A recessive allele of a gene in Tripsacum dactyloides L. (eastern gamagrass) changes staminate florets to pistillate or hermaphrodite, and restores fertility to suppressed florets. There were ten to 25 times more seeds in the mutant pistillate form, and these were 0.32 to 0.59 times smaller than seeds from the normal form. Seeds from pistillate plants had significantly lower germination rates (22% vs. 50%), and seedlings grew 20% slower than those of normal plants in a greenhouse experiment. Pistillate seedling survival rates were lower in both high- (18.8% vs. 62.6%) and low- (52.8% vs. 72.6%) competition environments in a field experiment, and surviving seedlings were smaller. The maternal parent of volunteer seedlings next to a plantation of normal and pistillate plants was determined by dissecting the attached fruitcases of 1,313 seedlings. Pistillate plants in the plantation produced 90% of all seeds falling on the site but only 29% of the volunteer seedlings. The pistillate macromutation is not likely to spread in the wild due to morphological constraints on seed size and packaging.  相似文献

Physiol.82(2): 460-468, 1997.Arterial vessel responses to sodiumnitroprusside (SNP) and acetylcholine (ACh) were measured in thespinotrapezius muscle of sedentary (Sed) and treadmill-trained (Tr)rats to determine whether these endothelium-dependent (ACh) and-independent (SNP) mechanisms contribute to thetraining-induced increase in functional vasodilation previouslyobserved. Control and maximal vessel diameters were similar between Sedand Tr. After 8 wk of training, functional dilation (2-, 4-, and 8-Hzcontractions) was enhanced in all orders of vessels studied[terminal feed artery (FA), largest arterioles (1A), andintermediate-sized arterioles (2A)], but responses to SNP wereincreased only in FA. Responses to ACh were not significantly increasedin any vessel order. After 16 wk of training, functional dilation hadregressed in Tr such that only the FA response to 4 Hz wassignificantly elevated relative to Sed. However, the FA and 1Aresponses to SNP were significantly greater in Tr than in Sed, as werethe 1A and 2A responses to ACh. These results show a dissociation offunctional dilation and SNP- or ACh-mediated responses, as well asage-dependent interactions, a time-dependent progression, and vesselorder specificity in the adaptations to training.

The synthesis of ovine hypothalamic somatostatin-28 (Ser-Ala-Asn-Ser-Asn-Pro-Ala-Met-Ala-Pro-Arg-Glu-Arg-Lys-Ala-Gly-Cys-Lys-Asn-Phe-Phe-Trp-Lys-Thr-Phe-Thr-Ser-Cys-OH) has been accomplished by solid phase methodology. The structure of the synthetic material was verified by: (1) direct sequence analysis with a Beckman 89°C sequencer, (2) correlation of the amino acid analyses of the isolated tryptic peptide fragments with their theoretical compositions, and (3) comparison, using high performance liquid chromatography, of the synthetic methionine-sulfoxide and methionine-sulfone modified NH2-terminal peptides (residues 1–11) with the corresponding tryptic fragment from somatostatin-28.  相似文献

A simple and reliable method is described for breeding spined loach, Cobitis taenia Linnaeus, 1758, under laboratory conditions. Three 40 L aquaria were each provided with a thick tuft of moss placed on top of a gauze-covered plastic box to serve as a spawning site. Ovoposition always occurred in the most densely available vegetation. The nonadhesive eggs fell through the gauze into the box. The spined loach is a fractional spawner releasing eggs in 14–18 batches at intervals of 2–23 (median 6) days. The total number of eggs per female was 2905–4282 during the reproductive season of 101–120 days. Yolks measured 1.14 ± 0.07 mm and eggs were 2.54 ± 0.22 mm in diameter. Total larval length was 5.03 ± 0.34 mm at hatching and 6.77 ± 0.34 mm at the beginning of exogenous feeding. The method allowed the rearing of numerous young for conservation measures and experimental investigations.  相似文献

