Spectrally Tunable Optical Transmission of Titanium Nitride Split Ring Resonators

In this paper, the optical properties of titanium nitride split ring resonators as an intermetallic metamaterial nanostructure were studied. Our simulation shows the presence of plasmon and LC resonances in the transmission spectrum of a cell consists of four u-shape split ring resonators. The effect of different parameters of resonator such as size, periodic constant, and the material between arms in addition to the polarization of incident beam was examined on the resonance behavior of the system. Also, the optical properties of a cell consist of four complementary split ring resonators within titanium nitride thin film were investigated. An excited mode was observed at λ = 840 nm that was attributed to the plasmon resonance. Changing the arrangement and configuration of the system from C _1v to C _2v symmetry led to the presence of the LC mode beside the plasmon mode in the transmission spectrum. Also, we explored a connection between the complementary split ring resonators and orderly perforated surface plasmon systems. It was determined that a transition occurred from resonator-type to surface plasmon behavior by increasing the size of resonator above 170 nm.     

Recombinant expression and purification of human placental growth factor 1 and specific camel heavy chain polyclonal antibody preparation

Placental growth factor (PlGF) is a member of the vascular endothelial growth factor (VEGF) family. Unlike VEGF, PlGF is dispensable for normal cell development as well as playing various roles in pathological angiogenesis which occurs in tissue ischemia, inflammation, and malignancy. The PlGF-1 has been considered as a potential candidate for the diagnosis and targeting of pathological angiogenesis. Camelidae serum contains an important fraction of functional antibodies, called heavy-chain antibodies (HcAbs) that are naturally devoid of light chains. Camelid HcAbs recognize their cognate antigens by a single variable-domain, referred to as VHH or Nanobody.Here, we describe the expression and purification of recombinant human PlGF-1 (rhPlGF-1). This protein was subsequently used for the preparation of camel heavy chain polyclonal antibody against rhPlGF-1.The recombinant expression plasmid pET-26b-hPlGF-1 was introduced into Escherichia coli BL21 cells to express the rhPlGF-1 protein. Purified rhPlGF-1 was used to immunize camel, the specific reactivity of HcAb was determined with ELISA and western blot. Western blot analysis indicated that the antiserum specifically reacted to the recombinant protein. The rhPlGF-1 protein and its antibody may be used for the development of detection assays needed for clinical research.     

Development of a novel engineered antibody targeting human CD123

Antibody engineering involves a range of custom modifications of immunoglobulins to improve their affinity, valency, and pharmacokinetics, ensuring a better target therapy achievement. A number of therapeutic antibodies have been used for cell surface receptor blockage, interfering with the ligand binding and inhibiting receptor-driven activation of cells. Here we describe the construction and characterization of a recombinant bivalent single-chain Fv (biscFv) that targets CD123. On conversion of anti-CD123 scFv to biscFv format, the recognition of the cognate ligand is not altered. Moreover, the increased overall efficacy of the anti-CD123 biscFv in binding and inhibition of CD123/IL-3 (interleukin-3) interactions in TF-1 cells is demonstrated.
