The influence of ungulate grazers on nutrient cycling and ecosystem productivity in grasslands has been shown to differ with moisture, nutrient availability, and feedbacks between above- and belowground activities. We examined the movement of nitrogen (N), applied as (15NH4)2SO4, through both dry and mesic sites in the northern range of Yellowstone National Park to test the hypothesis that plants were more able to acquire added N in grazed relative to ungrazed sites. Previous studies showed enhanced N mineralization in grazed areas, and detritus removal by grazers was predicted to enhance early-season plant growth. Thirteen months after tracer addition, there were no differences in plant 15N as a function of grazing, but historically ungrazed sites retained more 15N in accumulated litter than at grazed sites. This result demonstrated the importance of detritus in regulating redistribution of incoming N and the role of grazers in this process. Site moisture status influenced 15N recovery in all pools—soils, microbial biomass, and plants—and greater plant 15N acquisition occurred in roots at dry relative to mesic sites. Understanding how grazers influence nutrient cycling at the landscape scale requires further investigation of interactions among soil moisture, plant production, litter accumulation, grazing intensity, and belowground processes.  相似文献

Radiation protection of Escherichia coli B/r by hydroxyl radical scavengers   总被引:1,自引:0,他引:1  
We have used Escherichia coli B/r to test the proposal that hydroxyl radicals (.OH) are major contributors to lethal damage when bacteria in equilibrium with air or 100% nitrogen are exposed to ionizing radiation. In addition, we have tested the hypothesis that oxygen sensitizes bacterial cells to radiation by reacting at radical sites previously formed by reactions of .OH. Our results with B/r indicate that the involvement of OH radicals in damage may have been overestimated. We believe that simple .OH removal provides B/r with only a relatively small amount of protection in N2 and air. Although some .OH scavengers can have large protective effects in air, evidence supports the tentative conclusion that these effects are not based on simple .OH removal. If this conclusion is correct, then radiation sensitization by oxygen--at least of this bacterial strain--would be unrelated to reactions of .OH.  相似文献

Previous work with procaryotic cells has identified one kind of lethal damage from ionizing radiation which occurs only within a specific range of low O2 concentrations, about 10(-6) to 10(-4) M. Within this range, protection can occur in three ways: through the enzymatic decomposition of hydrogen peroxide (H2O2) by added catalase, through the enzymatic degradation of superoxide anion radicals (.O2-) by added superoxide dismutase (SOD), and through scavenging hydroxyl radicals (.OH) by various additives. These results indicate that three radiolytic products, H2O2, .OH, and .O2- (and/or the conjugate acid, the perhydroxyl radical, .HO2) are involved in this single kind of radiation-induced damage. Although the radiolytic productions of H2O2 and .O2- are strongly enhanced in higher O2 concentrations, neither enzyme protects when these air-equilibrated bacteria are irradiated. These experiments address this apparent contradiction and focus on the specific issue of why the addition of SOD protects at low but not at high O2 concentrations. We propose that, at a given O2 concentration, .O2- (and/or .HO2) may either react (with some cellular component?) to cause damage or react (with itself) to form hydrogen peroxide (H2O2). The specific O2 concentration during irradiation would determine the relative rates of these competing reactions and therefore the O2 concentration itself would establish whether or not we will observe damage from .O2-.  相似文献

HydroLink materials represent a novel family of gels composed of unique polymer matrices. The applications of HydroLink to molecular biology and, specifically, to DNA technology have been carefully investigated. Our results indicate that the HydroLink matrix developed for double-stranded DNA (dsDNA) is an excellent tool for electrophoretic separations in fixed electric fields. Excellent linear resolution from 100 to 5000 base pairs is easily achieved with good resolution albeit non-linear from 6000 to 23000 base pairs. The broad range of separation in addition to increased mechanical strength of dsDNA HydroLink represents a distinct advantage over other matrices currently used in DNA electrophoretic analysis.  相似文献

In the selection of acid-tolerantRhizobium meliloti, procedures for the collection and isolation of rhizobia, and the assessment of acid tolerance, have not been critically evaluated. Such procedures form the basis of this study. Root nodules were collected fromMedicago spp. found growing on acid soil in Sardinia. Their encumbent bacteria were isolated directly onto media adjusted over a range of pH values, and then assessed for acid tolerance in both the laboratory and field. Strains ofRhizobium meliloti isolated onto low pH media were, in general, more acid-tolerant than sister isolates from high pH media, when tested in both the laboratory and field. Dilution (10 or 100 fold) of the inocula used in the laboratory assessment did not greatly influence the rating derived, although there was some effect of bacterial colony type on growth rating. The link between polysaccharide production and acid tolerance was not strong. There was a poor correlation between the growth ratings derived from the laboratory screening and acid tolerance as expressed in the field.  相似文献

