Multiple isoforms of tropoelastin, the soluble precursor of elastin, are the products of translation of splice-variant mRNAs derived from the single-copy tropoelastin gene. Previous data had demonstrated DNA sequence heterogeneity in three domains of rat tropoelastin mRNA, indicating alternative splicing of several exons of the rat tropoelastin gene. Rat tropoelastin genomic clones encompassing the sites of alternative splicing were isolated and sequenced. Two sites of alternative splicing identified in rat tropoelastin mRNA sequences corresponded to exons 13-15 and exon 33 of the rat tropoelastin gene. Furthermore, the variable inclusion of an alanine codon in exon 16 resulted from two functional acceptor sites separated by three nucleotides. DNA sequences flanking exons subject to alternative splicing were analyzed. These exons contained splicing signals that differed from consensus sequences and from splicing signals of constitutively spliced exons. Introns immediately 5' of exons 14 and 33, for example, lacked typical polypyrimidine tracts and had weak, overlapping branch point sequences. Further, a region of secondary structure encompassing the acceptor site of exon 13 may influence alternative splicing of this exon. These results demonstrate that multiple cis-acting sequence elements may contribute to alternative splicing of rat tropoelastin pre-mRNA.  相似文献

The effects of plant age and infestation level of twospotted spider mite, Tetranychus urticae Koch (Acari: Tetranychidae), on visible plant damage, and the effect of plant age on spider mite population growth on impatiens, Impatiens wallerana Hook.f. (Ericales: Balsaminaceae), were determined by inoculating impatiens plants of three different ages with two densities of spider mites. Each plant was inoculated with either one adult female mite per three leaves or six leaves based on the average number of leaves on plants of each of the three age classes. Subsequently, leaf damage was correlated with mite-days (cumulative spider mite density) per leaf. The results showed that older aged plants exhibited greater damage than younger plants. Regression models of damage thresholds for each plant age suggest that monitoring for spider mites must be done periodically throughout the entire plant production cycle, but that more attention should be given toward the end of the cycle. Measurements of visible leaf damage were correlated with plant marketability. Specifically, the level of damage (proportion of damaged leaves per plant) at which plant marketability changes from a "premium" to a "discounted" category was 0.04-0.06. Thus, regression equations of the damage threshold could be used to estimate a cumulative spider mite density or mite-days equivalent to the economic threshold. Based on these equations, 5% leaf damage corresponds to 2.1, 1.51, and 1.25 mite-days for youngest, intermediate, and oldest plants, respectively. Because the damage threshold on impatiens was shown to be very low, the action threshold for biological control is essentially zero, and predators would need to be released as soon as damage is observed.  相似文献

We evaluated the effects of predator release pattern and prey distribution on rate of suppression of the twospotted spider mite, Tetranychus urticae Koch (Acari, Tetranychidae) and visual damage to the ornamental plant, Impatiens wallerana Hook.f., in a greenhouse. Sixteen impatiens plants were arranged in a square and infested with the same total number of spider mites distributed either evenly (equal numbers on all plants) or clumped (divided equally among the 4 central plants), simulating a “hot spot.” The predatory mite, Phytoseiulus persimilis Athias-Henriot, was released at a 1:4 predator:prey ratio based on total spider mites in the experimental unit, but the pattern of release was either even or clumped, which simulated broadcast or point-release strategies, respectively. Nine days after predator release, spider mite populations were reduced in all treatments, but only in the clumped pest-clumped predator treatment were spider mites undetectable. Poorest pest suppression occurred in the clumped spider mite-even predator treatment. Eighteen days after predator release, spider mites were eliminated in all treatments, but a reduction in average plant damage occurred only in treatments in which the predator release pattern matched the spider mite distribution (i.e., even-even or clumped-clumped) with the greatest reduction in the even-even treatment. Results suggest that there is an advantage to releasing predators in “hot spots” provided that the recommended predator:prey ratio is maintained within infested patches. If more uniform predator releases are planned, overall predator numbers need to be kept sufficiently high so that the predator:prey ratio of 1:4 shown to prevent damage on impatiens is achieved in higher-density spider mite patches.  相似文献

