异戊二烯是橡胶合成的重要前体物质。为了提高菌株的异戊二烯产量,本实验室在研究中构建了一株异戊二烯产气的菌株BW-01,基于蛋白质预算理论的指导,理性设计通过改变质粒拷贝数、增加稀有密码子等合成生物学手段调控关键限速酶编码基因表达,从而提高大肠杆菌外源MVA代谢途径的异戊二烯产量。摇瓶发酵实验中我们构建的新产气菌株BW-07比原有的产气菌株BW-01的产量提高了73%,达到了761.1 mg/L。为后续菌株改造及进行发酵罐实验奠定了基础。  相似文献

Background/Aims

Many anti-fibrotic drugs with high in vitro efficacies fail to produce significant effects in vivo. The aim of this work is to use a statistical approach to design a numerical predictor that correlates better with in vivo outcomes.

Methods

High-content analysis (HCA) was performed with 49 drugs on hepatic stellate cells (HSCs) LX-2 stained with 10 fibrotic markers. ∼0.3 billion feature values from all cells in >150,000 images were quantified to reflect the drug effects. A systematic literature search on the in vivo effects of all 49 drugs on hepatofibrotic rats yields 28 papers with histological scores. The in vivo and in vitro datasets were used to compute a single efficacy predictor (Epredict).

Results

We used in vivo data from one context (CCl4 rats with drug treatments) to optimize the computation of Epredict. This optimized relationship was independently validated using in vivo data from two different contexts (treatment of DMN rats and prevention of CCl4 induction). A linear in vitro-in vivo correlation was consistently observed in all the three contexts. We used Epredict values to cluster drugs according to efficacy; and found that high-efficacy drugs tended to target proliferation, apoptosis and contractility of HSCs.

Conclusions

The Epredict statistic, based on a prioritized combination of in vitro features, provides a better correlation between in vitro and in vivo drug response than any of the traditional in vitro markers considered.  相似文献

基因修饰技术是用于基因组定点改造的分子工具,目前主要有锌指核酸酶(ZFN)技术、转录激活子样效应物核酸酶(TALEN)技术和CRISPR-Cas核酸酶(CRISPR-Cas)技术。这些核酸酶都可以在DNA靶位点产生双链断裂(DSB),诱发细胞内源性的修复机制,激活体内非同源末端连接(NHEJ)或同源重组(HR)两种不同的修复机制,从而实现内源基因的敲除或外源基因的定点敲入。近年来,基因修饰技术已成功应用到细菌、酵母、人类细胞、果蝇、斑马鱼、小鼠、大鼠、家畜、食蟹猴、拟南芥、水稻、烟草、玉米、高粱、小麦和大麦等多种生物,显示了其强大的基因编辑优势。特别是新近出现的CRISPR-Cas9技术,降低了成本,使基因编辑变得简洁、高效和易于操作,得到了很多研究人员的关注。本文系统介绍了以上3种技术的原理及最新研究进展,并对未来的研究和应用做出了展望。  相似文献

The aggregate luminescence behaviors of polymeric luminescent materials have been attracting great attention. However, the importance of the polymerization process on luminescence, namely, polymerization-induced emission (PIE), has rarely been overviewed. In this review, recent advances in polymerization with PIE effects are summarized, including PIE with aromatic rings based on one-/two-/multi-component polymerizations, and PIE without aromatic rings according to disparate mechanisms of polymerizations. Typical examples are selected to elaborate the basic design principles, as well as the properties and potential applications of the luminous polymers. Moreover, the challenges and perspectives in this area are also discussed.  相似文献

Activated by retinoids, metabolites of vitamin A, the retinoic acid receptors (RARs) and the retinoid X receptors (RXRs) play important roles in a wide variety of biological processes, including embryo development, homeostasis, cell proliferation, differentiation and death. In this review, we summarized the functional roles of nuclear receptor RAR/RXR heterodimers in liver physiology. Specifically, RAR/RXR modulate the synthesis and metabolism of lipids and bile acids in hepatocytes, regulate cholesterol transport in macrophages, and repress fibrogenesis in hepatic stellate cells. We have also listed the specific genes that carry these functions and how RAR/RXR regulate their expression in liver cells, providing a mechanistic view of their roles in liver physiology. Meanwhile, we pointed out many questions regarding the detailed signaling of RAR/RXR in regulating the expression of liver genes, and hope future studies will address these issues.  相似文献

[目的] 以秸秆等木质纤维素类生物质为原料生产液体生物燃料乙醇,目前生产成本高,大规模工业化生产尚有较大难度。构建能同化阿拉伯糖进行木糖还原生产木糖醇的重组酿酒酵母菌株,以实现原料中全糖利用、生产高附加值产品,实现产品多元化。[方法] 首先,利用CRISPR/Cas9基因编辑技术依次向出发菌株中导入阿拉伯糖代谢途径和木糖还原酶基因,使菌株获得代谢阿拉伯糖和将木糖转化为木糖醇的能力;其次,通过适应性驯化的进化工程手段,提高重组菌株对阿拉伯糖的利用效率;最后,通过混合糖发酵验证重组菌株利用阿拉伯糖和还原木糖产木糖醇的能力。[结果] 通过导入植物乳杆菌的阿拉伯糖代谢途径,酿酒酵母菌株获得了较好的利用阿拉伯糖生长繁殖的能力;进一步导入假丝酵母的木糖还原酶基因后,重组菌株在葡萄糖作为辅助碳源条件下可高效还原木糖产木糖醇,但阿拉伯糖的利用能力下降。利用以阿拉伯糖为唯一碳源的培养基进行反复批次驯化,阿拉伯糖的利用能力得以恢复和提升,得到表型较好的重组菌株KAX3-2。该菌株在木糖(50 g/L)和阿拉伯糖(20 g/L)混合糖发酵条件下发酵72 h时,对阿拉伯糖和木糖利用率分别达到42.1%和65.9%,木糖醇的收率为64%。[结论] 本研究成功构建了一株能有效利用阿拉伯糖并能将木糖转化为木糖醇的重组酿酒酵母菌株KAX3-2,为后续构建、获得阿拉伯糖代谢能力更强、木糖醇积累效率更高菌株的工作奠定了基础。  相似文献