In order to determine the effect of diabetic autonomic neuropathy (DAN) on the atrial natriuretic peptide (ANP) response to dynamic stimuli, we studied the ANP response to 60 degrees head-up and 60 degrees leg-up tilt in diabetic subjects with (DAN + ve, n = 8) and without (DAN - ve, n = 8) evidence of autonomic neuropathy and seven matched non-diabetic controls. Mean baseline plasma ANP concentrations were similar in all three groups. Head-up tilt was associated with a fall in plasma ANP in all seven healthy controls (21.8 (16.8-30.7) to 16.8 (7.1-29.1), P = 0.06, mean (range)), seven of the eight DAN - ve (16.9 (6.5-33.7) to 8.5 (3.0-21.1), P = 0.015) and all eight DAN + ve subjects (27.3 (8.5-101.5) to 15.4 (1.0-67.6), P = 0.044). Leg-up tilt caused a rise in plasma ANP in six of the seven healthy controls (17.6 (7.5-27.9) to 22.4 (15.2-48.1), P = 0.041), six of the eight DAN - ve (12.5 (7.8-27.8) to 15.5 (7.3-31.3), P = 0.054) and seven of the eight DAN + ve subjects (18.2 (2.8-55.1) to 25.1 (4.5-92.8), P = 0.013). There was no significant difference in the fall in plasma ANP during head-up tilt or in the rise in plasma ANP during leg-up tilt between the three groups. We conclude that the regulation of ANP secretion is normal in diabetes mellitus, and is unaffected by the presence of autonomic neuropathy.  相似文献

Methods were devised to purify the cell-associated, iron-binding compounds known as mycobactins from the closely related species Mycobacterium avium, Mycobacterium intracellulare, and Mycobacterium scrofulaceum (i.e., the MAIS complex of organisms). The mycobactins from these three species showed a structure that is common to the mycobactins from all the mycobacteria examined to date. However, these mycobactins were unique in that they had more than one alkyl chain. The M. scrofulaceum mycobactins differed from other MAIS mycobactins by a shift in the position of the double bond in the R1 alkyl chain. Traces of other mycobactin types were observed in ethanol extracts of the three species, and examination of the chromatographic properties of these mycobactins showed that each species produced five mycobactin types. Each mycobactin could be subdivided further by the length of its R1 alkyl chain. No differences in the production of these novel mycobactin were observed among species. Mycobactins from three strains of Mycobacterium paratuberculosis and two wood pigeon strains of Mycobacterium avium which had lost their original growth requirements for mycobactin after repeated subculturing in laboratory growth media were examined by thin-layer chromatography and high-pressure liquid chromatography. Each organism produced a mycobactin with similar chromatographic properties to those synthesized by MAIS organisms. M. paratuberculosis NADC 18 produced at least two components in our laboratory, and nuclear magnetic resonance analysis of the major component showed this mycobactin to be identical to that produced by M. intracellulare M12. However, a sample of mycobactin J isolated by Merkal and McCullough (Curr. Microbiol. 7:333-335, 1982) from M. paratuberculosis NADC 18 was different from our isolates and appeared to correspond to a minor mycobactin component we had seen by thin-layer chromatography. No reason for this difference could be evinced. Our findings indicate that there is a close taxonomic relationship between M. paratuberculosis and the MAIS complex.  相似文献

The anoxic sensitization of bacterial spores by added H2O2 has been studied. Two mechanistic pathways for damage from H2O2 were found; one of these requires the presence of OH radicals. For this kind of damage, the relationship between H2O2 and OH appears to be that they are reactants. O-2 (and/or HO2), the product of such a reaction, is likely the agent which actually causes damage. These results with reagent H2O2 are compared with results of experiments in which H2O2 and OH are present as radiolytic products.  相似文献

This paper considers a deterministic model, where selection acts at both the diploid and haploid phases of development. The stability criteria for the maintenance of a protected polymorphism are derived. The implications of the model are examined through several examples, with emphasis on the comparison of these conditions to those of models that view selection as acting only at the diploid or haploid phases. It is found that the consideration of both phases broadens the conditions over such models.  相似文献