Our recent studies have shown that extracellular-regulated protein kinase (ERK) promotes cell death in cerebellar granule neurons (CGN) cultured in low potassium. Here we report that the "death" phenotypes of CGN after potassium withdrawal are heterogeneous, allowing the distinction between plasma membrane (PM)-, DNA-, and PM/DNA-damaged populations. These damaged neurons display nuclear condensation that precedes PM or DNA damage. Inhibition of ERK activation either by U0126 or by dominant-negative mitogen-activated protein kinase/ERK kinase (MEK) overexpression results in a dramatic reduction of PM damaged neurons and nuclear condensation. In contrast, overexpression of constitutively active MEK potentiates PM damage and nuclear condensation. ERK-promoted cellular damage is independent of caspase-3. Persistent active ERK translocates to the nucleus, whereas caspase-3 remains in the cytoplasm. Antioxidants that reduced ERK activation and PM damage showed no effect on caspase-3 activation or DNA damage. These data identify ERK as an important executor of neuronal damage involving a caspase-3-independent mechanism.  相似文献

Livestock significantly affect wetland soils and vegetation but their impacts on wetland nutrient dynamics are poorly understood. We set up a full factorial laboratory experiment to assess the effects of Juncus effusus, grazing exclusion, and flooding on P flux from intact cores collected from seasonal wetlands in cattle pastures in south Florida. We collected intact cores from Juncus tussocks and plant interspaces inside and outside 4-year grazing exclosures in five replicate wetlands. We incubated the cores for 50 days under continuous flooding or weekly 1-day flooding cycles and measured P concentrations in surface and pore water. Grazing exclosures had less Juncus (17%) and bare ground (2%) than adjacent grazed areas (Juncus, 48%; bare ground, 12%), but did not affect P fluxes. Initial fluxes of soluble reactive P (SRP) were much higher in cores with Juncus (242 ± 153 mg P m−2 day−1) than without Juncus (14 ± 20 mg P m−2 day−1). In weekly flooded cores P fluxes fell to 19.7 ± 13.4 mg P m−2 day−1 in cores with and 2.7 ± 2.6 in cores without Juncus. The strong effect of Juncus on P flux was an indirect effect of cattle grazing, but 4 years of grazing exclusion did not have a significant effect on P fluxes.  相似文献

The effects of iontophoretically ejected angiotensin II (Ang II) on the firing rate of neurons in the basolateral complex and the central and cortical amygdala were investigated in two strains of urethane anesthetized rats. In normotensive Sprague-Dawley rats, Ang II induced a significant increase in the discharge rate of responsive amygdaloid neurons. In contrast, in the hypertensive transgenic [TGR(mREN-2)27] rats with higher brain Ang II level, Ang II more often caused inhibitory effects on the amygdaloid firing rate in comparison with controls. The distribution of nonresponsive, excited, and inhibited neurons differed significantly in the two rat strains. Moreover, the responsiveness of amygdaloid neurons was significantly higher in transgenic rats in comparison with controls. Both the increase and the decrease in the firing rate caused by Ang II could be blocked either by angiotensin AT(1) or by AT(2) receptor-specific antagonists. In many cases, the Ang II-induced decrease in the firing rate was antagonized by bicuculline, a gamma-aminobutyric acid (GABA(A)) antagonist. The higher responsiveness of amygdaloid neurons in transgenic rats as well as the predominance of inhibitory effects, presumedly mediated by GABAergic interneurons, could change the output of the amygdala and its influence on thirst, kidney, and cardiovascular function or on processes of learning and anxiety.  相似文献

Cloning and functional expression of a human heparanase gene.   总被引:32,自引:0,他引:32  
We have cloned a gene (HSE1) from a human placental cDNA library that encodes a novel protein exhibiting heparanase activity. The cDNA was identified through peptide sequences derived from purified heparanase isolated from human SK-HEP-1 hepatoma cells. HSE1 contains an open reading frame encoding a predicted polypeptide of 543 amino acids and possesses a putative signal sequence at its amino terminus. Northern blot analysis suggested strong expression of HSE1 in placenta and spleen. Transient transfection of HSE1 in COS7 cells resulted in the expression of a protein with an apparent molecular mass of 67-72 kDa. HSE1 protein was detectable in conditioned media but was also associated with the membrane fraction following cell lysis. The HSE1 gene product was shown to exhibit heparanase activity by specifically cleaving a labeled heparan sulfate substrate in a similar manner as purified native protein.  相似文献
