Antibody-dependent cytotoxic activity of the lymphocytes and the cytological characteristics of immune cell complexes in vitro

Bovine blood lymphocytes showed a high antibody-dependent (AD) cytotoxicity against target cultured L cells. The cytological analysis of immune cellular complexes, formed by affector lymphocytes with target cells during the cytotoxic reaction in vitro, revealed that adsorption of AD lymphocytes to target cells was completed within the first hours of incubation. Temporal parameters of contact between effector and target cells and dissociation of adsorbed lymphocytes varied widely, but after 48 hours of incubation the number of immune cellular complexes decreased by 5.6%. The morphological modification of AD effector lymphocytes during interaction with target cells is described.     

Effect of short-term culture of blood lymphocytes in vitro on the ability of Fcgamma-receptors to bind IgG1 and IgG2 and mediation of antibody-dependent cytotoxicity

The comparative study incubation influence at 37 degrees C on the lymphocyte Fc gamma-receptor (Fc gamma R) binding ability to IgG1 and IgG2 and on the antibody-dependent cell mediated cytotoxicity (ADCC) was performed. By means of the fluorometric binding assay it was determined that during 30 min of incubation the binding ability of Fc gamma R decreased while after 180 min of incubation it increased. Upon comparing both IgG1 and IgG2 binding abilities to lymphocytes no statistically important changes were noticed (p < 0.05). The changes in the binding ability of Fc gamma R for IgG1 and IgG2 had a similar pattern to the changes.     

Quantitative parameters of the IgG binding by Fc receptors and the functional activity of blood lymphocytes in healthy cattle and cattle with chronic lympholeukemia

The binding of IgG to lymphocyte Fc receptors in the blood of healthy cattle and of cattle with chronic lympholeukemia has been studied by fluorometric techniques before and after the incubation of lymphocytes in a serum-free medium at 37 degrees C. The study has shown that changes in the intensity of binding of IgG to Fc receptors of normal and leukemic lymphocytes correlate with changes in the cell-mediated cytotoxic activity of the corresponding lymphocytes. Lower cell-mediated cytotoxic activity of leukemic lymphocytes in comparison with that of normal lymphocytes was parallelled by a lower association constant of IgG with leukemic lymphocytes Fc receptors.
