THE mechanism, known as the carrier effect, whereby immunity to one or more determinant groups enhances the response to other determinants on the same multivalent antigen, was first recognized in delayed hypersensitivity to haptens, in which, for an appreciable response, the hapten must be coupled to the same protein carrier for priming and challenge1, 2. Carrier specificity has also been demonstrated in the secondary antibody responses to hapten protein conjugates3. Two alternative hypotheses have been advanced to explain this specificity. The “local environment” hypothesis supposes that the hapten-sensitive cell recognizes both the hapten and the carrier determinants. However, the antihapten antibodies produced do not distinguish details of the carrier molecule and so do not reflect the specificity of the cellular receptor. Furthermore, inert spacer molecules inserted between hapten and carrier do not interfere with carrier specificity in the antibody response3. Reflecting current views on the cooperation between thymus-derived (T) and bone marrow derived (B) lymphocytes in the antibody response to various antigens4, the second hypothesis invokes two or more cells, one with receptors directed towards the hapten (hapten-sensitive cell), the others specific for the carrier molecule proper (carrier-reactive cells). Supporting this is the observation that pre-immunization to a particular protein carrier alone could potentiate the primary or secondary antihapten response to a hapten conjugated to that protein5. In an adoptive transfer system, moreover, the efficiency of antihapten antibody production by cells primed to a particular hapten-protein conjugate and stimulated with the hapten conjugated on a heterologous protein, is significantly enhanced by the introduction of cells primed to the heterologous carrier alone. Anti-carrier serum antibody does not cause such enhancement6. The carrier-reactive cells must therefore cooperate in increasing the efficiency of the hapten-sensitive cells in some way other than by providing humoral anti-carrier antibody. While there is currently intense effort to examine the 13C signal of CO2 evolved in the dark, less is known on the isotope composition of day‐respired CO2. This lack of knowledge stems from technical difficulties to measure the pure respiratory isotopic signal: day respiration is mixed up with photorespiration, and there is no obvious way to separate photosynthetic fractionation (pure ci/ca effect) from respiratory effect (production of CO2 with a different δ13C value from that of net‐fixed CO2) at the ecosystem level. Here, we took advantage of new simple equations, and applied them to sunflower canopies grown under low and high [CO2]. We show that whole mesocosm‐respired CO2 is slightly 13C depleted in the light at the mesocosm level (by 0.2–0.8‰), while it is slightly 13C enriched in darkness (by 1.5–3.2‰). The turnover of the respiratory carbon pool after labelling appears similar in the light and in the dark, and accordingly, a hierarchical clustering analysis shows a close correlation between the 13C abundance in day‐ and night‐evolved CO2. We conclude that the carbon source for respiration is similar in the dark and in the light, but the metabolic pathways associated with CO2 production may change, thereby explaining the different 12C/13C respiratory fractionations in the light and in the dark.  相似文献

Abstract: The supposed vestige of a cephalopod gladius from Turonian platy limestones at Vallecillo, north‐east Mexico, named Palaeoctopus pelagicus by Fuchs et al. in 2008, is reinterpreted and shown to be a gular plate of a coelacanth fish, possibly of the genus Megacoelacanthus. In addition to the gular plate, two extrascapulars and fin rays of all fins are preserved on one slab and its counterpart. This is the first record of a coelacanth from these lower Turonian strata at Vallecillo, which are rich in fish.  相似文献

One of the most important changes in high‐latitude ecosystems in response to climatic warming may be the thawing of permafrost soil. In upland tundra, the thawing of ice‐rich permafrost can create localized surface subsidence called thermokarst, which may change the soil environment and influence ecosystem carbon release and uptake. We established an intermediate scale (a scale in between point chamber measurements and eddy covariance footprint) ecosystem carbon flux study in Alaskan tundra where permafrost thaw and thermokarst development had been occurring for several decades. The main goal of our study was to examine how dynamic ecosystem carbon fluxes [gross primary production (GPP), ecosystem respiration (Reco), and net ecosystem exchange (NEE)] relate to ecosystem variables that incorporate the structural and edaphic changes that co‐occur with permafrost thaw and thermokarst development. We then examined how these measured ecosystem carbon fluxes responded to upscaling. For both spatially extensive measurements made intermittently during the peak growing season and intensive measurements made over the entire growing season, ecosystem variables including degree of surface subsidence, thaw depth, and aboveground biomass were selected in a mixed model selection procedure as the ‘best’ predictors of GPP, Reco, and NEE. Variables left out of the model (often as a result of autocorrelation) included soil temperature, moisture, and normalized difference vegetation index. These results suggest that the structural changes (surface subsidence, thaw depth, aboveground biomass) that integrate multiple effects of permafrost thaw can be useful components of models used to estimate ecosystem carbon exchange across thermokarst affected landscapes.  相似文献

