In this study, mainly biological treatment of prawn waste for chitin production was investigated. Lactic acid and protease fermentations were applied to extract chitin from prawn waste in the presence of various glucose concentrations. The results obtained were also compared with those of chemical method which was consisted of first mineral removal and then protein removal sequence. Different strategies were applied using lactic acid producing bacterium, Lactococcus lactis, and a protease producer, marine bacterium Teredinobacter turnirae. Both bacteria were first cultivated individually and then cofermented. In their individual cultivation, L. lactis removed the inorganic materials efficiently, while T. turnirae performed better in deproteinization process. Cofermentation of both bacteria was also conducted using three different protocols. The highest process yield (95.5%) was obtained when T. turnirae was first inoculated. Although the extraction of chitin by biological treatment was incomplete compared to the chemical method, the biological treatment employed here could still be considered as an alternative method in a more environmentally benign approach.  相似文献

Perfluorodecalin, a perfluorocarbon (PFC), was used in this investigation as a dissolved oxygen carrier in the media of Streptomyces coelicolor cultures. The effects of different concentrations of PFC, PFC emulsified with pluronic F-68 and pluronic alone were investigated in the shake-flask cultures using both defined and complex media. In the defined medium with PFC alone, the maximum biomass and actinorhodin concentrations and the volumetric substrate consumption rates increased with increasing PFC concentration. They decreased dramatically, however, when the PFC concentration exceeded 50% (v/v). Emulsifying the PFC with pluronic F-68 resulted in a significant increase in antibiotic concentration while growth was unaffected. The inclusion of more than 4 g/l pluronic alone in the fermentation medium inhibited the growth. In the complex medium with 40% (v/v) PFC, although the final antibiotic concentration was unaffected, the onset of actinorhodin accumulation was 2 days earlier than that in the control. It was demonstrated that PFC and emulsified PFC did not have any deleterious effects on S. coelicolor cultures.  相似文献

Diclofenac sodium (DS) is used primarily to treat fever and to alleviate pain and inflammation. We investigated the effects of DS exposure during gestation on the testes of rat pups to investigate the safety of its use during the prenatal period. Pregnant rats were separated into control, saline, low dose, medium dose and high dose groups. DS was given between weeks 15 and 21 of gestation. Total numbers of spermatogonia and Sertoli cells were counted in the testes of 7-day-old male rats using the physical disector method. By the end of the study, the total number of Sertoli cells was decreased significantly in a dose dependent manner in the medium and high dose groups compared to controls. No significant differences were found in the total number of spermatogonia in the control, saline and low dose DS groups. Medium and high dose DS administration reduced the total number of spermatogonia compared to other groups. We suggest that prenatal administration of DS can cause deleterious effects on the testis development, especially in high doses.  相似文献

The growth of Saccharomyces cerevisiae was enhanced by using perfluorocarbon (PFC) while it was inhibited by pluronic F-68 alone. The use of 15%(v/v) PFC resulted in almost twice as much biomass concentration compared to the control. The inclusion of PFC emulsion in the culture medium also increased the maximum biomass concentration, but not as much as in the case of pure PFC.  相似文献

Summary The production of actinorhodin by Streptomyces coelicolor in a defined medium was examined using spore and vegetative inocula. The spore inoculum yielded higher concentrations of biomass and actinorhodin as well as a higher maximum specific growth rate compared with the vegetative inoculum. Nevertheless, the productivity of the batch culture for actinorhodin formation with vegetative inoculum was higher than that with spore inoculum.  相似文献

Parkinson disease (PD) is a common neurodegenerative condition associated with degeneration of dopaminergic neurons in the zona compacta of the substantia nigra. There is increasing evidence that genetic factors play a role in the etiology of PD, although genetic heterogeneity is likely. An autosomal dominant syndrome with many similarities to sporadic PD has been mapped to 4q21-22 in a large Italian pedigree and has been found to be due to mutation of the alpha-synuclein gene. However, this gene appears to account for only a minority of PD, and a susceptibility locus for autosomal dominant parkinsonism has recently been mapped, on 2p13. Autosomal recessive juvenile parkinsonism (JP), which shows marked clinical similarity to PD, maps to 6q25.2-q27. We found linkage to this region in a group of 15 families from four distinct ethnic backgrounds. A full genomic screen excluded other candidate regions. We have constructed a detailed genetic map of the linked region and have mapped the position of the manganese superoxide dismutase gene (SOD2). Recombination events restricted the JP locus to a 6.9-cM region and excluded SOD2. The apparent homozygosity for null alleles at D6S955 in one family suggested a deletion and finer localization of the JP locus.  相似文献

Actinorhodin production by Streptomyces coelicolor A3(2) immobilised naturally in a porous support material was investigated in a 20 l bioreactor. The effect of pH-control on actinorhodin fermentation was conducted by performing different strategies. The combination of pH-control and immobilisation of the cells produced a different result than pH-control with freely suspended cells. The control of pH at 7.2 in an immobilised cell system altered the physiology of the cells shifting the product, actinorhodin, to another pigmented secondary metabolite, undecylprodigiosin. Although the cells were biologically active in such an environment, they were unable to produce actinorhodin. In the freely suspended cell system, however, actinorhodin biosynthesis was not hindered by controlling the medium pH.  相似文献
