Insulin-like growth factor II binding and action in human fetal fibroblasts   总被引:5,自引:0,他引:5  
To investigate the role of insulin-like growth factor II (IGF-II) in human prenatal growth, IGF-II binding and biological action were studied in four lines of fetal and three lines of postnatal human fibroblasts. Specific binding of IGF-II was similar in both groups: 15.7% and 14.9% for fetal and postnatal fibroblasts, respectively. This was 5-10 times the amount of IGF-I binding found in these cells. IGF-I and IGF-II caused dose-dependent increases in [14C]aminoisobutyric acid (AIB) uptake. IGF-II was sevenfold less potent than IGF-I in stimulating this metabolic response in both fetal and postnatal fibroblasts. The maximal effect of IGF-II in stimulating [14C]AIB uptake approach that of IGF-I. Similar results were obtained when IGF-I and IGF-II stimulation of [3H]thymidine incorporation was compared in fetal and postnatal fibroblasts. Incubation in the presence of alpha IR-3, a monoclonal antibody to the type I IGF receptor, inhibited the ability of both IGF-I and IGF-II to stimulate [14C]AIB uptake and [3H]thymidine incorporation in fetal and postnatal cells. A monoclonal antibody to the insulin receptor did not affect IGF action. These data indicate that IGF-II is a potent metabolic and mitogenic stimulus for human fetal fibroblasts. However, despite the presence of abundant type II IGF receptors on both fetal and postnatal human fibroblasts, IGF-II stimulation of amino acid transport and DNA synthesis appears to be mediated through the type I rather than through its own type II IGF receptor.  相似文献

Summary Zooplankton and water samples were collected at weekly intervals between April 25 and May 30, 1986 in Barrow Strait, N.W.T. (Canadian Arctic Archipelago). In tows from 0–30 m, the zooplankton community (>202 m) was dominated by Pseudocalanus. The population was apparently growing and developing as shown by an increase in the proportion of adults (stage VI) and decreases in the proportion of stages III, IV, and V as the season progressed. Respiration and excretion rates of the Pseudocalanus populations were probably linked, there being an immediate increase in excretion rate, accompanying an increase in feeding rate when chlorophyll concentrations increased, which was followed by a smaller increase in respiration rate after a time lag. Hence, there was a large decrease in the ON ratio. Increased metabolism coincided with changes in the population structure, as did protease and bodily protein, but could not be clearly linked to dietary acclimation. Only laminarinase activity could be statistically related to an identifiable fraction of potential nutritional value in the water, particulate soluble carbohydrate, but neither showed overall seasonal change.  相似文献

The in vitro activity of several new imidazoles, cloconazole, sulconazole, butoconazole, isoconazole and fenticonazole, were compared with those of amphothericin B, flucytosine, and three azoles: econazole, miconazole and ketoconazole against isolates of pathogenic Candida. A total of 186 clinical isolates of 10 species of the genus Candida and two culture collection strains were tested by an agar-dilution technique. Isoconazole was the most active azole, followed by butoconazole and sulconazole. Differences between some of the species in their susceptibility to the antifungal agents were noted. Sulconazole and cloconazole had the highest activity in vitro against 106 isolates of C. albicans. Butoconazole and isoconazole were also very active against isolates of C. albicans, and were the most active azole compounds against 80 isolates of Candida spp.  相似文献

After conjugation in Tetrahymena thermophila, the old macronuclei degenerate, and new macronuclei (anlagen) develop. During anlagen development a number of DNA sequences found in the micronuclear genome (micronuclear limited sequences) are eliminated from the anlagen. A cloned copy of a repetitive micronuclear limited sequence has been used to determine the developmental stage at which micronuclear limited sequences are eliminated. DNAs from anlagen of various developmental stages were examined by Southern analysis. It was found that micronuclear limited sequences are present in 4C anlagen and essentially absent in 8C and 16C anlagen. The precipitous loss of these sequences in the 8C anlagen rules out under-replication as the mechanism for the loss and suggests that these sequences are specifically degraded early during anlagen development.  相似文献

Failure of epididymal spermatozoa from T/t mutant mice, but not from t/t individuals, to fertilize oocytes in vitro was partially overcome by opening a small aperture in the zona pellucida with acidified Tyrode's solution to permit direct access of the spermatozoon to the vitellus. This study provides a model system to evaluate requirements for successful zona drilling in the treatment of human infertility and further insights into the effects of the t complex on sperm fertility.  相似文献

A novel experimental method was developed which allows the determination of the threshold concentration of sucrose by use of a linear sucrose gradient in water. With this method a continuous tasting of the test-liquid is possible. A panel of 15 persons experienced in taste-testing was used. Three gradients of different steepness were applied: 0 to 1.5% (w/w) sucrose in 2 min (I), 3 min (II) and 4 min (III). The results of the new method were compared with those of the standard method (DIN). With gradients I and II we found values which were significantly higher than those of the standard method (I: 0.49% (w/w); II: 0.46% (w/w); DIN: 0.31% (w/w)), whereas with gradient III the same threshold value was found as with the DIN-Method (III: 0.32% (w/w)).  相似文献

Synopsis The spawning periodicity and mating behavior of a northern population of the Atlantic silverside, Menidia menidia, was studied both in the field (Salem Harbor, Massachusetts) and in laboratory experiments. Spawning in the field coincided with new and full moons. Under conditions of unlimited food availability in artificial outdoor pools that received natural celestial illumination or in indoor aquaria receiving no evening illumination, spawning occurred every 1–3 days rather than fortnightly. These results suggest that tidal influences are a primary factor that synchronizes spawning in M. menidia. Published reports of diel time of spawning in both marine and freshwater species of Menidia indicate a generic tendency to spawn during mid-morning. This pattern coupled with high tides that cover suitable spawning substrates during mid-morning only at fortnightly intervals could account for the semilunar spawning cycle in marine populations of Menidia. Although general characteristics of the reproductive ecology of M. menidia in Massachusetts were similar to a thoroughly studied population in South Carolina, several differences were evident. The breeding season was shorter in Massachusetts (late; April–June), and occurred over a much lower range of temperatures (9–21°C). Intensity and frequency of spawning was correlated with the height of high tide. Eggs were deposited only on mats of intertidal, filamentous algae rather than on roots or stems of Spartina alterniflora or other intertidal vegetation. Promiscuous spawning occurred in small, highly male-dominated groups of fish, primarily after the tide had begun to recede, and often at the water's edge. No predation on spawning adults was observed but the mummichog, Fundulus heteroclitus, fed daily on developing embryos during high tide.
