We describe a development of a novel high-throughput phagocytosis assay based on a pH-sensitive cyanine dye, CypHer5E, which is maximally fluorescent in an acidic environment. This dye is ideally suited for the study of phagocytosis because of the acidic conditions generated in the intracellular phagocytic vesicles after particle uptake. Use of CypHer5E-labeled particles results in greatly reduced background from noninternalized particles and makes the assay more robust. Additionally, CypHer5E-labeled particles are resistant to fluorescence quenching observed in the aggressive and acidic environment of the phagosome with traditional dyes. The CypHer5E-based assay has been shown to work reliably in a variety of cell types, including primary human monocytes, primary human dendritic cells, primary human endothelial cells, human monocytic THP-1 cell line, and human/mouse hybrid macrophage cell line WBC264-9C. Inhibition of CypHer5E bead uptake by cytochalasin D was studied, and the 50% inhibition concentration (IC50) was determined. The assay was performed in 96- and 384-well formats, and it is appropriate for high-throughput cellular screening of processes and compounds affecting phagocytosis. The CypHer5E phagocytosis assay is superior to existing protocols because it allows easy distinction of true phagocytosis from particle adherence and can be used in microscopy-based measurement of phagocytosis.  相似文献

Experiments on the transfection of cultured SKOV3 tumor cells of human ovarian adenocarcinoma and HeLa cells of human cervical carcinoma with gene Bax have demonstrated that SKOV3 cells are highly sensitive to the protein product of this gene, whereas the sensitivity of HeLa cells is substantially lower. HeLa cells obtained as a result of Bax transfection and subsequent selection are characterized by an extremely high Bax protein content and a hypersensitivity to doxorubicin. All Bax-transfected SKOV3 cells with an increased Bax content have died. In the SKOV3 cell line, a Bax exon 3 mutation has been found that corresponds to genotype G7/G9, whereas the native type of the Bax gene corresponds to genotype G8/G8. Analysis of the expression of genes encoding myelin basic protein (MBP) and glial fibrillary acidic protein (GFAP) in human glial tumors was carried out for determination of the expression specificity of these genes according to tumor types and their malignancy. Low levels of MBP mRNA in astrocytoma specimens of malignancy grades II-IV and significantly higher levels in perifocal zones adjacent to them have been determined by Northern hybridization. Diffuse astrocytomas and anaplastic astrocytomas are characterized mostly by a low level of MBP gene expression and high level of GFAP gene expression, but distinct subtypes of diffuse and anaplastic astrocytomas with a high level of GFAP gene expression can also be detected that may be the reflection of different oncogenic pathways. Very low levels or even absence of MBP mRNA were revealed in oligodendroglioma and all oligoastrocytomas. Thus, Northern hybridization data are correlated with serial analysis of gene expression (SAGE). Obtained results show that MBP is a nonspecific marker for tumors of oligodendroglial origin, but determination of relative levels of MBP and GFAP mRNAs may be useful for glial tumor recognition. In such a way, these two genes together with YKL-40 and TSC-22, which we found previously, can be included into the gene panel for determination of so-called “gene signatures” of brain tumors. However, strict requirements in relation to a clinical value of these “gene signatures” cannot be formulated without verifying them on a large number of clinical samples of tumors and valid control.  相似文献

