Low molecular mass proteoglycans (PG) were isolated from human articular cartilage and from pig laryngeal cartilage, which contained protein cores of similar size (Mr 40-44 kDa). However, the PG from human articular cartilage contained dermatan sulphate (DS) chains (50% chondroitinase AC resistant), whereas chains from pig laryngeal PG were longer and contained only chondroitin sulphate (CS). Disaccharide analysis after chondroitinase ABC digestion showed that the human DS-PG contained more 6-sulphated residues (34%) than the pig CS-PG (6%) and both contained fewer 6-sulphated residues than the corresponding high Mr aggregating CS-PGs from these tissues (86% and 20% from human and pig respectively). Cross-reaction of both proteoglycans with antibodies to bovine bone and skin DS-PG-II and human fibroblasts DS-PG suggested that the isolated proteoglycans were the humans DS-PG-II and pigs CS-PG-II homologues of the cloned and sequenced bovine proteoglycan. Polyclonal antibodies raised against the pig CS-PG-II were shown to cross-react with human DS-PG-II. SDS/polyacrylamide-gel analysis and immunoblotting of pig and human cartilage extracts showed that some free core protein was present in the tissues in addition to the intact proteoglycan. The antibodies were used in a competitive radioimmunoassay to determine the content of this low Mr proteoglycan in human cartilage extracts. Analysis of samples from 5-80 year-old humans showed highest content (approximately 4 mg/g wet wt.) in those from 15-25 year-olds and lower content (approximately 1 mg/g wet wt.) in older tissue (greater than 55 years). These changes in content may be related to the deposition and maintenance of the collagen fibre network with which this class of small proteoglycan has been shown to interact.  相似文献

Aeromonas aminopeptidase contains two nonidentical metal binding sites that have been shown by both spectroscopy and kinetics to be capable of interacting with one another [Prescott, J.M., Wagner, F.W., Holmquist, B., & Vallee, B.L. (1985) Biochemistry 24, 5350-5356]. The effects of metal ion substitutions on the susceptibility of the p-nitroanilides of L-alanine, L-valine, and L-leucine--substrates that are hydrolyzed at widely differing rates by native Aeromonas aminopeptidase--were studied by determining values of kcat and Km for the 16 metalloenzymes that result from all possible combinations of Zn2+, Co2+, Ni2+, and Cu2+ in each of the two sites. The different combinations of metal ions and substrates yield a broad range in kinetic values; kcat varies by more than 1800-fold, Km by 3000-fold, and kcat/Km ratios by more than 10,000. L-Leucine-p-nitroanilide is by far the most susceptible of the three substrates, and the hyperactivation previously observed with aminopeptidase containing either Ni2+ or Cu2+ in the first binding site and Zn2+ in the second site occurs only with the two poorer substrates, L-alanine-p-nitroanilide and L-valine-p-nitroanilide. Although the enzyme with Zn2+ in both sites hydrolyzes the substrates with N-terminal alanine and valine poorly, it is extremely effective toward L-leucine-p-nitroanilide. Neither metal binding site can be identified as controlling either Km or kcat; both parameters are influenced by the identity of the metal ions, by the site each occupies, and, most strongly, by the substrate.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

A cold-sensitive, streptomycin-sensitive mutant of Saccharomyces cerevisiae accumulates a 28S ribonucleoprotein particle when grown at low temperature. This particle contains 17S ribosomal ribonculeic acid which is degraded when exposed to ribonuclease. The particle does not serve as a precursor to 60 and 40S ribosomal subunits nor is it turned over when growth is allowed to resume at the permissive temperature; rather it is only diluted by growth. That streptomycin sensitivity (allelic with cold sensitivity) is ribosomal is evidenced by the inhibition of protein synthesis in vitro by streptomycin and the binding of labeled streptomycin to the mutant but not the parental 40S ribosomal subunit.  相似文献

Genetic variation of apoA1/C3/A4 is associated with hyperlipidaemia and coronary heart disease. We report the polymerase chain reaction (PCR) conditions for determining three polymorphic sites in the 5 flanking region of apoA1 using DNA prepared from small aliquots of whole blood. Analysis of the purified proteoglycans extracted from normal human articular cartilage with 4M-guanidinium chloride showed that there was an age-related increase in their content of protein and keratan sulphate. 2. The hydrodynamic size of the dissociated proteoglycans also decreased with advancing age, but there was little change in the proportion that could aggregate. 3. Results suggested that some extracts of aged-human cartilage had an increased content of hyaluronic acid compared with specimens from younger patients. 4. Dissociated proteoglycans, from cartilage of all age groups, bind to hyaluronic acid and form aggregates in direct proportion to the hyaluronic acid concentration. 5. Electrophoretic heterogeneity of the dissociated proteoglycans was demonstrated on polyacrylamide/agarose gels. The number of proteoglycan species observed was also dependent on the age of the patient.  相似文献

Summary In South Australian mangroves Lepsiella vinosa feeds on two species of barnacles, Balanus amphitrite and Elminius modestus, and only attacks other types of prey if no barnacles are available. Laboratory experiments indicate that L. vinosa can be trained to either species and switches in terms of Murdoch's (1969) model.The distribution of barnacles is heterogeneous and distinct prey patches are found in the mangroves. In high density patches, that is over 25 B. amphitrite per metre length of pneumatophore, L. vinosa concentrated its attacks on B. amphitrite irrespective of the relative abundance of the two species. In moderate density patches, that is less than 25 B. amphitrite per metre length of pneumatophore, L. vinosa fed on the two species as expected on the basis of relative abundance. There was, however, also an association between changes in the absolute density of B. amphitrite and the proportion it formed in the diet.Prey selection in the moderate density patches was not random and was influenced by previous feeding history. L. vinosa which fed on the more abundant species, E. modestus, selected this species at a higher rate than expected in subsequent surveys. This result suggests that ingestive conditioning took place in the field, and provides some support for the switching hypothesis.Field cage experiments indicate that L. vinosa shows switching provided that the density of the preferred prey was not too high. L. vinosa had a weak preference for B. amphitrite over E. modestus, c was calculated to be 1.26, when equal numbers of each species were present. The preference appears to be consistent between individual L. vinosa as few individuals fed predominantly on only one prey species.The switching response in L. vinosa was asymmetrical with regard to the two prey species. L. vinosa switched to B. amphitrite when it was more abundant irrespective of prior training. L. vinosa trained to E. modestus, or unfed, prior to the experiment switched to E. modestus when it was more abundant, but, those trained to B. amphitrite did not. L. vinosa also showed asymmetry in its feeding rate. Those trained to E. modestus, or unfed, attacked prey at the average rate observed in the field irrespective of the prey presented. L. vinosa trained to B. amphitrite showed a marked reduction in feeding rate when B. amphitrite was less abundant.Absolute density as well as the relative density of the two prey species influenced prey selection. Higher predation on B. amphitrite was found, despite a fixed relative abundance of one B. amphitrite to five E. modestus, when the barnacle density was increased from 11.2 to 56 per metre of pneumatophore.  相似文献
