Antigenic similarities between Plasmodium and Babesia parasites of the phylum Apicomplexa have been previously demonstrated primarily by the serological cross reactivity observed in the indirect fluorescent antibody (IFA) test. We have now studied the antigenic relationship between the human malaria parasite, Plasmodium falciparum, and the hemoparasitic agent of cattle, Babesia bovis, using rabbit monospecific antibodies produced against individual culture-derived P. falciparum polypeptides and bovine polyspecific antibodies to B. bovis exoantigens. These respective antibodies were found to be distinctly cross reactive in the IFA test using infected erythrocytes (squirrel monkey—P. falciparum; bovine—B. bovis) as antigen substrates. Immunofluorescence was shown to be highly specific for parasite surfaces. Additionally, the degree of reactivity with soluble exoantigens contained in Plasmodium and Babesia culture supernatants was monitored by a two-site enzyme immunoassay employing the cross-reactive antibodies. Further evidence for antigenic cross reactivity between P. falciparum and B. bovis parasites was shown with the in vitro inhibition assay. Antibodies to P. falciparum and B. bovis were found to be highly inhibitory for the in vitro growth of P. falciparum in human erythrocytes.  相似文献

1. Predation‐exclusion experiments have highlighted that top‐down control is pervasive in terrestrial communities, but most of these experiments are simplistic in that they only excluded a single group of predators and the effect of removal was evaluated on a few species from the community. The main goal of our study was to experimentally establish the relative effects of ants and birds on the same arthropod assemblage of canopy trees. 2. We conducted 1‐year long manipulative experiments in an organic citrus grove intended to quantify the independent effects of bird and ant predators on the abundance of arthropods. Birds were excluded with plastic nets whereas ants were excluded with sticky barriers on the trunks. The sticky barrier also excluded other ground dwelling insects, like the European earwig Forficula auricularia L. 3. Both the exclusion of ants and birds affected the arthropod community of the citrus canopies, but the exclusion of ants was far more important than the exclusion of birds. Indeed, almost all groups of arthropods had higher abundance in ant‐excluded than in control trees, whereas only dermapterans were more abundant in bird‐excluded than in control trees. A more detailed analysis conducted on spiders also showed that the effect of ant exclusion was limited to a few families rather than being widespread over the entire diverse spectrum of spiders. 4. Our results suggest that the relative importance of vertebrate and invertebrate predators in regulating arthropod populations largely depends on the nature of the predator–prey system.  相似文献

The location of major quantitative trait loci (QTL) contributing to stem and leaf [Na+] and [K+] was previously reported in chromosome 7 using two connected populations of recombinant inbred lines (RILs) of tomato. HKT1;1 and HKT1;2, two tomato Na+‐selective class I‐HKT transporters, were found to be closely linked, where the maximum logarithm of odds (LOD) score for these QTLs located. When a chromosome 7 linkage map based on 278 single‐nucleotide polymorphisms (SNPs) was used, the maximum LOD score position was only 35 kb from HKT1;1 and HKT1;2. Their expression patterns and phenotypic effects were further investigated in two near‐isogenic lines (NILs): 157‐14 (double homozygote for the cheesmaniae alleles) and 157‐17 (double homozygote for the lycopersicum alleles). The expression pattern for the HKT1;1 and HKT1;2 alleles was complex, possibly because of differences in their promoter sequences. High salinity had very little effect on root dry and fresh weight and consequently on the plant dry weight of NIL 157‐14 in comparison with 157‐17. A significant difference between NILs was also found for [K+] and the [Na+]/[K+] ratio in leaf and stem but not for [Na+] arising a disagreement with the corresponding RIL population. Their association with leaf [Na+] and salt tolerance in tomato is also discussed.  相似文献

A comparative study of karyotype morphology and heterochromatin patterns in Brunnera and Cynoglottis (Boraginaceae) was carried out with traditional methods and Giemsa C-banding. Two polymorphic species of Cynoglottis , each with two subspecies, and two of Brunnera were investigated using native population samples from the central-eastern Mediterranean and Middle East. Pollen size of these samples was measured to investigate relationships with ploidy level. C. barrelieri subsp. barrelieri and subsp. serpentinicola are characterized by In = 18 and smaller pollen grains in contrast to C. chetikiana subsp. chetikiana and subsp. paphlagonica , which are fundamentally tetraploid with 2n = 36. The occurrence of cytotypes with 2n –/2 and 2n = 24 in both subspecies of C. chetikiana , however, would suggest x = 6 as the original haploid number and x = 9 as a derived one. Furthermore, the finding of a hypoploid cytotype with 2n = 16 in C. barrelieri ssp. barrelieri was consistent with previous reports and suggested relationships with Anchusa. Karyoevolutionary processes possibly associated with such a wide chromosome variation in Cynoglottis are discussed. Brunnera macrophylla and B. orientalis share a complement of 2n= 12 and an apparently identical karyotype, which differs from Cynoglottis in terms of asymmetry, chromosome size and morphology. A basic C-banding style was present in Brunnera and Cynoglottis , but heterochromatin content increased from the former to the latter. The parallel increase in chromosome number, heterochromatin content and size of the pollen from Brunnera to Cynoglottis may reflect an evolutionary progression, and is consistent with the supposed ancient origin of Brunnera.  相似文献

Chrysomela populi beetles feed on poplar leaves and extensively damage plantations. We investigated whether olfactory cues orientate landing and feeding. Young, unexpanded leaves of hybrid poplar emit constitutively a blend of monoterpenes, primarily ( E )- β -ocimene and linalool. This blend attracts inexperienced adults of C. populi that were not previously fed with poplar leaves. In mature leaves constitutively emitting isoprene, insect attack induces biosynthesis and emission of the same blend of monoterpenes, but in larger amount than in young leaves. The olfactometric test indicates that inexperienced beetles are more attracted by adult than by young attacked leaves, suggesting that attraction by induced monoterpenes is dose dependent. The blend does not attract adults that previously fed on poplar leaves. Insect-induced emission of monoterpenes peaks 4 d after the attack, and is also detected in non-attacked leaves. Induced monoterpene emission is associated in mature leaves with a larger decrease of isoprene emission. The reduction of isoprene emission is faster than photosynthesis reduction in attacked leaves, and also occurs in non-attacked leaves. Insect-induced monoterpenes are quickly and completely labelled by 13C. It is speculated that photosynthetic carbon preferentially allocated to constitutive isoprene in healthy leaves is in part diverted to induced monoterpenes after the insect attack.  相似文献
