A DNA macroarray was developed to provide the ability to detect multiple foodborne pathogens in fresh chicken meat. Probes targeted to the 16S rRNA and genus- and species-specific genes, including fimY, ipaH, prfA, and uspA, were selected for the specific detection of Salmonella spp., Shigella spp., Listeria monocytogenes, and Escherichia coli, respectively. The combination of target gene amplification by PCR and a DNA macroarray in our system was able to distinguish all target bacteria from pure cultures with a detection sensitivity of 105 c.f.u. ml?1. The DNA macroarray was also applied to 10 fresh chicken meat samples. The assay validation demonstrated that by combining the enrichment steps for the target bacteria and the DNA macroarray, all 4 target bacteria could be detected simultaneously from the fresh chicken samples. The sensitivity of L. monocytogenes and Shigella boydii detection in the fresh chicken samples was at least 10 and 3 c.f.u. of the initial contamination in 25 g samples, respectively. The advantages of our developed protocol are high accuracy and time reduction when compared to conventional culture. The macroarray developed in our investigation was cost effective compared to modern oligonucleotide microarray techniques because there was no expensive equipment required for the detection of multiple foodborne pathogens.  相似文献

Introduction

We have previously reported results of precipitation studies for neonatal parenteral nutrition solutions containing calcium chloride and sodium phosphate using visual methods to determine compatibility. The purpose of this study was to do further testing of compatibility for solutions containing calcium chloride using more sensitive methods.

Methods

Solutions of Trophamine (Braun Medical Inc, Irvine, CA) and Premasol (Baxter Pharmaceuticals, Deerfield, IL) were compounded with calcium chloride and potassium phosphate. Controls contained no calcium or phosphate. After incubation at 37° for 24 hours solutions without visual precipitation were analyzed to determine mean particle size using dynamic light scattering from a laser light source.

Results

Particle sizes were similar for control solutions and those without visual precipitation and a mean particle size <1000 nm. Compatible solutions were defined as those with added calcium and phosphate with no visual evidence of precipitation and mean particle size <1000 nm. In solutions containing 2.5–3% amino acids and 10 mmol/L of calcium chloride the maximum amount of potassium phosphate that was compatible was 7.5 mmol/L.

Conclusion

Maximum amounts of phosphate that could be added to parenteral nutrition solutions containing Trophamine and calcium chloride were about 7.5–10 mmol/L less for a given concentration of calcium based upon laser methodology compared to visual techniques to determine compatibility. There were minor differences in compatibility when adding calcium chloride and potassium phosphate to Premasol versus Trophamine.  相似文献

The purpose of this study is to access the existing awareness of nearly forgotten Thai detergent plants by the use of chemometrics tool. A Northern Thai forest dependent community was chosen as it played vital role on knowledge retaining of plant utilisations. For initial perception, ethnobotanical survey was conducted to determine usage of plants by the community. Then the utilised plant parts were screened for phytochemicals and their relationships with the defined cleansing terms (viz., shampoo, scrub, detergent, soap, scent and spiritual) were analysed by Principal Component Analysis (PCA). From the results, the most cited plants as known, used and found were Acacia concinna, Clitoria ternetea, Oryza sativa and Citrus hystrix. Biometric analyses advised that knowledge of detergent plant utilisation was well preserved at all age ranges and it was not variable with genders. Cluster analysis described that term ‘spiritual’ was not narrated with cleansing properties. For phytochemical analysis, plant extracts showed positive variable of bioactive ingredients and the main compounds in the extracts was saponins. These findings confirmed that the knowledge of indigenous plant utilisation was reserved by the forest dependent community and the information is beneficial toward local plant conservation movement.  相似文献
