Maintenance of the cellobiose utilization genes of Escherichia coli in a cryptic state 总被引:6,自引:1,他引:5
The genes for cellobiose utilization are normally cryptic in Escherichia
coli. The cellobiose system was used as a model to understand the process
by which silent genes are maintained in microbial populations. Previously
reported was (1) the isolation of a mutant strain that expresses the
cellobiose-utilization (Cel) genes and (2) that expression of those genes
allows utilization of three beta- glucoside sugars: cellobiose, arbutin,
and salicin. The Cel gene cluster has now been cloned from that mutant
strain. In the course of locating the Cel genes within the cloned DNA
segment, it was discovered that inactivation of the Cel-encoded hydrolase
rendered the host strain sensitive to all three beta-glucosides as potent
inhibitors. This sensitivity arises from the accumulation of the
phosphorylated beta- glucosides. Because even the fully active genes
conferred some degree of beta-glucoside sensitivity, the effects of
cellobiose on a series of five Cel+ mutants of independent origin were
investigated. Although each of those strains utilizes cellobiose as a sole
carbon and energy source, cellobiose also acts as a potent inhibitor that
reduces the growth rate on glycerol 2.5-16.5-fold. On the other hand,
wild-type strains that cannot utilize cellobiose are not inhibited. The
observation that the same compound can serve either as a nutrient or as an
inhibitor suggests that, under most conditions in which cellobiose will be
present together with other resources, there is a strong selective
advantage to having the cryptic (Cel0) allele. In those environments in
which cellobiose is the sole, or the best, resource, mutants that express
the genes (Cel+) will have a strong selective advantage. It is suggested
that temporal alternation between these two conditions is a major factor in
the maintenance of these genes in E. coli populations. This alternation of
environments and fitnesses was predicted by the model for cryptic-gene
maintenance that was previously published.

Selection-induced mutations are nonrandom mutations that occur as specific
and direct responses to environmental challenge. Examples of
selection-induced mutations have been reported both in bacteria and in
yeast. I previously showed (Hall 1988) that excisions of the mobile genetic
element IS150 from within bglF are selection induced and argued that they
occurred because they were potentially advantageous under the selective
conditions employed. Mittler and Lenski (Mittler and Lenski 1992) have
argued that such excisions are not selection induced but that they occur
randomly in nondividing cells. Here I provide further evidence that IS150
excisions are induced by selection and that the excisions are immediately,
rather than only potentially, advantageous to the cell. I also provide
evidence that excisions, which Mittler and Lenski claim occur randomly in
saturated broth cultures, actually occur after samples from those cultures
are plated onto selective medium.

Reproductive patterns are analysed in annual legumes of west Asia, and their relationships to increasing aridity determined by multivariate analysis. Dormancy, seed size, dispersal and fecundity are shown to be partially substitutable in terms of their effect on survival and population growth.The range of patterns show greatest diversity under mesic conditions in coastal, mediterranean areas with high winter rainfall, low incidence of frost and long growing season. Increasing aridity leads to increasing reproductive homogeneity, in which a subset of patterns-those composed of high levels of seed dormancy, high seed to pod ratios, restricted dispersal capabilities and early flowering-become predominant. These findings corroborate earlier theoretical and empirical evidence concerning desert annuals.The majority of widespread species are shown to possess arid-type patterns. This exerts considerable influence on both the type and degree of ecotypic differentiation within species. The only feature that responds consistently to climatic change is flowering time, particularly among species characterized by high seed dormancy. In the few widespread species that do not display arid-type characteristics, ecotypic differentiation appears more frequently in a larger suite of traits.The unequal environmental demands made on species showing different levels of dormancy (e.g. variation in length of growing season from year to year and place to place) leads to strong asymmetries governing the relationships between reproduction and ecological amplitude. In this respect arid-type strategists with high seed dormancy appear to have greater chances of expansion than others. Some of the important controlling events regulating eukaryotic S-phase progression are considered to occur late in the G1 stage of the cell cycle. We show here that stimulation of DNA synthesis in bone marrow-derived macrophages (BMM) by macrophage CSF-1 is preceded by G1 expression of three genes which encode proteins associated with the DNA synthesis machinery--the M1 and M2 subunits of ribonucleotide reductase and proliferating cell nuclear Ag (PCNA). Increased expression for these genes correlated well with the mitogenic response and sustained expression required de novo RNA and protein synthesis and also the presence of CSF-1 for at least most of G1. Inhibitors of BMM proliferation (LPS, TNF-alpha, IFN-gamma, and cAMP elevating agents) suppressed CSF-1-induced expression of M1, M2, and PCNA mRNA measured at 22 h. This suppression occurred even when added up to 12 h after the CSF-1, a period coinciding with the G1/S-phase boundary. The delayed kinetics of this effect parallels the ability of these agents to maximally inhibit CSF-1-induced BMM DNA synthesis when added at similar times. Decreased expression of M1, M2, and PCNA was not merely a consequence of DNA synthesis inhibition because the S-phase inhibitor, hydroxyurea, did not suppress CSF-1-induced gene expression. These results suggest that inhibition of DNA synthesis by antiproliferative agents involves inhibition of expression of several genes associated with the DNA synthesis machinery. 相似文献

High-level expression of the Endo-beta-N-acetylglucosaminidase F2 gene in E.coli: one step purification to homogeneity 总被引:1,自引:0,他引:1
The Endo F2gene was overexpressed in E.coli as a fusion protein joined to
the maltose-binding protein. MBP-Endo F2was found in a highly enriched
state as insoluble, inactive inclusion bodies. Extraction of the inclusion
bodies with 20% acetic acid followed by exhaustive dialysis rendered the
fusion protein active and soluble. MBP-Endo F2was digested with Factor
Xaand purified on Q-Sepharose. The enzyme was homogeneous by SDS-PAGE, and
appeared as a single symmetrical peak on HPLC. Analysis of the
amino-terminus demonstrated conclusively that recombinant Endo F2was
homogeneous and identical to the native enzyme.

Primatologists have recently suggested that they may be full species and that the Bornean species could be divided into 3–5
subspecies. Statistical analyses of factors affecting the mortality, fertility, and well-being of captive orangutans have
revealed some important differences between Bornean, Sumatran, and hybrid orangutans that could support the suggestion. Hybrid
orangutans appeared to be genetically weaker, with much lower survival rates than pure subspecific individuals. The weakness
also may have contributed to their higher rate of infant rejection. In addition, infertility was 3 times higher in Bornean
