Long-chain fatty acids in germination-inhibiting animal-wastecomposts were identified by gas chromatography-mass spectrometryas myristic, palmitic, stearic, oleic, linoleic, and linolenicacids. These acids were found at concentrations greater than0.25 mg (g dry compost)–1. The identified acids, togetherwith lauric acid, and five kinds of short- and medium-chainfatty acid, were tested for their effects on the germinationprocess of sorghum seeds. The authentic long-chain fatty acids, which were dissolved ina 1:9 (v/v) mixture of methanol and distilled water at 40 mgliter–1, significantly reduced the -amylase activity,physiological water uptake, and ATP content of the germinatingseeds during the first 24h of imbibition, as well as the rateof germination of seeds. Among the tested fatty acids, myristicand palmitic acids were the most potent inhibitors of germination.The inhibitory effects of long-chain fatty acids were strongerthan those of the phenolic acids. The short- and medium-chainfatty acids did not have any significant germination-inhibitoryeffects at 40 mg liter–1. The results indicate that thelong-chain fatty acids are the dominant inhibitors of germinationin animal-waste composts, and that the inhibition of the -amylaseactivity in germinating sorghum seeds is one aspect of the modeof action of these long-chain fatty acids. 1On leave from the Department of Crop Science, Faculty of Agriculture,University of Peradeniya, Sri Lanka.  相似文献

Orthophosphate (H2PO4?, Pi) is an essential macronutrient integral to energy metabolism as well as a component of membrane lipids, nucleic acids, including ribosomal RNA, and therefore essential for protein synthesis. The Pi concentration in the solution of most soils worldwide is usually far too low for maximum growth of crops, including rice. This has prompted the massive use of inefficient, polluting, and nonrenewable phosphorus (P) fertilizers in agriculture. We urgently need alternative and more sustainable approaches to decrease agriculture's dependence on Pi fertilizers. These include manipulating crops by (a) enhancing the ability of their roots to acquire limiting Pi from the soil (i.e. increased P‐acquisition efficiency) and/or (b) increasing the total biomass/yield produced per molecule of Pi acquired from the soil (i.e. increased P‐use efficiency). Improved P‐use efficiency may be achieved by producing high‐yielding plants with lower P concentrations or by improving the remobilization of acquired P within the plant so as to maximize growth and biomass allocation to developing organs. Membrane lipid remodelling coupled with hydrolysis of RNA and smaller P‐esters in senescing organs fuels P remobilization in rice, the world's most important cereal crop.  相似文献

SG2NA, a WD40 repeat protein of the Striatin subfamily, has four splicing and one messenger RNA edit variants. It is fast emerging as a scaffold for multimeric signaling complexes with roles in tissue development and disease. The green fluorescent protein (GFP)‐tagged variants of SG2NA were ectopically expressed in NIH3T3 cells and their modulation by serum and GSK3β‐ERK signaling were monitored. The 87, 78, and 35 kDa variants showed a biphasic modulation by serum till 24 h but the 52 kDa variant remained largely unresponsive. Inhibition of phosphatases by okadaic acid increased the levels of the endogenous 78 kDa and the ectopically expressed GFP‐tagged 87 and 78 kDa SG2NAs. Contrastingly, okadaic acid treatment reduced the level of GFP‐tagged 35 kDa SG2NA, suggesting differential modes of their stability through phosphorylation‐dephosphorylation. The inhibition of GSK3β by LiCl showed a gradual decrease in the levels of 78 kDa. In the case of the other variants viz, GFP‐tagged 35, 52, and 87 kDa, inhibition of GSK3β caused an initial increase followed by a decrease with a subtle difference in kinetics and intensities. Similar results were also seen upon inhibition of GSK3β by small interfering RNA. All the variants showed an increase followed by a decrease upon inhibition of extracellular‐signal‐regulated‐kinase (ERK). These variants are localized in the plasma membrane, endoplasmic reticulum, mitochondria, and the nucleus with different propensities and no discernable subcellular distribution was seen upon stimulation by serum and the inhibition of phosphatases, GSK3β, and ERK. Taken together, the variants of SG2NA are modulated by the kinase‐phosphatase network in a similar but characteristic manner.  相似文献

