The chemical composition of surface waters of two Dutch moorland pools and of incident precipitation, was monitored from 1982
to 1990. For this period, sulfur and water budgets were calculated using a hydrochemical model developed for well-mixed non-stratifying
lakes. Total atmospheric deposition of S decreased significantly after 1986 at both locations. A model describing the sulfur
budget in terms of input, output and reduction/oxidation processes predicted a fast decrease of pool water SO42− concentrations after a decrease of atmospheric input. However, SO42− concentrations in the surface water was lowered only slightly or remained constant. Apparently a source within the lake caused
the unexpectedly high SO42− concentrations. The possible supply of SO42− from the sediment through regulation by (K-)Al-SO4 containing minerals or desorption of SO42− from positively charged surfaces in the sediment was evaluated. Solubility calculations of pore water with respect to alunite,
basaluminite and jurbanite indicated that SO42− concentration was not regulated by these minerals. It is suggested here (1) that desorption of SO42− from peaty sediments may account for the estimated SO42− supply provided that the adsorption complex is periodically recharged by partial oxidation of the upper bottom sediments
and (2) that because of exposure of a part of the pool bottom to the atmosphere during dry summers and subsequent oxidation
of reduced S, the amount of SO42− may be provided which complements the decreasing depositional SO42− input. In future research these two mechanisms need to be investigated. 相似文献
To evaluate the ecological importance of differences in underground morphology, ten individuals per species were excavated for three species pairs in a coniferous forest in the Cascade Mountains of Oregon. The two species of each pair had similar aboveground morphologies, divergent underground morphologies, and different geographical ranges and habitat relationships. Erythronium montanum and Clintonia uniflora have one or two elliptic leaves per ramet but E. montanum has a short, segmented rhizome and only one ramet per genet, whereas C. uniflora spreads vegetatively via long rhizomes. Streptopus roseus and Smilacina stellata have similar, determinant aerial shoots and spread by long rhizomes, but S. stellata has a dimorphic root and rhizome system that allows it to occupy a wider range of habitats. Rubus pedatus and Rubus lasiococcus both spread via stolons, but R. lasiococcus has deeper roots and can occupy drier habitats than R. pedatus. The differences in underground morphology within the species pairs are consistent with the species’ geographical ranges and habitats occupied, and may be causal in determining distribution patterns. 相似文献
In an oligotrophic moorland pool in The Netherlands, S cycling near the sediment/water boundary was investigated by measuring (1) SO42– reduction rates in the sediment, (2) depletion of SO42– in the overlying water column and (3) release of35S from the sediment into the water column. Two locations differing in sediment type (highly organic and sandy) were compared, with respect to reduction rates and depletion of SO42– in the overlying water.Sulfate reduction rates in sediments of an oligotrophic moorland pool were estimated by diagenetic modelling and whole core35SO42– injection. Rates of SO42– consumption in the overlying water were estimated by changes in SO42– concentration over time in in situ enclosures. Reduction rates ranged from 0.27–11.2 mmol m–2 d–1. Rates of SO42– uptake from the enclosed water column varied from –0.5, –0.3 mmol m–2 d–1 (November) to 0.43–1.81 mmol m–2 d–1 (July, August and April). Maximum rates of oxidation to SO42– in July 1990 estimated by combination of SO42– reduction rates and rates of in situ SO42– uptake in the enclosed water column were 10.3 and 10.5 mmol m–2 d–1 at an organic rich and at a sandy site respectively.Experiments with35S2– and35SO42– tracer suggested (1) a rapid formation of organically bound S from dissimilatory reduced SO42– and (2) the presence of mainly non SO42–-S derived from reduced S transported from the sediment into the overlying water. Several models of activation mechanisms were proposed for G protein-coupled receptors (GPCRs), yet no direct methods exist for their elucidation. The availability of constitutively active mutants has given an opportunity to study active receptor conformations within acceptable limits using models such as the angiotensin II type 1 (AT1)1 receptor mutant N111G-hAT1 which displays an important constitutive activity. Recently, by using methionine proximity assay, we showed for the hAT1 receptor that TMD III, VI, and VII form the ligand-binding pocket of the C-terminal amino acid of an antagonistic AngII analogue. In the present contribution, we investigated whether the same residues would also constitute the ligand-binding contacts in constitutively activated mutant (CAM) receptors. For this purpose, the same Met mutagenesis strategy was carried out on the N111G double mutants. Analysis of 43 receptors mutants in the N111G-hAT1 series, photolabeled and CNBr digested, showed that there were only subtle structural changes between the wt-receptor and its constitutively active form. 相似文献
