Agricultural intensification and land-use changes are major factors impacting farmland biodiversity. The Ortolan Bunting Emberiza hortulana is the long-distance trans-Saharan migratory passerine that has undergone the most dramatic decline among all European farmland birds. The factors responsible for this decline may originate from the breeding grounds, migration stopovers and/or overwintering quarters. Very little is known about conditions on the species' wintering grounds, but a recent study has highlighted the utmost importance of the traditionally managed agroecosystems in the Ethiopian Highlands as a key wintering area, apparently harbouring as much as 90% of the world's Ortolan Bunting population. Using radiotracking and line transect surveys, this study aimed to provide fine-grained information about species–habitat relationships in the Ortolan Bunting overwintering quarters. Our results showed the importance, at the landscape scale, of small-scale agriculture, notably of traditionally managed, cereal-dominated fields interspersed with semi-natural structures. At a foraging-site scale, on the other hand, patches of bare ground in combination with large areas of post-harvesting stubble represented key habitat features. Stubbles provide an essential food resource and bare ground promotes ground foraging by enhancing food accessibility. The maintenance of a traditional agricultural economy will be essential to maintain the habitat potential for the Ortolan Buntings overwintering in the Ethiopian Highlands and will be instrumental in preserving its world population from further decline.  相似文献

BACKGROUND: beta-Lactam compounds are the most widely used antibiotics. They inactivate bacterial DD-transpeptidases, also called penicillin-binding proteins (PBPs), involved in cell-wall biosynthesis. The most common bacterial resistance mechanism against beta-lactam compounds is the synthesis of beta-lactamases that hydrolyse beta-lactam rings. These enzymes are believed to have evolved from cell-wall DD-peptidases. Understanding the biochemical and mechanistic features of the beta-lactam targets is crucial because of the increasing number of resistant bacteria. DAP is a D-aminopeptidase produced by Ochrobactrum anthropi. It is inhibited by various beta-lactam compounds and shares approximately 25% sequence identity with the R61 DD-carboxypeptidase and the class C beta-lactamases. RESULTS: The crystal structure of DAP has been determined to 1.9 A resolution using the multiple isomorphous replacement (MIR) method. The enzyme folds into three domains, A, B and C. Domain A, which contains conserved catalytic residues, has the classical fold of serine beta-lactamases, whereas domains B and C are both antiparallel eight-stranded beta barrels. A loop of domain C protrudes into the substrate-binding site of the enzyme. CONCLUSIONS: Comparison of the biochemical properties and the structure of DAP with PBPs and serine beta-lactamases shows that although the catalytic site of the enzyme is very similar to that of beta-lactamases, its substrate and inhibitor specificity rests on residues of domain C. DAP is a new member of the family of penicillin-recognizing proteins (PRPs) and, at the present time, its enzymatic specificity is clearly unique.  相似文献

This study was undertaken to determine the current population size, structure and habitat preference of Soemmerring's gazelle [Nanger soemmerringii (Cretzschmar 1828)] in the Alledeghi Wildlife Reserve, NE Ethiopia. Animals were counted, both during dry and wet seasons, along 12 line transects each in three habitat types (grassland, tree‐scattered grassland and bushland) in 2015/16. Habitat type had nonsignificant effect on mean population density of Soemmerring's gazelle, but wet season mean density was significantly higher than dry season mean density. Estimated weighted mean (±95% CI) population density of the species in the reserve was 1.90 (±0.17) and 5.99 (±0.370) individuals/km2 during the dry and wet seasons, respectively. Total population size of the species in the Alledeghi Wildlife Reserve was estimated at 826 ± 77 and 2,562 ± 158 individuals during the dry and wet seasons, respectively. Over half of the total population of Soemmerring's gazelle was represented by adult females during both seasons. Seasonal habitat preference of Soemmerring's gazelle was statistically significant, with greater preference for grassland habitat during wet season and for bushland habitat during dry season. In conclusion, this study has provided valuable data that will be used as a baseline for future population monitoring.  相似文献

In the present study, the elemental compositions of fat and nerve tissue during their plasma mediated laser ablation are studied in the context of tissue differentiation for laser surgery applications by using Laser‐Induced Breakdown Spectroscopy (LIBS). Tissue samples of porcine fat and nerve were prepared as ex vivo experimental objects. Plasma mediated laser ablation is performed using an Nd : YAG laser in open air and under normal stray light conditions. The performed measurements suggest that the two tissue types show a high similarity in terms of qualitative elemental composition while at the same time revealing a distinct difference in the concentration of the constituent elements. Different analysis approaches are evaluated and discussed to optimize the tissue‐differentiation performance of the LIBS approach.

