Using the immunofluorescence approach, we have determined that the recently detected protein kinases, among which are RhoA-activated kinase, integrin-linked kinase, zipper interacting protein kinase, and death-associated protein kinase, which are capable of phosphorylating myosin, are localized in the Z-lines sarcomeres of human myocardium. Additionally, we studied the content of integrin-linked and zipper interacting protein kinases in human embryonic myocardium, as well as in normal and hypertrophic adult human heart. The content of these protein kinases in adult normal myocardium increases in comparison with the embryonic heart. The content of integrin-linked and zipper interacting protein kinases in hypertrophic myocardium is higher compared with the normal adult heart. The data obtained suggest the involvement of these protein kinases in the development and hypertrophy of human heart.  相似文献

Incubation with microbial culture supernatants improved essential oil yield from Aquilaria subintegra woodchips. The harvested woodchips were incubated with de man, rogosa and sharpe (MRS) agar, yeast mold (YM) agar medium and six different microbial culture supernatants obtained from Lactobacillus bulgaricus, L. acidophilus, Streptococcus thermophilus, Lactococcus lactis, Saccharomyces carlsbergensis and S. cerevisiae prior to hydrodistillation. Incubation with lactic acid bacteria supernatants provided higher yield of agarwood oil (0.45% w/w) than that obtained from yeast (0.25% w/w), agar media (0.23% w/w) and water (0.22% w/w). The composition of agarwood oil from all media and microbial supernatant incubations was investigated by using gas chromatography-mass spectrometry. Overall, three major volatile profiles were obtained, which corresponded to water soaking (control), as well as, both YM and MRS media, lactic acid bacteria, and yeast supernatant incubations. Sesquiterpenes and their oxygenated derivatives were key components of agarwood oil. Fifty-two volatile components were tentatively identified in all samples. Beta-agarofuran, α-eudesmol, karanone, α-agarofuran and agarospirol were major components present in most of the incubated samples, while S. cerevisiae-incubated A. subintegra provided higher amount of phenyl acetaldehyde. Microbial culture supernatant incubation numerically provided the highest yield of agarwood oil compared to water soaking traditional method, possibly resulting from activity of extracellular enzymes produced by the microbes. Incubation of agarwood with lactic acid bacteria supernatant significantly enhanced oil yields without changing volatile profile/composition of agarwood essential oil, thus this is a promising method for future use.  相似文献

We have shown that Y-27632, an inhibitor of Rho-associated kinase (ROCK), delays sarcomere assembly in cultured rat neonatal cardiomyocytes pretreated with angiotensin II. Y-27632 affects the beat rate of cardiomyocytes, but this effect is only observed at high cell density and, therefore, seems to be related to impairment of cell-cell interactions. Consistent with this suggestion, ROCK2 is localized in myofibrillar Z-disks of human myocardium as well as in intercalated disks. We propose that ROCK participates in maturation of the myocardial contractile system through phosphorylation of its molecular targets in Z-disks and in intercalated disks.  相似文献

The skeletal myosin light chain kinase (skMLCK) was identified in human and chicken embryo myocardium but not in embryo and adult rat heart using western blotting. The content of skMLCK and myosin-activating protein kinases: RhaA-activated protein kinase (ROCK), integrin-linked protein kinase (ILK), and zipper-interacting protein kinase (ZIPK) was compared in normal human myocardium and the hearts of patients with dilated cardiomyopathy (DCM). It was demonstrated that the content of skMLCK, ROCK and ILK increases in DCM whereas the content of ZIPK decreases. The results obtained may reflect compensatory processes in cardiomyocytes in DMC, which are aimed at increasing their viability and contractility.  相似文献

