The arbuscular mycorrhizal (AM) symbiosis is widespread throughout the plant kingdom and important for plant nutrition and ecosystem functioning. Nonetheless, most terrestrial ecosystems also contain a considerable number of non‐mycorrhizal plants. The interaction of such non‐host plants with AM fungi (AMF) is still poorly understood. Here, in three complementary experiments, we investigated whether the non‐mycorrhizal plant Arabidopsis thaliana, the model organism for plant molecular biology and genetics, interacts with AMF. We grew A. thaliana alone or together with a mycorrhizal host species (either Trifolium pratense or Lolium multiflorum) in the presence or absence of the AMF Rhizophagus irregularis. Plants were grown in a dual‐compartment system with a hyphal mesh separating roots of A. thaliana from roots of the host species, avoiding direct root competition. The host plants in the system ensured the presence of an active AM fungal network. AM fungal networks caused growth depressions in A. thaliana of more than 50% which were not observed in the absence of host plants. Microscopy analyses revealed that R. irregularis supported by a host plant was capable of infecting A. thaliana root tissues (up to 43% of root length colonized), but no arbuscules were observed. The results reveal high susceptibility of A. thaliana to R. irregularis, suggesting that A. thaliana is a suitable model plant to study non‐host/AMF interactions and the biological basis of AM incompatibility.  相似文献

Bioenergy production is driving modifications to native plant species for use as novel biofuel crops. Key aims are to increase crop growth rates and to enhance conversion efficiency by reducing biomass recalcitrance to digestion. However, selection for these biofuel‐valuable traits has potential to compromise plant defenses and alter interactions with pests and pathogens. Insect‐vectored plant viruses are of particular concern because perennial crops have potential to serve as virus reservoirs that influence regional disease dynamics. In this study, we examined relationships between growth rates and biomass recalcitrance in five switchgrass (Panicum virgatum) populations, ranging from near‐wildtype to highly selected cultivars, in a common garden trial. We measured biomass accumulation rates and assayed foliage for acid detergent lignin, neutral detergent fiber, in vitro neutral detergent fiber digestibility and in vitro true dry matter digestibility. We then evaluated relationships between these traits and susceptibility to a widely distributed group of aphid‐transmitted Poaceae viruses (Luteoviridae: Barley and cereal yellow dwarf viruses, B/CYDVs). Virus infection rates and prevalence were assayed with RT‐PCR in the common garden, in greenhouse inoculation trials, and in previously established switchgrass stands across a 300‐km transect in Michigan, USA. Aphid host preferences were quantified in a series of arena host choice tests with field‐grown foliage. Contrary to expectations, biomass accumulation rates and foliar digestibility were not strongly linked in switchgrass populations we examined, and largely represented two different trait axes. Natural B/CYDV prevalence in established switchgrass stands ranged from 0% to 28%. In experiments, susceptibility varied notably among switchgrass populations and was more strongly predicted by potential biomass accumulation rates than by foliar digestibility; highly selected, productive cultivars were most virus‐susceptible and most preferred by aphids. Evaluation and mitigation of virus susceptibility of new biofuel crops is recommended to avert possible unintended consequences of biofuel production on regional pathogen dynamics.  相似文献

Disentangling the relative roles of biotic and abiotic forces influencing forest structure, function, and local community composition continues to be an important goal in ecology. Here, utilizing two forest surveys 20‐year apart from a Central American dry tropical forest, we assess the relative role of past disturbance and local climatic change in the form of increased drought in driving forest dynamics. We observe: (i) a net decrease in the number of trees; (ii) a decrease in total forest biomass by 7.7 Mg ha?1 but when calculated on subquadrat basis the biomass per unit area did not change indicating scale sensitivity of forest biomass measures; (iii) that the decrease in the number of stems occurred mainly in the smallest sizes, and in more moist and evergreen habitats; (iv) that there has been an increase in the proportion of trees that are deciduous, compound leaved and are canopy species, and a concomitant reduction in trees that are evergreen, simple‐leaved, and understory species. These changes are opposite to predictions based on recovery from disturbance, and have resulted in (v) a uniform multivariate shift from a more mesic to a more xeric forest. Together, our results show that over relatively short time scales, community composition and the functional dominance may be more responsive to climate change than recovery to past disturbances. Our findings point to the importance of assessing proportional changes in forest composition and not just changes in absolute numbers. Our findings are also consistent with the hypothesis that tropical tree species exhibit differential sensitivity to changes in precipitation. Predicted future decreases in rainfall may result in quick differential shifts in forest function, physiognomy, and species composition. Quantifying proportional functional composition offers a basis for a predictive framework for how the structure, and diversity of tropical forests will respond to global change.  相似文献

1. Sedimentary akinetes (resting stages) may represent significant potential inocula for nuisance blooms of cyanobacteria. We studied the effects of salinity and sediment source on the germination and subsequent growth of Anabaena flos‐aquae akinetes from a shallow, tidally influenced lake. 2. Surface sediments collected from littoral and open‐water sites were used as inocula to culture A. flos‐aquae akinetes in four salinities (0.1, 2.2, 4.4 and 6.5) over 22 days. Akinete germination and development was followed by counting developmental stages every second day. 3. Filament growth, but not akinete germination, was inhibited by salinity and there were significantly fewer filaments at 6.5 than at 0.1 and 2.2. Cultures inoculated with littoral sediment had more akinetes, germlings and filaments than those inoculated with open‐water sediment. 4. Sediment is a potential source of inocula for Anabaena blooms in the lake, which potentially could develop solely from this source because germination and subsequent filament growth do not depend on the existence of an initial pelagic Anabaena population.  相似文献

Thymus and Bursa Dependence of Lymphocyte Mitogenic Factor in the Chicken   总被引:5,自引:0,他引:5  
AMONG the soluble products generated during activation of rodent or human lymphocytes1, lymphocyte-stimulating (‘mitogenic’) factors are detected by their ability to accelerate DNA metabolism of freshly cultured allogeneic or syngeneic lymphocytes2–8. If such lymphocyte activation products function as mediators of cellular immunity1,9–11, their activities will be generated independently of the antibody-forming system. In the chicken, antigen-stimulation of sensitized lymphocytes in vitro involves both thymus (T-) and bursa-(B-)-dependent cells12–14. If antigen-induced lymphocyte transformations are generally mediated by lymphocyte mitogenic factors, both T-cells and B-cells should produce lymphocyte mitogenic factors when stimulated by specific antigen. We have therefore determined whether antigen-stimulated chicken lymphocytes generate a lymphocyte mitogenic factor and whether this response is affected by neonatal thymectomy or bursectomy.  相似文献

Molluscs have become increasingly popular in the study of centralneural mechanisms. More recently, there have been attempts torelate activity in central neurons with behavior in animalsof this phylum. The latter studies necessitate an understandingof the effectors of such behaviors. This requires not only informationabout the neuromuscular junction, but also an awareness of thecapabilities of the muscles themselves. Therefore, we have discussedsome structural and related functional characteristics of molluscanmuscle. We suggest that invertebrate mucles might be comparedon three scales: the amount of myofilament organization, theamount of vesicular specialization and organization, and theamount of paramyosin. We have considered some characteristicsof the widely-studied sustained contraction, known as "catch."Finally, we have discussed the neuromuscular junction—thetypes of junctions, the multiplicity of innervation, and someaspects of pharmacology. The results of such a study indicatedmany areas in which further research is essential before wecan understand behavior in terms of activity in the centralnervous system.  相似文献
