Nitrate provision has been found to regulate the capacity forChara corallina cells to take up nitrate. When nitrate was suppliedto N sufficient cells maximum nitrate uptake was reached after8 h. Prolonged treatment of the cells in the absence of N alsoresulted in the apparent ability of these cells to take up nitrate.Chlorate was found to substitute partially for nitrate in the‘induction’ step. The effects on nitrate reductionwere separated from those on nitrate uptake by experiments usingtungstate. Tungstate pretreatment had no effect on NO3uptake ‘induced’ by N starvation, but inhibitedNO3 uptake associated with NO3 pretreatment. Chloridepretreatment similarly had no effect on NO3 uptake ‘induced’by N deprivation, but inhibited NO3 uptake followingNO3 pretreatment. The data suggest that there are atleast two mechanisms responsible for the ‘induction’of nitrate uptake by Chara cells, one associated with NO3reduction and ‘induced’ by CIO3 or NO3and one associated with N deprivation. ABSTRACT. Myrionecta rubra, a ubiquitous planktonic ciliate, has received much attention due to its wide distribution, occurrence as a red tide organism, and unusual cryptophyte endosymbiont. Although well studied in coastal waters, M. rubra is poorly examined in the open ocean. In the Irminger Basin, North Atlantic, the abundance of M. rubra was 0–5 cells/ml, which is low compared with that found in coastal areas. Distinct patchiness (100 km) was revealed by geostatistical analysis. Multiple regression indicated there was little relationship between M. rubra abundance and a number of environmental factors, with the exception of temperature and phytoplankton biomass, which influenced abundance in the spring. We also improve on studies that indicate distinct size classes of M. rubra; we statistically recognise four significantly distinct width classes (5–16, 12–23, 18–27, 21–33 μm), which decrease in abundance with increasing size. A multinomial logistic regression revealed the main variable correlated with this size distribution was ambient nitrate concentration. Finally, we propose a hypothesis for the distribution of sizes, involving nutrients, feeding, and dividing of the endosymbiont.  相似文献

One‐tailed statistical tests are often used in ecology, animal behaviour and in most other fields in the biological and social sciences. Here we review the frequency of their use in the 1989 and 2005 volumes of two journals (Animal Behaviour and Oecologia), their advantages and disadvantages, the extensive erroneous advice on them in both older and modern statistics texts and their utility in certain narrow areas of applied research. Of those articles with data sets susceptible to one‐tailed tests, at least 24% in Animal Behaviour and at least 13% in Oecologia used one‐tailed tests at least once. They were used 35% more frequently with nonparametric methods than with parametric ones and about twice as often in 1989 as in 2005. Debate in the psychological literature of the 1950s established the logical criterion that one‐tailed tests should be restricted to situations where there is interest only in results in one direction. ‘Interest’ should be defined; however, in terms of collective or societal interest and not by the individual investigator. By this ‘collective interest’ criterion, all uses of one‐tailed tests in the journals surveyed seem invalid. In his book Nonparametric Statistics, S. Siegel unrelentingly suggested the use of one‐tailed tests whenever the investigator predicts the direction of a result. That work has been a major proximate source of confusion on this issue, but so are most recent statistics textbooks. The utility of one‐tailed tests in research aimed at obtaining regulatory approval of new drugs and new pesticides is briefly described, to exemplify the narrow range of research situations where such tests can be appropriate. These situations are characterized by null hypotheses stating that the difference or effect size does not exceed, or is at least as great as, some ‘amount of practical interest’. One‐tailed tests rarely should be used for basic or applied research in ecology, animal behaviour or any other science.  相似文献