Bioassay‐guided fractionation was conducted on dichloromethane extract from the rhizomes of Globba schomburgkii Hook.f., which have previously been reported as the part with the highest antibacterial activity. 10 fractions and 20 sub‐fractions were obtained and evaluated for their potency against various strains of bacteria. The most active sub‐fractions were 8 times more effective against Staphylococcus aureus and Micrococcus luteus than the original crude extract. Moreover, two pure compounds, namely petasol and (E)‐15,16‐dinorlabda‐8(17),11‐dien‐13‐one, were successfully isolated and characterized for the first time from this plant species. Untargeted compound analysis of all fractions and sub‐fractions was performed by gas chromatography hyphenated with mass spectrometry, leading to positive identification of 167 compounds according to comparison with the mass spectrum and retention index database, 137 of which have never been reported for G. schomburgkii. The correlation between antibacterial activity and composition of each fraction suggests that the bioactive compounds could be 4,8‐β‐epoxycaryophyllene, methyl isocostate, (E)‐labda‐8(17),12‐diene‐15,16‐dial, α‐kessyl acetate, zederone, clovanediol, ledene oxide‐(I), alantolactone, or 8α,11‐elemadiol.  相似文献

Skeletal myosin light chain kinase in the myocardia of various animal species was identified by immunoblotting. The myocardial concentrations of this protein and myosin-activating protein kinases (RhoA-activated kinase, integrin-linked kinase, and zipper-interacting kinase) were compared in healthy humans and patients with dilated cardiomyopathy. Skeletal myosin light chain kinase was detected in the human and chicken embryo hearts, rather than in the embryonic and adult rat hearts. In the myocardium of patients with dilated cardiomyopathy, the concentrations of myosin light chain kinase, RhoA-activated kinase, and integrin-linked kinase increase and the concentration of zipper-interacting kinase decreases. The results obtained are likely to characterize compensatory processes in cardiomyocytes in dilated cardiomyopathy that are aimed at increasing their viability and contractility.  相似文献

The ability of giant hogweeds to form monodominant communities and even pure monostands in invaded areas has been well documented. Understanding of the mechanisms leading to monostand formation can aid in determining the limitations of existing community ecology models and establishing an effective management plan for invasive species elimination. The aim of this observational study was to investigate traits of Heracleum sosnowskyi plants (demography, canopy structure, morphology and physiology) of the plants in a pure stand in an invaded area useful for understanding potential monostand formation mechanisms. All measurements were performed in one typical Heracleum sosnowskyi monostand located in an abandoned agriculture field located in Syktyvkar city suburb (North-east Russia). This monostand consisted of five main plant growth stages: seed, seedling, juvenile, vegetative adult, and generative adult. Plants of all stages began to grow simultaneously shortly after the snowmelt, at the same time as spring ephemeral plant species grew. The density of generative plants did not change during the vegetation period, but the density of the other plant stages rapidly decreased after the formation of a tall (up to 2–2.5 m) and dense (Leaf area index up to 6.5) canopy. The canopy captured approximately 97% of the light. H. sosnowskyi showed high (several orders of magnitude higher than average taiga zone grasses) photosynthetic water use efficiency (6–7 μM CO2/μM H2O). Formation of H. sosnowskyi monostands occurs primarily in disturbed areas with relatively rich and well-moistened soils. Early commencement of growth, rapid formation of a dense canopy, high efficiency of light and water use during photosynthesis, ability of young plants to survive in low light conditions, rapid recovery of above-ground plant parts after damage, and the high density of the soil seed bank are the most important traits of H. sosnowskyi plants for monostand formation in invaded areas.  相似文献

Myosin light chain kinase (MLCK) is a key regulator of various forms of cell motility involving actin and myosin II. MLCK is widely present in vertebrate tissues including the myocardium. However, the role of MLCK in cardiomyocyte function is not known. Previous attempts to gain insight into possible roles and identify potential molecular partners were disappointing and equivocal due to cross reactivity of early antibodies with striated muscle MLCK, which has a different genetic locus and a divergent amino acid sequence from the above mentioned enzyme. Using an immunofluorescence approach and a panel of antibodies directed against MLCK, cytoskeletal, and sarcomeric proteins, we localized MLCK to myofibril precursors and Z-lines of sarcomeres in embryonic and adult cardiomyocytes. The same structures contained nonmuscle myosin IIB implicating this protein as a possible target of MLCK. Our results suggest a role for MLCK in cardiomyocyte differentiation and contraction through regulation of nonmuscle myosin IIB.  相似文献