Peripheral nerve transection and neuroma-in-continuity injuries are associated with permanent functional deficits, often despite successful end-organ reinnervation. Axonal misdirection with non-specific reinnervation, frustrated regeneration and axonal attrition are believed to be among the anatomical substrates that underlie the poor functional recovery associated with these devastating injuries. Yet, functional deficits associated with axonal misdirection in experimental neuroma-in-continuity injuries have not yet been studied. We hypothesized that experimental neuroma-in-continuity injuries would result in motor axon misdirection and attrition with proportional persistent functional deficits. The femoral nerve misdirection model was exploited to assess major motor pathway misdirection and axonal attrition over a spectrum of experimental nerve injuries, with neuroma-in-continuity injuries simulated by the combination of compression and traction forces in 42 male rats. Sciatic nerve injuries were employed in an additional 42 rats, to evaluate the contribution of axonal misdirection to locomotor deficits by a ladder rung task up to 12 weeks. Retrograde motor neuron labeling techniques were utilized to determine the degree of axonal misdirection and attrition. Characteristic histological neuroma-in-continuity features were demonstrated in the neuroma-in-continuity groups and poor functional recovery was seen despite successful nerve regeneration and muscle reinnervation. Good positive and negative correlations were observed respectively between axonal misdirection (p<.0001, r2=.67), motor neuron counts (attrition) (p<.0001, r2=.69) and final functional deficits. We demonstrate prominent motor axon misdirection and attrition in neuroma-in-continuity and transection injuries of mixed motor nerves that contribute to the long-term functional deficits. Although widely accepted in theory, to our knowledge, this is the first experimental evidence to convincingly demonstrate these correlations with data inclusive of the neuroma-in-continuity spectrum. This work emphasizes the need to focus on strategies that promote both robust and accurate nerve regeneration to optimize functional recovery. Maintenance of a high degree of biodiversity in homogeneous environments is poorly understood. A complex cheese starter culture with a long history of use was characterized as a model system to study simple microbial communities. Eight distinct genetic lineages were identified, encompassing two species: Lactococcus lactis and Leuconostoc mesenteroides. The genetic lineages were found to be collections of strains with variable plasmid content and phage sensitivities. Kill-the-winner hypothesis explaining the suppression of the fittest strains by density-dependent phage predation was operational at the strain level. This prevents the eradication of entire genetic lineages from the community during propagation regimes (back-slopping), stabilizing the genetic heterogeneity in the starter culture against environmental uncertainty. 相似文献
Several processes bury plants, but sediment can also be subsequently removed, often by delayed erosion. Thus, the ability to survive multiple years of burial and to respond when released are important to vegetation changes and population dynamics. We experimentally evaluated the effects of delayed removal of tephra (aerially transported volcanic ejecta) in an old-growth forest understory near Mount St. Helens, using 1-m2 plots assigned to three treatments: tephra removed 4 months after deposition (50 plots), tephra removed 28 months after deposition (the delayed erosion treatment, 50 plots), and undisturbed, natural tephra (100 plots). Prior to tephra removal, species density, cover, shoot density, and shoot size in the delayed erosion treatment were all similar to values in natural plots and significantly less than values in plots cleared initially, indicating that 24 months of additional burial adversely affected understory plants. However, all attributes eventually approached pre-eruption values for shrubs and herbs, indicating that erosion greatly facilitated vegetation recovery. Responses varied substantially among species and growth forms. Overall, our experimental results indicate that some plants of most species can respond effectively after release from burial of at least three growing seasons. In addition, the delay of erosion retards ecosystem recovery relative to early erosion, facilitates recovery relative to no erosion, and modifies the trajectory of post-disturbance vegetation change.
Mitochondria of the yeast Saccharomyces cerevisiae constitute a perfect model to study the outer membrane channel modulation as besides the TOM complex channel they contain only a single isoform of the VDAC channel and it is possible to obtain viable mutants devoid of the channel. Here, we report that the fraction of the intermembrane space isolated from wild type and the VDAC channel-depleted yeast mitochondria, except of the well-known VDAC channel modulator activity, displays also the TOM complex channel modulating activity as measured in the reconstituted system and with intact mitochondria. The important factor influencing the action of both modulating activities is the energized state of mitochondria. Moreover, the presence of the VDAC channel itself seems to be crucial to properties of the intermembrane space protein (s) able to modulate the outer membrane channels because in the case of intact mitochondria quantitative differences are observed between modulating capabilities of the fractions isolated from wild type and mutant mitochondria. 相似文献