Bot. 37: 429–439. and have been used to investigate various properties of nitrate uptake and translocation intoHordeum vulgare L seedlings. Short term / influx into seed lings grown in CaSO4 was stimulated by after a lag of 2 h. The apparent kinetics of shortterm / influx over the concentration range 0?0–0?7mol m fitted Michaelis-Menten equations The apparent Vmax didnot change when seedlings were used that had been pretreatedin 10 or 100 mmol m–3 and Vmax=3.77 and 3?56µmol g–1 fr. wt. h–1respectively. The apparent Michaelis constants were also similarand Km=0?47 and 0?45 mol m–3 respectively. Longer term pulse chase experiments with the heavy isotope 15Nhave shown that feeding roots with resulted in the preferential appearance of 15N labelled aminoacids in the xylem sap. Pulse chase experiments with the radioisotope13N have shown that feeding shoots with resulted in a radial pattern of distnbution of labelin the leaf veins, which can be detected using autoradiography. Hemocyanins are high molecular weight oxygen-carrying proteinsthat occur in the molluscs and arthropods. The oxygen-bindingsite in these proteins is a pair of copper atoms bound directlyto ammo acid side chains. The biscopper sites of these proteinsbind single molecules of dioxygen or carbon monoxide. In arthropodsthere are two copper atoms per approximately 70 000 daltonsof protein. This corresponds to the molecular weight of theminimum polypeptide chain. In molluscs however there are twocopper atoms per 50,000 daltons of protein. This does not correspondto the minimum polypeptide chain although it does correspondto the minimal functional unit. The minimal polypeptide chainin molluscan hemocyanins is approximately 400,000 daltons andappears to be composed of eight or more 50 000 dalton unitslinked together like a string of pearls. In the molluscs, thenative hemocyanins found in the hemolymph generally occur asgiant cylindrical molecules 350 Å in diameter, 380 Ålong, with molecular weights of 9 000,000. These large moleculesare composed of approximately 20 polypeptide chains. In thehemolymph of the arthropods depending upon the species, hemocyaminsoccur as hexamers dodecamers 24-mers and 48-mers. The molecularweight of the 48-mers is about 3 600,000. The respiratory functionsof hemocyamns show a wide range of allosteric properties. Thelarge molecules commonly show cooperativity in oxygen binding.The number of interacting subunits is in some cases dependentupon external conditions of pH and ionic composition. The oxygenaffinity is usually sensitive to pH and to low molecular weightcofactors like chloride, calcium and magnesium ions. The intrinsicoxygen binding properties of an organism s hemocyanin and itsallosteric control by modulators allow organisms possessingthese giant molecules to adapt to their environmental conditions.  相似文献

Abstract. Nitrate uptake into Chara corallina cells at different external pH (pHo) after different NO3 pretreatment conditions has been investigated. Following NO3 pretreatment (0.2 mol m−3 NO3) there was little effect of pHo on subsequent net NO3 uptake into Chara cells. After N deprivation (2 mmol m−3 NO3) there was a pronounced effect of pHo on nitrate uptake, the maximum rate occurring at pHo 4.7. There was no consistent relationship between OH efflux and NO3 uptake in short term experiments (< 1 h). NO3 efflux was also sensitive to pHo, the maximum rate occurring at ∼ pHo 5.0. An inhibitor of the proton pump, DES, immediately stimulated NO3 uptake into cells pretreated with NO3 and prevented the time-dependent decrease in NO3, uptake into Chara cells that had been previously treated with low N (2 mmol m−3 NO3). NO3 efflux was found to be very sensitive to DES with Ki= 0.7 mmol m−3. At the optimum pHo for NO3 uptake the effect of DES on membrane potential (ψm) were slight, and only apparent after 30 min. The results are interpreted in context of current models relating NO3 uptake and H+ pump activity. A new model for NO3 uptake is proposed which involves NO3/NO3 exchange at steady state.  相似文献

The mature egg of Pteridium aquilinum is surrounded by an acetolysis-resistantmembrane. Cytochemical tests indicate that this consists oflipid, possibly in a polymerized form. There is no evidencefor the presence of polysaccharide, protein, lignin, suberin,or sporopollenin. The membrane is believed to be formed frommaterials secreted by the egg itself, and to be analogous tothe inner portion of the megaspore membrane of a gymnospermwhich has a comparable origin and staining properties.  相似文献

The goal of the present study was to obtain valuable microsatellite markers useful in genetic approaches of the shrimp Litopenaeus vannamei, an important aquaculture species of the world. Eight loci produced suitable polymorphic patterns with allele number ranging from two to 12 and expected heterozygosity from 0.18 to 0.89. Cross‐species amplification was successfully performed in five other penaeid species (Litopenaeus schmitti, Farfantepenaeus brasiliensis, Farfantepenaeus paulensis, Xiphopenaeus kroyeri and Rimapenaeus constrictus), indicating that some these loci can be useful in studies of other related species.  相似文献
