Salicylic Acid Levels in Thermogenic and Non-Thermogenic Plants   总被引:7,自引:0,他引:7  
The natural trigger for heat production in the thermogenic inflorescencesof Sauromatum guttatum Schott (voodoo lily) was recently identifiedas salicylic acid (SA), which induced heat production at levelsas low as 13 ng g f. wt–1. Since then the levels of SAwere determined in other thermogenic and non-thermogenic plantspecies. In thermogenic inflorescences of five aroid species,and in male cones of at least four thermogenic cycads SA levelsduring heat production exceeded 1 µg g f. wt–1.SA was not detected in the thermogenic flowers of a water lily,Victoria regia Lindl. (Nymphaeaceae), and Bactris major Jacq.(Palmae). Levels of salicylic acid varied substantially in thefloral parts of seven non-thermogenic species and in the leavesof 27 non-thermogenic species. Moss oxalic acid oxidase freed from catalase by boiling is stronglyinhibited by the "metal-complexing" compounds such as thiocyanate,azide, diethyldithiocarbamate, and hydrosulfite. Inactivatedby dialysis against thiocyanate or azide, the enzyme can bereactivated to a considerable extent by the addition of ferricsalt, cytochrome-c or hemoglobin, not by other metal ions, suchas Cu2+, Zn2+ , Mn2+, and Fe2+. Nitrate, chlorate, monoiodoacetate,and iodide also act as strong inhibitors towards moss oxalicacid oxidase. Some enzyme fractions which were obtained by the sodium sulfateprecipitation method were stimulated by Fe3+, but not by cytochrome-cor by other metallic ions. Plants respond to wounding by means of a multitude of reactions, with the purpose of stifling herbivore assault. Phospholipase D (PLD) has previously been implicated in the wounding response. Arabidopsis ( Arabidopsis thaliana ) AtPLD α 1 has been proposed to be activated in intact cells, and the phosphatidic acid (PA) it produces to serve as a precursor for jasmonic acid (JA) synthesis and to be required for wounding-induced gene expression. Independently, PLD activity has been reported to have a bearing on wounding-induced MAPK activation. However, which PLD isoforms are activated, where this activity takes place (in the wounded or non-wounded cells) and what exactly the consequences are is a question that has not been comprehensively addressed. Here, we show that PLD activity during the wounding response is restricted to the ruptured cells using 32Pi-labelled phospholipid analyses of Arabidopsis pld knock-out mutants and PLD -silenced tomato cell-suspension cultures. pldα1 knock-out lines have reduced wounding-induced PA production, and the remainder is completely eliminated in a pldα1 / δ double knock-out line. Surprisingly, wounding-induced protein kinase activation, AtLOX2 gene expression and JA biosynthesis were not affected in these knock-out lines. Moreover, larvae of the Cabbage White butterfly ( Pieris rapae ) grew equally well on wild-type and the pld knock-out mutants.  相似文献

The identification of demographically independent populations and the recognition of management units have been greatly facilitated by the continuing advances in genetic tools. Managements units now play a key role in short-term conservation management programmes of declining species, but their importance in expanding populations receives comparatively little attention. The endangered yellow-eyed penguin ( Megadyptes antipodes ) expanded its range from the subantarctic to New Zealand's South Island a few hundred years ago and this new population now represents almost half of the species' total census size. This dramatic expansion attests to M. antipodes' high dispersal abilities and suggests the species is likely to constitute a single demographic population. Here we test this hypothesis of panmixia by investigating genetic differentiation and levels of gene flow among penguin breeding areas using 12 autosomal microsatellite loci along with mitochondrial control region sequence analyses for 350 individuals. Contrary to our hypothesis, however, the analyses reveal two genetically and geographically distinct assemblages: South Island vs. subantarctic populations. Using assignment tests, we recognize just two first-generation migrants between these populations (corresponding to a migration rate of < 2%), indicating that ongoing levels of long-distance migration are low. Furthermore, the South Island population has low genetic variability compared to the subantarctic population. These results suggest that the South Island population was founded by only a small number of individuals, and that subsequent levels of gene flow have remained low. The demographic independence of the two populations warrants their designation as distinct management units and conservation efforts should be adjusted accordingly to protect both populations.  相似文献

