菌168中异源表达,通过重组菌枯草芽孢杆菌168-bap的代谢物检测非核糖体肽类化合物Surfactin。【结果】Bap为Sfp型PPTase,检测到重组菌枯草芽孢杆菌168-bap中Surfactin的产生。【结论】本研究为海洋萎缩芽孢杆菌中NRPS基因簇的异源表达奠定了基础。 相似文献

Both cucumber mosaic virus (CMV) and tobacco mosaic virus (TMV) coat protein (CP) genes have been transferred to chilli pepper (Capsicum annuum var. Longunt) cultivar 8212 by a modified procedure of Agrobacterium tumefaciens-mediated transformation using hypocotyl as the explant. PCR analysis revealed the presence of both CMV and TMV CP genes in at least 11 primary transformants out of 49 kanamycin-resistant chili pepper plants. Ten T1 lines from five independent transformation events were identified as putative homozygous transgenic lines based on the rooting assay of their T2 seedlings on the kanamycin-containing media. Integration and expression of CMV CP and TMV CP transgenes in one of the homozygous line, 16-13, were confirmed bySouthern blot, RT-PCR and western blot analyses. Line 16-13 was highly resistant to infection of homologous CMV and TMV strains in greenhouse conditions when successively challenged with CMV and TMV or challenged with TMV alone.Futhermore, field trials on T2, T3 and T4 progenies of Line 16-13 were performed on scales of 123, 300 and 10,000 plants, respectively, in consecutive years 1996, 1997 and 1998 with the permission of the Chinese government authority. The transgenic plants displayed delayed symptom development and significantly milder disease severity in field conditions when compared to untransformed chili pepper plants, resulting in 47 and 110% increase in pepper fruit yield in surveys conducted in 1997 and 1998 trials, respectively. Finally, quality analysis and biosafety assesment were performed on transgenic chili pepper fruit concurrently with the control fruit, and demonstrated that the transgenic chili pepper fruit is substantially equivalent to the non-transgenic pepper in terms of the quality and biosafety when consumed as a food additive. 相似文献

Highly ordered Ni nanowire arrays (NiNWAs) were synthesized for the first time using a template-directed electropolymerization strategy with a nanopore polycarbonate (PC) membrane template, and their morphological characterization were examined by scanning electron microscopy (SEM) and transmission electron microscope (TEM). A NiNWAs based electrode shows very high electrochemical activity for electrocatalytic oxidation of glucose in alkaline medium, which has been utilized as the basis of the fabrication of a nonenzymatic biosensor for electrochemical detection of glucose. The biosensor can be applied to the quantification of glucose with a linear range covering from 5.0 × 10−7 to 7.0 × 10−3 M, a high sensitivity of 1043 μA mM−1 cm−2, and a low detection limit of 1 × 10−7 M. The experiment results also showed that the sensor exhibits good reproducibility and long-term stability, as well as high selectivity with no interference from other oxidable species. 相似文献

采用抗菌谱测定以及BOXAIR-PCR、生理生化特征和16S rDNA序列分析等方法对分离自10种作物根际的55株拮抗细菌的多样性及主要拮抗菌类群进行了分析.结果表明: 根际拮抗细菌的拮抗作用具有丰富的多样性;BOXAIR-PCR分析中供试菌在721%相似性水平上可以聚为7个群,850%相似性水平上聚为25个群;所有供试根际拮抗菌分别属于芽孢杆菌属(Bacillus)芽孢杆菌属(Paenibacillus)、短芽孢杆菌属(Brevibacillus)、假单胞菌属(Pseudomonas)和产碱菌属(Alcaligenes).作物根际拮抗菌具有丰富的遗传多样性和拮抗性能多样性. 相似文献

矮牵牛花同源异型基因fbp2的克隆及其对烟草花形态的影响 总被引:6,自引:0,他引:6
以矮牵牛(Petunia hybrida L.)栽培品种为材料,取开放前的花蕾分离mRNA,反转录合成cDNA,以cDNA为模板,通过PCR扩增,对获得的目的片段进行序列分析。结果表明,分离的目的片段含有686个核苷酸(含有起始密码和终止密码)。核苷酸序列与文献报道相比,同源率为99.6%,只有3个碱基发生改变,5’端的MADS盒区域完全相同。将得到的矮牵牛花同源异型基因fbp2的cDNA(yfbp2)与CaMV355启动子和NOS3’终止子融合,构建了表达载体pBBP2。表达载体通过农杆菌(Agrobacterium tumefaciens)LBA4404(pAL4404)介导转化烟草(NicotianatabacumL.)叶片,在含有100mg/L卡那霉素的抗性培养基上再生成株。对抗性株进行总DNASouthern杂交和总RNA的点杂交,证明目的基因已导入烟草细胞中,整合到烟草基因组上,并且在烟草细胞中转录。同源异型基因fbp2导入烟草后导致烟草花型改变,在雄蕊上产生了花瓣。 相似文献