Genetic data from extant donkeys (Equus asinus) have revealed two distinct mitochondrial DNA haplogroups, suggestive of two separate domestication events in northeast Africa about 5000 years ago. Without distinct phylogeographic structure in domestic donkey haplogroups and with little information on the genetic makeup of the ancestral African wild ass, however, it has been difficult to identify wild ancestors and geographical origins for the domestic mitochondrial clades. Our analysis of ancient archaeological and historic museum samples provides the first genetic information on the historic Nubian wild ass (Equus africanus africanus), Somali wild ass (Equus africanus somaliensis) and ancient donkey. The results demonstrate that the Nubian wild ass was an ancestor of the first donkey haplogroup. In contrast, the Somali wild ass has considerable mitochondrial divergence from the Nubian wild ass and domestic donkeys. These findings resolve the long-standing issue of the role of the Nubian wild ass in the domestication of the donkey, but raise new questions regarding the second ancestor for the donkey. Our results illustrate the complexity of animal domestication, and have conservation implications for critically endangered Nubian and Somali wild ass.  相似文献

Photoacoustic endoscopy (PAE) is an emerging imaging modality, which offers a high imaging penetration and a high optical contrast in soft tissue. Most of the developed endoscopic photoacoustic sensing systems use miniaturized contact ultrasound transducers or complex optical approaches. In this work, a new fiber‐based detection technique using speckle analysis for contact‐free signal detection is presented. Phantom and ex vivo experiments are performed in transmission and reflection mode for proof of concept. In summary, the potential of the technique for endoscopic photoacoustic signal detection is demonstrated. The new technique might help in future to broaden the applications of PAE in imaging or guiding minimally invasive laser procedures.   相似文献

Light has direct effects on sleep and wakefulness causing arousal in diurnal animals and sleep in nocturnal animals. In the present study, we assessed the modulation of light-induced sleep by melanopsin and the histaminergic system by exposing mice to millisecond light flashes and continuous light respectively. First, we show that the induction of sleep by millisecond light flashes is dose dependent as a function of light flash number. We found that exposure to 60 flashes of light occurring once every 60 seconds for 1-h (120-ms of total light over an hour) induced a similar amount of sleep as a continuous bright light pulse. Secondly, the induction of sleep by millisecond light flashes was attenuated in the absence of melanopsin when animals were presented with flashes occurring every 60 seconds over a 3-h period beginning at ZT13. Lastly, the acute administration of a histamine H3 autoreceptor antagonist, ciproxifan, blocked the induction of sleep by a 1-h continuous light pulse during the dark period. Ciproxifan caused a decrease in NREMS delta power and an increase in theta activity during both sleep and wake periods respectively. The data suggest that some form of temporal integration occurs in response to millisecond light flashes, and that this process requires melanopsin photoreception. Furthermore, the pharmacological data suggest that the increase of histaminergic neurotransmission is sufficient to attenuate the light-induced sleep response during the dark period.  相似文献

The research work presented in this paper focuses on qualitative tissue differentiation by monitoring the intensity ratios of atomic emissions using ‘Laser Induced Breakdown Spectroscopy’ (LIBS) on the plasma plume created during laser tissue ablation. The background of this study is to establish a real time feedback control mechanism for clinical laser surgery systems during the laser ablation process. Ex‐vivo domestic pig tissue samples (muscle, fat, nerve and skin) were used in this experiment. Atomic emission intensity ratios were analyzed to find a characteristic spectral line for each tissue. The results showed characteristic elemental emission intensity ratios for the respective tissues. The spectral lines and intensity ratios of these specific elements varied among the different tissue types.