Microcalorimetric analysis of heat-production by single intactsporophylls from thermogenic male cones of two cycad species(Ceratozamia miqueliana H. Wendl. and Zamia furfuracea L. f.)showed an oscillatory behaviour for three consecutive days duringpollen maturation and shedding. In sporophylls from C. rumphiiMiq., a weakly thermogenic species with no maximum of heat-production,such oscillations were not detected. The presence of the alternativerespiratory pathway in the sporophylls of the male cones ofZamia pumila L. has been demonstrated. The thermogenic carpellary appendages of the flowers of Victoriacruziana d'Orb. and the thermogenic staminal appendages of Nelumbolutea (Willd.) Pers. possess the cyanide-insensitive, ‘alternative’respiratory pathway. The presence of this pathway was demonstratedin tissue slices as well as in mitochondria. The thermogenicactivity was accompanied by ultrastructural changes in the Victoriamitochondria. Before anthesis, mitochondria with well-developedcristae were present in appendage tissue in large numbers. Duringanthesis, lamelliform cristae appeared in a different orientation. Antipsychotic (AP) drugs have the potential to cause prolongation of the QT interval corrected for heart rate (QTc). As this risk is dose-dependent, it may be associated with the number of AP drugs concurrently prescribed, which is known to be associated with increased cumulative equivalent AP dosage. This study analysed whether AP dose mediates the relationship between polypharmacy and QTc interval. We used data from a cross-sectional survey that investigated the prevalence of QTc lengthening among people with psychiatric illnesses in Italy. AP polypharmacy was tested for evidence of association with AP dose and QTc interval using the Baron and Kenny mediational model. A total of 725 patients were included in this analysis. Of these, 186 (26%) were treated with two or more AP drugs (AP polypharmacy). The mean cumulative AP dose was significantly higher in those receiving AP polypharmacy (prescribed daily dose/defined daily dose = 2.93, standard deviation 1.31) than monotherapy (prescribed daily dose/defined daily dose = 0.82, standard deviation 0.77) (z = −12.62, p < 0.001). Similarly, the mean QTc interval was significantly longer in those receiving AP polypharmacy (mean = 420.86 milliseconds, standard deviation 27.16) than monotherapy (mean = 413.42 milliseconds, standard deviation 31.54) (z = −2.70, p = 0.006). The Baron and Kenny mediational analysis showed that, after adjustment for confounding variables, AP dose mediates the association between polypharmacy and QTc interval. The present study found that AP polypharmacy is associated with QTc interval, and this effect is mediated by AP dose. Given the high prevalence of AP polypharmacy in real-world clinical practice, clinicians should consider not only the myriad risk factors for QTc prolongation in their patients, but also that adding a second AP drug may further increase risk as compared with monotherapy.  相似文献

There is growing awareness that microevolutionary dynamics may alter ecological processes. Rising temperatures under global change are expected to open windows for establishment of species and genotypes from warmer regions. Yet, microevolutionary tracking of temperature change by local populations may reduce establishment success of these immigrants. We exposed a UK population of the water flea, Daphnia magna , to two temperature regimes during a 1.5-year experimental evolution trial, and subsequently compared competitive strength of non-warm-adapted and warm-adapted D. magna in competition with French genotypes. Our results indicate that local microevolutionary responses to global warming may reduce establishment success of immigrant genotypes that are preadapted to warmer climate. Simulation modeling shows that microevolution results in a reduced likelihood and speed of displacement of local populations by immigrant genotypes under realistic immigrant/resident ratios. We conclude that local evolutionary dynamics may shift the relative impact of local and regional processes in response to global change.  相似文献