Communal nesting lizards may be vulnerable to climate warming, particularly if air temperatures regulate nest temperatures. In southeastern Australia, velvet geckos Oedura lesueurii lay eggs communally inside rock crevices. We investigated whether increases in air temperatures could elevate nest temperatures, and if so, how this could influence hatching phenotypes, survival, and population dynamics. In natural nests, maximum daily air temperature influenced mean and maximum daily nest temperatures, implying that nest temperatures will increase under climate warming. To determine whether hotter nests influence hatchling phenotypes, we incubated eggs under two fluctuating temperature regimes to mimic current ‘cold’ nests (mean = 23.2 °C, range 10–33 °C) and future ‘hot’ nests (27.0 °C, 14–37 °C). ‘Hot’ incubation temperatures produced smaller hatchlings than did cold temperature incubation. We released individually marked hatchlings into the wild in 2014 and 2015, and monitored their survival over 10 months. In 2014 and 2015, hot‐incubated hatchlings had higher annual mortality (99%, 97%) than cold‐incubated (11%, 58%) or wild‐born hatchlings (78%, 22%). To determine future trajectories of velvet gecko populations under climate warming, we ran population viability analyses in Vortex and varied annual rates of hatchling mortality within the range 78– 96%. Hatchling mortality strongly influenced the probability of extinction and the mean time to extinction. When hatchling mortality was >86%, populations had a higher probability of extinction (PE: range 0.52– 1.0) with mean times to extinction of 18–44 years. Whether future changes in hatchling survival translate into reduced population viability will depend on the ability of females to modify their nest‐site choices. Over the period 1992–2015, females used the same communal nests annually, suggesting that there may be little plasticity in maternal nest‐site selection. The impacts of climate change may therefore be especially severe on communal nesting species, particularly if such species occupy thermally challenging environments.  相似文献

The abundance and diversity of chemotactic heterotrophic bacteria associated with Arctic cyanobacteria was determined. The viable numbers ranged between 104 and 106 cell g?1 cyanobacterial biomass. A total of 112 morphotypes, representing 22 phylotypes based on their 16S rRNA sequence similarity were isolated from the samples. All the phylotypes were Gram-negative with affiliation to the proteobacterial and bacteroidetes divisions. Among the 22 phylotypes, 14 were chemotactic to glucose. Majority of the phylotypes were psychrotolerant showing growth up to 30 °C. Representatives of Alphaproteobacteria, the genus Flavobacterium and the gammaproteobacterial Alcanivorax sp, were psychrophilic with growth at or below 18 °C. A significant percentage of phylotypes were pigmented (~68 %), rich in unsaturated membrane fatty acids and tolerated pH values and NaCl concentrations between 5.0–8.0 and 0.15–1.0 M, respectively. The percentages of phylotypes producing extracellular cold-active enzymes at 4 °C were amylase (18.18 %), lipase and urease (45.45 %), caseinase (59.09 %) and gelatinase (31.8 %).  相似文献

Major contribution (over 90 %) to the world’s rice production is coming from Asia, where metal contamination of agricultural lands is often reported. Thus the present paper reviews the sources and current status of heavy metal contamination of paddy lands in the region. Apart from the natural sources, agrochemicals, wastewater irrigation, sewage sludge application, livestock manures, mining and fly ash etc., could be identified as the key sources of metal contamination in Asia. Accumulation of heavy metals and metalloids (Cd, Zn, Cu, Pb, Cr, Ni, Fe, Zn, Co, Hg and As) in different parts of the rice plant (roots, straw, hull and grain) is reported at varying degrees. Rice grain accumulates the least amount of toxic metals compared to hull, straw and roots. Most importantly, a greater number of investigations confirmed that the metal contents in rice grains are within the permissible limits of Codex recommendations of joint Food and Agriculture Organization/World Health Organization Food Standards Programme and/or food regulations imposed by the respective governments. However, due to the fact that rice is a main route of human exposure to heavy metals, appropriate preventive and remedial measures should be enforced in the areas with potential risk of metal contamination.  相似文献
