Truffles have evolved from epigeous (aboveground) ancestors in nearly every major lineage of fleshy fungi. Because accelerated rates of morphological evolution accompany the transition to the truffle form, closely related epigeous ancestors remain unknown for most truffle lineages. This is the case for the quintessential truffle genus Tuber, which includes species with socio-economic importance and esteemed culinary attributes. Ecologically, Tuber spp. form obligate mycorrhizal symbioses with diverse species of plant hosts including pines, oaks, poplars, orchids, and commercially important trees such as hazelnut and pecan. Unfortunately, limited geographic sampling and inconclusive phylogenetic relationships have obscured our understanding of their origin, biogeography, and diversification. To address this problem, we present a global sampling of Tuberaceae based on DNA sequence data from four loci for phylogenetic inference and molecular dating. Our well-resolved Tuberaceae phylogeny shows high levels of regional and continental endemism. We also identify a previously unknown epigeous member of the Tuberaceae – the South American cup-fungus Nothojafnea thaxteri (E.K. Cash) Gamundí. Phylogenetic resolution was further improved through the inclusion of a previously unrecognized Southern hemisphere sister group of the Tuberaceae. This morphologically diverse assemblage of species includes truffle (e.g. Gymnohydnotrya spp.) and non-truffle forms that are endemic to Australia and South America. Southern hemisphere taxa appear to have diverged more recently than the Northern hemisphere lineages. Our analysis of the Tuberaceae suggests that Tuber evolved from an epigeous ancestor. Molecular dating estimates Tuberaceae divergence in the late Jurassic (∼156 million years ago), with subsequent radiations in the Cretaceous and Paleogene. Intra-continental diversification, limited long-distance dispersal, and ecological adaptations help to explain patterns of truffle evolution and biodiversity.  相似文献

Series of purine and pyrazolo[3,4-d]pyrimidine inhibitors of phosphatidylinositol-3-kinases (PI3K) have been prepared. The optimized purine inhibitors show good potency in a PI3K p110α (PI3K-α) fluorescence polarization assay with good selectivity versus PI3K p110γ (PI3K-γ) and the mammalian target of rapamycin (mTOR). The related pyrazolo[3,4-d]pyrimidines show potent PI3K-α and mTOR inhibition with good selectivity versus PI3K-γ. Representative compounds showed activity in a cellular proliferation assay against Caco-2 colorectal, LoVo colorectal and PC3MM2 prostate adenocarcinoma cancer cells. Signaling through the PI3K pathway was confirmed via inhibition of phospho-AKT in MDA-361 cells.  相似文献

A series of mono-morpholino 1,3,5-triazine derivatives (8a8q) bearing a 3-oxa-8-azabicyclo[3.2.1]octane were prepared and evaluated for PI3-kinase/mTOR activity. Replacement of one of the bis-morpholines in lead compound 1 (PKI-587) with 3-oxa-8-azabicyclo[3.2.1]octane and reduction of the molecular weight yielded 8m (PKI-179), an orally efficacious dual PI3-kinase/mTOR inhibitor. The in vitro activity, in vivo efficacy, and PK properties of 8m are discussed.  相似文献

Background

Early diagnosis of dengue can assist patient triage and management and prevent unnecessary treatments and interventions. Commercially available assays that detect the dengue virus protein NS1 in the plasma/serum of patients offers the possibility of early and rapid diagnosis.

Methodology/Principal Findings

The sensitivity and specificity of the Pan-E Dengue Early ELISA and the Platelia™ Dengue NS1 Ag assays were compared against a reference diagnosis in 1385 patients in 6 countries in Asia and the Americas. Platelia was more sensitive (66%) than Pan-E (52%) in confirmed dengue cases. Sensitivity varied by geographic region, with both assays generally being more sensitive in patients from SE Asia than the Americas. Both kits were more sensitive for specimens collected within the first few days of illness onset relative to later time points. Pan-E and Platelia were both 100% specific in febrile patients without evidence of acute dengue. In patients with other confirmed diagnoses and healthy blood donors, Platelia was more specific (100%) than Pan-E (90%). For Platelia, when either the NS1 test or the IgM test on the acute sample was positive, the sensitivity versus the reference result was 82% in samples collected in the first four days of fever. NS1 sensitivity was not associated to disease severity (DF or DHF) in the Platelia test, whereas a trend for higher sensitivity in DHF cases was seen in the Pan-E test (however combined with lower overall sensitivity).

Conclusions/Significance

Collectively, this multi-country study suggests that the best performing NS1 assay (Platelia) had moderate sensitivity (median 64%, range 34–76%) and high specificity (100%) for the diagnosis of dengue. The poor sensitivity of the evaluated assays in some geographical regions suggests further assessments are needed. The combination of NS1 and IgM detection in samples collected in the first few days of fever increased the overall dengue diagnostic sensitivity.  相似文献

A pure mechanical anisotropic model of a tree trunk has been developed based on the 3D finite element method. It simulates the microscopic structure of vessels in the trunk of a European beech (Fagus sylvatica) in order to study and analyse its mechanical behaviour with different configurations of pressures in the conduits of xylem and phloem. The dependence of the strains at the inner bark was studied when sap pressure changed. The comparison with previously published experimental data leads to the conclusion that a great tensile stress—or ‘negative pressure’—must exist in the water column in order to achieve the measured strains if only the mechanical point of view is taken into account. Moreover, the model can help to design experiments where qualitatively knowing the strains and the purely mechanical behaviour of the tree is required.  相似文献

The main objective of this paper is to discuss new procedures of the isolation of Hyaluronan. Hyaluronic acid can be obtained from human umbilical cord residual, which is obtained from other biopharmaceutical productions. The route involves treatment of human umbilical cord residuals with sodium chloride solution, followed by ammonium quaternary salt solution precipitation; the solid is re-suspended in calcium chloride solution in order to dissociate the hyaluronan ammonium quaternary salt complex followed by ethanol-induced precipitation to give a product. The product was purified four times by chloroform extraction, and characterized by chemical methods such as the Blumenkrantz and Asboe-Hansen uronic technique for uronic acid determination, Elson Morgan qualitative tests for hexosamines, intrinsic viscosity, ion-exchange chromatography, and 13C NMR spectroscopy. The results showed that the product might be used in the formulation of ointment, lotion and cream for the treatment of skin diseases.  相似文献

 We followed the colonization frequency of ectomycorrhizal (EM), vesicular-arbuscular mycorrhizal (VAM), and dark septate (DS) fungi in 1- to 5-month-old bishop pine seedlings reestablishing after a wildfire. Seedlings were collected on a monthly basis at either a VAM-dominated chaparral scrub site or an EM-dominated forest site, both of which were burned. In both vegetation types, fully developed EM were observed from the third month after germination. EM fungi observed on the seedlings from the scrub site were limited to Rhizopogon subcaerulescens, R. ochraceorubens and Suillus pungens. Seedlings from the forest were colonized by a greater variety of EM fungi including Amanita spp., Russula brevipes and a member of the Cantharellaceae. VAM structures (vesicles, arbuscules or hyphal coils) were observed in the seedling root systems beginning 1 month after germination at the scrub site and 3 months after germination at the forest site. Seedlings from the scrub site consistently had more frequent VAM fungal colonization than those from the forest site through the fifth month after germination. DS fungi were observed in most seedlings from both the scrub and forest sites beginning in the first month post-germination. We propose that these fungi survived as a resident inoculum in the soils and did not disperse into the sites after the fire. Accepted: 14 February 1998  相似文献