生态地貌学作为一个“新兴”的跨学科的概念,受到地貌学家、生态学家及政策制定者的日益关注。地貌在控制许多生态系统过程中起着基础性的作用,而生态系统又可以对许多地貌形态和地貌过程产生深远的影响。过去几十年,在地貌学和生态系统生态学的交叉点上开展了大量的研究。通过梳理地貌学和生态系统生态学相似的发展历程,讨论了两个学科相结合的必要性;归纳了两者相结合的研究现状;总结了两个学科融合研究过程中存在的问题;提出未来需要进一步加强对地貌过程与生态过程相互作用的研究,并将生态地貌学与现行生态保护与生态修复建设及相关政策有机结合,促进生态地貌学的有序和可持续发展。正确认识地球的地貌形态与生态系统之间的关系,有助于拓展地貌学与生态系统生态学的研究尺度,促进学科体系发展;深入研究生态地貌学,对于维持生态平衡,保护生态多样性及增强未来对生态系统的管理和恢复都有重要意义。  相似文献

Long non‐coding RNAs (lncRNAs) have been indicated for the regulatory roles in cardiovascular diseases. This study determined the expression of lncRNA TNK2 antisense RNA 1 (TNK2‐AS1) in oxidized low‐density lipoprotein (ox‐LDL)‐stimulated human aortic smooth muscle cells (HASMCs) and examined the mechanistic role of TNK2‐AS1 in the proliferation and migration of HASMCs. Our results demonstrated that ox‐LDL promoted HASMC proliferation and migration, and the enhanced proliferation and migration in ox‐LDL‐treated HASMCs were accompanied by the up‐regulation of TNK2‐AS1. In vitro functional studies showed that TNK2‐AS1 knockdown suppressed cell proliferation and migration of ox‐LDL‐stimulated HASMCs, while TNK2‐AS1 overexpression enhanced HASMC proliferation and migration. Additionally, TNK2‐AS1 inversely regulated miR‐150‐5p expression via acting as a competing endogenous RNA (ceRNA), and the enhanced effects of TNK2‐AS1 overexpression on HASMC proliferation and migration were attenuated by miR‐150‐5p overexpression. Moreover, miR‐150‐5p could target the 3’ untranslated regions of vascular endothelial growth factor A (VEGFA) and fibroblast growth factor 1 (FGF1) to regulate FGF1 and VEGFA expression in HASMCs, and the inhibitory effects of miR‐150‐5p overexpression in ox‐LDL‐stimulated HASMCs were attenuated by enforced expression of VEGFA and FGF1. Enforced expression of VEGFA and FGF1 also partially restored the suppressed cell proliferation and migration induced by TNK2‐AS1 knockdown in ox‐LDL‐stimulated HASMCs, while the enhanced effects of TNK2‐AS1 overexpression on HASMC proliferation and migration were attenuated by the knockdown of VEGFA and FGF1. Collectively, our findings showed that TNK2‐AS1 exerted its action in ox‐LDL‐stimulated HASMCs via regulating VEGFA and FGF1 expression by acting as a ceRNA for miR‐150‐5p.  相似文献

The addition of surfactant in tablet was a well-defined approach to improve drug dissolution rate. While the selected surfactant played a vital role in improving the wettability of tablet by medium, it was equally important to improve the dissolution rate by permeation effect due to production of pores or the reduced inter-particle adhesion. Furthermore, understanding the mechanism of dissolution rate increased was significant. In this work, contact angle measurement was taken up as an alternative approach for understanding the dissolution rate enhancement for tablet containing surfactant. Ethylcellulose, as a substrate, was used to prepare tablet. Four surfactants, sodium dodecyl sulfate (SDS), sodium dodecylbenzenesulfonate (SDBS), dodecyltrimethylammonium bromide (DTAB), and sodium lauryl sulfonate (SLS), were used. Berberine hydrochloride, metformin hydrochloride, and rutin were selected as model drugs. The contact angle of tablet in the absence and presence of surfactant was measured to explore the mechanism. The dissolution test was investigated to verify the mechanism and to establish a correlation with the contact angle. The result showed that the mechanism was the penetration effect rather than the wetting effect. The dissolution increased with a reduction in the contact angle. DTAB was found to obtain the highest level of dissolution enhancement and the lowest contact angle, while SDS, SDBS, and SLS were found to be the less effective in both dissolution enhancement and contact angle decrease. Therefore, contact angle was a good indicator for dissolution behavior besides exploring the mechanism of increased dissolution, which shows great potential in formula screening.  相似文献
