As in cotyledons of Agrostemma githago, synthesis of DNA takesplace after germination in cotyledons of Vaccaria pyramidataand is followed by the formation of hydrolases, in particular,-amylase and acid phosphatase. If DNA synthesis is inhibitedby hydroxyurea, no, or only slight, enzyme activity develops.The possible role of this DNA synthesis is discussed. A method was developed to produce radiolabeled 3-hydroxy-L-kynurenine by injection of [14C]-L-tryptophan into pupae of the heliconid butterfly, Heliconius charitonia, which was converted into [14C]-3-hydroxy-L-kynurenine and deposited as a wing pigment. Extractions of 3-hydroxykynurenine (3-OHK) with 60% methanol from wings yielded in 14.4 μg per mg dry weight. In extracts from yellow wing areas, 3-OHK represented 100% of detectable amino acids. Resulting specific radioactivity of [14C]-3-OHK was between 0.05 and 0.07 mCi/mmol when 0.5 μCi [14C]-tryptophan was injected into pupae 1 or 2 days before emergence of the butterfly. Incorporation of [14C]-3-OHK into wing ommochromes was studied in nymphalid butterflies, Araschnia levana and Precis coenia. After injection into pupae [I4C]-3-OHK as well as [14C]-tryptophan were specifically incorporated into red and red-brown wing scales as shown by autoradiography. The same incorporation occurred in isolated wings after incubation in Grace's medium containing [14C]-3-OHK. In Araschnia levana, [14C]-3-OHK offered to left wing pairs was incorporated into dihydroxanthommatin six times more effectively than [14C]-tryptophan offered to right wing pairs from the same specimen. Therefore, 3-OHK seems to be the ultimate precursor of wing ommatins.  相似文献

1. Invasions of zebra and quagga mussels have had long‐term, large‐scale impacts on lake ecosystems in the USA as characterised by high abundance, broad‐scale spread and effective adaption to new environmental conditions. Due to their high filtering capacity, decreases in chlorophyll a (Chl) and total phosphorus (TP) concentrations have been reported in many affected lakes. 2. In 25 US lakes, we analysed the effects of dreissenid invasions on changes in Chl and TP concentrations, measured as the probability of a concentration decrease in the post‐invasion period and changes in Chl–TP relationships using Bayesian hierarchical regressions. We also examined whether changes in Chl and TP concentrations and in the Chl–TP relationship differed according to lake hydromorphology, such as mean depth or mixing status (mixed versus stratified lakes). 3. Our results showed that dreissenid invasions were often, but not always, associated with subsequent TP and Chl declines. Dreissenid effects on Chl and TP may be influenced by lake thermal structure. Decreases in Chl and TP were consistently found in mixed lakes where benthic–pelagic coupling is tight, while the effects were less predictable in stratified lakes. Within stratified lakes, Chl and TP reductions were more clearly discernible in deeper lakes with long water residence times. 4. Regression results demonstrated that a joint increase in slope and decrease in intercept and a tighter correlation of the Chl–TP relationship were likely to occur in dreissenid‐invaded lakes; this does not support the idea of a shift from bottom‐up to top‐down control of primary production. These results have important implications for management, suggesting that a relaxation of TP standards would be unwarranted. 5. Across lakes, the slope of the Chl–TP relationship for mixed lakes was substantially higher than that for stratified lakes before mussel invasion, indicating an important role of light in limiting primary production. The slope differences between mixed and stratified lakes decreased in the post‐invasion period, possibly because mussel filtration results in a relaxation of light limitation that is more pronounced in deeper, stratified lakes.  相似文献

1. The world's population is increasingly urban, and streams and rivers, as the low lying points of the landscape, are especially sensitive to and profoundly impacted by the changes associated with urbanization and suburbanization of catchments. 2. River restoration is an increasingly popular management strategy for improving the physical and ecological conditions of degraded urban streams. In urban catchments, management activities as diverse as stormwater management, bank stabilisation, channel reconfiguration and riparian replanting may be described as river restoration projects. 3. Restoration in urban streams is both more expensive and more difficult than restoration in less densely populated catchments. High property values and finely subdivided land and dense human infrastructure (e.g. roads, sewer lines) limit the spatial extent of urban river restoration options, while stormwaters and the associated sediment and pollutant loads may limit the potential for restoration projects to reverse degradation. 4. To be effective, urban stream restoration efforts must be integrated within broader catchment management strategies. A key scientific and management challenge is to establish criteria for determining when the design options for urban river restoration are so constrained that a return towards reference or pre‐urbanization conditions is not realistic or feasible and when river restoration presents a viable and effective strategy for improving the ecological condition of these degraded ecosystems.  相似文献

Patterns of newly synthesized proteins in imbibing after-ripenedyoung and aged Agrostemma embryos show differences in the dynamicsof cotyledons and axes. In the course of imbibition the terminationof some syntheses in the embryonal parts of aged seeds is delayed.As was shown by in vitro translation, mRNAs coding for the synthesisof some proteins are still present in the embryos when proteintranslation has already finished. Organic matrix was isolated from the shell of the bivalve Argopectenirradians by decalcification. The capacity of the matrix toinitiate formation of crystals similar in form and orientationto the crystals of normal shell was investigated. Decalcifiedshell matrix placed in an inorganic recalcification solutioninitiated the formation of elongate crystals in parallel arrangementcorresponding to the parallel orientation observed in the matrixfibers and similar to the orientation of the long crystals innormal shell. The detailed form of the crystals deposited invitro was different from that of the normal shell crystals.Electron diffraction analysis of remineralized matrix demonstratedthat the material was calcite, the mineral of normal shell. In contrast, the calcareous tube of the serpulid Hydroides dianthushas crystals lacking uniform arrangement and a matrix whichdoes not have a well-oriented structure. The decalcified tubematrix was recalcified and the mineral posited showed some evidenceof normal orientation. The results demonstrate that matrices of Argopecten shell andHydroides tube can induce crystal formation in vitro. 1. The process‐based phytoplankton community model, PROTECH, was used to model the response of algal biomass to a range of mixed layer depths and extinction coefficients for three contrasting lakes: Blelham Tarn (eutrophic), Bassenthwaite Lake (mesotrophic) and Ullswater (oligotrophic). 2. As expected, in most cases biomass and diversity decreased with decreasing light availability caused by increasing the mixed depth and background extinction coefficient. The communities were generally dominated by phytoplankton tolerant of low light. Further, more novel, factors were identified, however. 3. In Blelham Tarn in the second half of the year, biomass and diversity did not generally decline with deeper mixing and the community was dominated by nitrogen‐fixing phytoplankton because that nutrient was limiting to growth. 4. In Bassenthwaite Lake, changing mixed depth influenced the retention time so that, as the mixed depth declined, the flushing rate in the mixed layer increased to the point that only fast‐growing phytoplankton could dominate. 5. In the oligotrophic Ullswater, changing the mixed depth had a greater effect through nutrient supply rather than light availability. This effect was observed when the mixed layer was relatively shallow (<5.5 m) and the driver for this was that the inflowing nutrients were added to a smaller volume of water, thus increasing nutrient concentrations and algal growth. 6. Therefore, whilst changes in mixed depth generally affect the phytoplankton via commonly recognized factors (light availability, sedimentation rate), it also affected phytoplankton growth and community composition through other important factors such as retention time and nutrient supply.  相似文献