The retinal pigment epithelium (RPE) of teleosts contains pigment granules that migrate in response to changes in light condition. Dissociated, cultured RPE cells in vitro can be triggered to aggregate or disperse pigment granules by the application of cAMP or dopamine, respectively. Previous research using the actin‐disrupting drug, cytochalasin D, suggested that pigment granule motility is actin dependent. To further examine the role of actin in pigment granule motility, we tested the effects of the actin‐stabilizing drug, jasplakinolide, on pigment granule motility. Pigment granules in previously dispersed RPE cells remained dispersed after jasplakinolide exposure (0.1–1 μM), but the drug halted movement of most pigment granules and stimulated rapid bi‐directional movements in a small subset of granules. Jasplakinolide also blocked net pigment granule aggregation and interfered with the maintenance of full aggregation. Although jasplakinolide did not block pigment granule dispersion, it did alter the motility of dispersing granules compared to control cells; rather than the normal saltatory, primarily centrifugal movements, granules of jasplakinolide‐treated cells demonstrated slow, creeping centrifugal movements and more rapid bi‐directional movements. Jasplakinolide also altered cell morphology; the length and thickness of apical projections increased, and enlarged, paddle‐like structures, which contained F‐actin appeared at the tips of projections. Actin antibody labeling of jasplakinolide‐treated cells revealed a more reticulated network of actin compared to antibody‐labeled control cells. These results indicate that jasplakinolide‐induced disruption of the actin network compromises normal pigment granule dispersion and aggregation in isolated RPE cells, thus providing further evidence that these movements are actin dependent.  相似文献

Lycaenid butterflies of the Aricia agestisartaxerxes complex pose an unresolved taxonomic and conservation problem in northwestern Europe. Two key issues require resolution: (i) how many species of Aricia occur in northwestern Europe and what are their distributions?; (ii) how is the morphological variation observed in northwestern Europe best explained? We investigated phylogenetic relationships and phylogeographic patterns in this species group using mitochondrial and nuclear markers in comparison with morphological variation. A 325 bp fragment of the mitochondrial cytochromeb gene was sequenced from 179 individuals representing 18 populations from the UK and Scandinavia. Seventeen enzymecoding loci were analysed from 538 individuals from the same populations. Highly congruent phylogenies between mitochondrial and allozyme markers demonstrate that the sample is composed of two closely related species, A. agestis and A. artaxerxes. Both marker types also suggest that Scottish and northern Scandinavian A. artaxerxes populations are conspecific, and consequently do not support the endemic status of A. artaxerxes in the UK. The subspecies division of British populations of A. artaxerxes is also not supported by phylogenetic analyses. Allozyme and mitochondrial analyses cluster two populations from the Peak District, UK, differently. The former suggests that they are A. artaxerxes whilst the latter suggests they are A. agestis. Further research is required to find the reason for this disagreement, which could be associated with the different dynamics of nuclear and mitochondrial genes across a hybrid zone between the two species. © 2002 The Linnean Society of London, Biological Journal of the Linnean Society, 2002, 75 , 27–37.  相似文献

SUMMARY: Six QAC detergent mixtures and the QACs alone were tested for bactericidal efficiency in hard and distilled water, using E. coli as test organism, contact times of 1, 2 and 5 min and temperatures of 5, 10 and 17°.
The efficiencies of the unformulated QACs were dissimilar. With all, 200 p/m in hard water was less efficient than 50 p/m in distilled water. The efficiencies of the QAC detergent mixtures were not related to that of the pure QAC, the efficiency of a QAC being increased by one detergent formulation and decreased by another. Freezing and thawing may alter element turnover and solute fluxes in soils by changing physical and biological soil properties. We simulated soil frost in replicated snow removal plots in a mountainous Norway spruce stand in the Fichtelgebirge area, Germany, and investigated N net mineralization, solute concentrations and fluxes of dissolved organic carbon (DOC) and of mineral ions (NH4+, NO3, Na+, K+, Ca2+, Mg2+). At the snow removal plots the minimum soil temperature was −5 °C at 5 cm depth, while the control plots were covered by snow and experienced no soil frost. The soil frost lasted for about 3 months and penetrated the soil to about 15 cm depth. In the 3 months after thawing, the in situ N net mineralization in the forest floor and upper mineral soil was not affected by soil frost. In late summer, NO3 concentrations increased in forest floor percolates and soil solutions at 20 cm soil depth in the snow removal plots relative to the control. The increase lasted for about 2–4 months at a time of low seepage water fluxes. Soil frost did not affect DOC concentrations and radiocarbon signatures of DOC. No specific frost effect was observed for K+, Ca2+ and Mg2+ in soil solutions, however, the Na+ concentrations in the upper mineral soil increased. In the 12 months following snowmelt, the solute fluxes of N, DOC, and mineral ions were not influenced by the previous soil frost at any depth. Our experiment did not support the hypothesis that moderate soil frost triggers solute losses of N, DOC, and mineral ions from temperate forest soils.  相似文献