Geochemical, biogeochemical, and molecular genetic investigation of the upper (0–5 cm) bottom sediments of the Yamal sector of the Kara Sea was carried out. The Yamal sector is well-protected from the massive inflow of river water. The sediments were oxidized at the surface and weakly reduced in the 3?5-cm layer. Corg content varied from 0.1 to 1.3%, while the level of dissolved СН4 was 1.9 to 20.3 μmol L–1. The isotopic composition of organic matter (OM) carbon, δ13Corg, varied from–27.5 to–22.2‰ (–25.4‰ on average). The share of terrigenous OM was 13.3 to 72.2% (48.9% on average). The rate of methane production, methane oxidation, and sulfate reduction varied from 0.8 to 9.0 (2.7 on average) nmol СН4 dm–3 day–1, from 9.9 to 103 (31.6 on average) nmol СН4 dm–3 day–1, and from 0.49 to 2.2 (1.1 on average) μmol S dm–3 day–1, respectively. High-throughput sequencing of the amplicons of the 16S rRNA genes was used to reveal the physiological groups of microorganisms responsible for the processes of methane production and oxidation, sulfate reduction, and oxidation of reduced sulfur compounds. Members of the phylum Woesearchaeota were predominant among archaea. Methanogenic archaea belonged to the families Methanobacteriaceae, Methanococcaceae, and Methanosarcinaceae (Euryarchaeota). Methanotrophs of the family Methylococcaceae were revealed among the Gammaproteobacteria, with their share in the sediments ~1%. In the class Deltaproteobacteria (15.4%), three orders of sulfate reducers were predominant: Desulfobacterales, Desulfovibrionales, and Desulfuromonadales. Oxidation of reduced sulfur compounds was carried out by chemolithoautotrophic bacteria of the genera Sulfurovum, Sulfurimonas, and Arcobacter of the class Epsilonproteobacteria (1.1% of the total microbial number).  相似文献

In the l = 3 Uragan-3M (U-3M) torsatron with a helical divertor the plasma is produced and heated by RF field in the ω ≤ ω ci range of frequencies. A two-temperature ion perpendicular energy distribution with a suprathermal tail sets in with heating. If the heating power is high enough, a spontaneous transition to an H-like confinement mode is observed. Recently, it has been supposed that the transition is connected with hotter and suprathermal ions (common name “fast ions“, FI) loss. The objective of this work is an experimental elucidation of the real link between the H-transition and FI loss. To do this, a transient regime of the RF discharge with two H-mode states is chosen, and the evolution is followed of electron density, FI content in the confinement volume, FI outflow to the divertor and edge potential. On the basis of juxtaposing of these processes, a conclusion is made that the H-mode transition in U-3M is really driven by FI loss. Possible mechanisms resulting in the transition are discussed, among them the ion orbit loss and the radial drift of helically-trapped ion orbits seem most probable.  相似文献

Dynamics of the composition of the microbial community was studied during start-up of a single-stage completely mixed constant flow laboratory setup for ammonium removal by the nitritation/anammox process from the filtrate of digested sludge of the Kuryanovo wastewater treatment plant (KWTP), Moscow. To decrease the period of the start-up, the setup was initially inoculated with two types of activated sludge (nitrifying sludge from a KWTP aeration tank and sludge from a sequencing batch reactor enriched with anammox bacteria). The start-up and adjustment stage was therefore decreased to 35–40 days, and nitrogen removal efficiency reached 80% after 120 days of the setup operation. Taxonomic analysis of the composition of the microbial community was carried out by pyrosequencing of the 16S rRNA fragments obtained using the universal and planctomycetes-specific primers. In the course of adaptation of activated sludge to increasing nitrogen load, microbial community of the setup became less diverse and more specialized. The contribution of anammox bacteria of the family Brocadiaceae, closely related to Candidatus “Brocadia caroliniensis,” increased gradually. Members of the order Nitrosomonadales were involved in ammonium oxidation to nitrite. While nitrite-oxidizing bacteria of the genus Nitrospira were also detected, their share decreased with accumulation of the activated sludge. The contribution of other bacteria varied as well: the shares of the phyla Ignavibacteria, Chloroflexi, and Acidobacteria increased significantly (up to 13, 12, and 10%, respectively of the total number of reads), while relative abundance of the Proteobacteria, Bacteroidetes, Actinobacteria, Firmicutes, Synergistetes, Aminicenantes, Thermotogae, and Cloacimonetes decreased. Thus, application of pyrosequencing made it possible to monitor succession of the bacterial community involved in nitrogen removal by nitritation/anammox process.  相似文献
