Iron saccharate complex ISC is an iron supplement used to optimize erythropoiesis in cases of iron deficiencies. Because of the lack of major mechanisms of iron excretion, excess iron unbound to protective molecules is believed to be involved in catalyzing the generation of reactive oxygen species and induction of oxidative stress. This study employed ISC for the purpose of inducing iron overload and hence investigating the consequent iron toxicity, lipid peroxidation and antioxidant extent in a murine species. Male Wistar rats were given iron as intraperitoneal injections of ISC in subacute (0.2 mg Fe kg?1 for 2 weeks) and subchronic (0.1 mg Fe kg?1 for 4 weeks) doses. In iron-overloaded rats, enhanced hepatic iron accumulation (P > 0.001) attended by increased serum concentrations of malondialdehyde (MDA) (P > 0.001) and activities of antioxidant enzymes (superoxide dismutase SOD, catalase CAT and glutathione peroxidase GPx) (P > 0.001) was pointed out. The demonstrated antioxidant boost is attributed to a sense of equilibrium prompted by the potential of iron-induced oxidative stress to modify antioxidant defense capacity and to modulate susceptibility to oxidative stress. Rats seemed to constantly suffer from oxidative stress based on the consistent rise in MDA that was not overwhelmed by the elevated antioxidant input. The current findings are of informative value in drawing attention to the health hazards of applying higher doses of the commercially used iron supplement ISC. Data are virtually significant in elucidating the higher magnitude of subchronic than subacute iron overload in initiating oxidative stress and antioxidant defense. Both pathways proceeded in a time-dependent rather than dose-dependent manner.  相似文献

Purpose

We extend a life cycle assessment (LCA) embracing both economic and social perspectives to develop an integrated solid waste management system for Kuwait. This study considers the domestic waste generated by households and waste generated commercially. Six municipal solid waste (MSW) scenarios (SR1, SR2, …, SR6) are evaluated using a triple bottom line (TBL) approach that incorporates environmental, financial, and social bottom lines (social BLs).

Methods

Within the TBL framework, the environmental BL employs LCA in accordance with ISO 14044. The financial BL is calculated using capital and operational costs as well as the associated recycling revenues. The social BL applies macro-economic indicators that examine the effects of a given MSW scenario (SR) on the inhabitants. To integrate the TBLs, we apply an analytic hierarchy process (AHP) because of its advantage of pairwise unit-free rescaling. The relative importance of each BL is determined by considering the political, legal, socio-cultural, and economic climates of the country. The relative weights are cross-multiplied with indicators from each BL to calculate a composite sustainability index (CSI) for the proposed MSW SR.

Results and discussion

The environmental BL (LCA) indicates that global warming, acidification, and human toxicity are the most adversely affected impact categories, considering the local conditions and waste composition. Environmentally, SR1 (landfilling) scored the worst in almost all impact categories and, thus, was labeled the worst-case scenario environmentally. SR6 (composting, recycling, and incineration) performed the best from an environmental perspective. Financially, landfilling (SR1) is the most economical scenario. Any SR that focused on incineration (SR2 and SR5) was financially unfavorable. The scenarios that involved composting were scored as financially reasonable (SR3, SR4, and SR6). From a social acceptability perspective, SR2 (incineration) scored the highest, while SR1 (landfills) scored the lowest. Finally, across the TBL framework, SR4 (composting and incineration) had the highest CSI based on the relative importance scheme adopted for each BL.

Conclusions

Although they are often overlooked in most LCA studies, the financial and social aspects are indispensable to proving feasibility and credibility at a strategic level. The complexity of financial and social formulations in LCA is inherited from the difficulty in quantifying emissions and other impacts. In addition, from a social perspective, the contingent risks and associated uncertainty vary widely across cultures, ideologies, and degrees of development and are further complicated because of the scarcity and uncertainty of the data.

Cadmium (Cd) is an environmental carcinogenic pollutant known to inactivate several proteins involved in DNA repair systems while at the same time creating an oxidative stress that can result in additional DNA lesions. The testis and the lung are the target organs for cadmium carcinogenesis. Increased production of oxidants in vivo can cause damage to intracellular macromolecules such as DNA, proteins and lipids, which in turn lead to oxidative injury. So, this investigation aimed to evaluate the protective role of L-Carnitine through up regulation of HSPs against DNA damage induced by cadmium chloride. The current study was carried out on forty adult male rats, each with average weight 220-250g., were divided into 4 equal groups. 1(st) group was received saline solution (0.5 ml/100 g body weight) and kept as control. 2(nd) group was received 500mg / kg body weight L-Carnitine intraperitoneally (IP). 3(rd) group was administered 1.2 mg cadmium chloride IP. 4(th) group was received both cadmium chloride and L-Carnitine simultaneously. The comet assay parameters showed significantly increased HSP70 and DNA damage in testis cells after 10 and 56 days in the third group. Meanwhile, HSP70 showed significantly decreased levels after 10 days and 56 days in the fourth group after L-Carnitine treatment simultaneously with cadmium chloride. The results of the present study demonstrate a damaging effect of cadmium chloride on DNA of the testis cells (with low stress response). This damaging effect increases the synthesis of HSP70 that upregulated by L-Carnitine treatment and showed ameliorative effect of the cells for recovery.  相似文献

Diabetes mellitus (DM) is a chronic disease that is characterized by deteriorating glycemic control. The disease is known to be caused by imbalance between reactive oxygen species (ROS) and antioxidant defense systems. Hyperglycemia is commonly observed in a wide variety of diseases, including cancer. Although, therapy against glycemic control, is used in all these diseases, the diabetic cancer patients are on additional therapy with anticancer drugs. The objective of present study was to study if Glucophage (metformin), a very popular antidiabetic agent can avert the mutagenicity and lipid peroxidation caused by adriamycin (ADR), which is a commonly used cytotoxic drug. The experimental protocol included oral treatment of mice with different doses (62.5, 125 and 250 mg/kg day) of metformin for 7 days. Some mice in each group were injected i.p. with ADR (15 mg/kg). In each case animals were killed, 30 or 24, 48 and 72 h after the last treatment and femurs were excised for cytological studies by micronucleus test. Additional experiments on estimation of glutathione (GSH) and malondialdehyde (MDA) were undertaken in blood and serum, respectively. Twenty-four hour after the treatment, blood from each mouse was collected from heart and preserved for analysis. The results obtained revealed that pretreatment with metformin: (i) reduced the ADR-induced frequency of micronuclei without any alteration in its cytotoxicity and (ii) protected against the ADR-induced increase and decrease of MDA and GSH, respectively. The exact mechanism of action is not known, however, the inhibition of ADR-induced clastogenicity and lipid peroxidation by metformin may be attributed to the antioxidant action of the latter. Our results demonstrate that metformin might be useful to avert secondary tumor risk by decreasing the accumulation of free radicals and inhibition of mutagenicity.  相似文献