The present study examined the webbing life type and behavioral response of date palm mite (DPM) Oligonychus afrasiaticus (McGregor) toward its webbing residues, in presence or absence of immature individuals, on leaves and fruits of date palm. DPM exhibited a complicated web life type on both the leaves and fruits of date palm, which is characterized by the following features: complicated irregular web structure; webbing density on the leaves is medium and on the fruits extremely high; eggs are always laid on the threads of web without web cover; fecal pellets are deposited on the substrate surface (both leaf and fruit); quiescent stages are on the silken web threads; and the preferred site for feeding and walking is under the web. Behavioral observation of DPM females revealed that the midrib of leaves and the base of fruits of date palm were the preferred sites for feeding and web construction. The number and development stage of DPM affected the behavioral response of females on date palm leaves but not on the fruits. DPM spent most time feeding under the web on both the leaves and fruits of date palm.  相似文献

Type 2 diabetes mellitus (T2DM) is a chronic and one of the most common metabolic diseases affecting large proportion of world population. Diabetes-induced changes in lipid and renal parameters are major risk factors contributing to diabetic complications such as diabetic nephropathy and cardiovascular diseases. Due to adverse effects associated with pharmacological intervention in the T2DM treatment, there is an increased interest in the research focussing on identifying novel plant based therapeutic agents. Here we report the effects of various coconut products on diabetic, lipid and renal parameters in streptozotocin (STZ)-induced diabetic rat model. Diabetic rats demonstrated a significant increase in serum glucose, and glycated haemoglobin levels (HbA1c). Lipid parameters including triglycerides, total cholesterol, low density lipoprotein cholesterol (LDL-cholesterol) and very low density lipoprotein cholesterol (VLDL-cholesterol) were found to be significantly increased, while high density lipoprotein cholesterol (HDL-cholesterol) was significantly declined in diabetic rats. Diabetic rats also displayed increased serum and kidney creatinine, urea, and total protein levels and increased urine glucose, urea, albumin and creatinine levels. Contrastingly, treatment with virgin and filtered coconut oils, coconut water and coconut milk resulted in a significant reversal in the levels of above studied parameters in diabetic rats. Further, these coconut products markedly prevented diabetes induced histopathological changes in kidney tissue. Collectively, the data demonstrate the antidiabetic, hypolipidemic and renal protective properties of various coconut products and underscore the importance of regular consumption of plant based medicinal products in the treatment of T2DM and its complications.  相似文献

Nepeta is a perennial herbal plant that belongs to the Lamiaceae mint family. Nepta has different species that are widely used in the agriculture, medicine, and pharmaceutical industries. A field experiment was conducted in 2015 and 2016 to determine the effects of potassium humate foliar application [0 (control) and 400 ppm] on growth, essential oil and constituents of three Nepta species [Nepeta cataria (catnip), Nepeta cataria var. citriodora (lemon catnip), and Nepeta grandiflora (giant catmint)] under Egyptian conditions. In all three species, the application of potassium humate increased growth indicators, essential oil content (%) and yield (mL/plant), and flavonoids in each of the two harvests. However, the height of giant catmint was significantly higher than that of lemon catnip, which was higher than that of catnip. Lemon catnip that received potassium humate and harvested second gave the highest essential oil content and yield. The major constituents were geraniol and nepetalactone in catnip; citronellol and geraniol in lemon catnip; and o-cymene, c-terpinene, p-cymene and carvacrol in giant catmint. The highest p-cymene in giant catmint, citronellol in lemon catnip, and geraniol in catnip were obtained from the application of potassium humate, while the highest o-cymene, c-terpinene and carvacrol in giant catmint, geraniol in lemon catnip, and nepetalactone in catnip were obtained from control plants. This study demonstrated the variations among species of nepeta, and how they respond to the application of potassium humate. The findings of this study can guide the customization of potassium humate applications to the three species for achieving desired growth and essential oil production outcomes.  相似文献

Long-persistent phosphorescent smart paints have the ability to continue glowing in the dark for a prolonged time period to function as energy-saving products. Herein, new epoxy/silica nanocomposite paints were prepared with different concentrations of lanthanide-doped aluminate nanoparticles (LAN; SrAl2O4:Eu2+,Dy3+). The LAN pigment was firstly coated with silicon dioxide (SiO2) utilizing the heterogeneous precipitation technique to provide LAN-encapsulated between SiO2 nanoparticles (LAN@SiO2). The epoxy/silica/lanthanide-doped aluminate nanoparticles (ESLAN) nanocomposite paints were coated on steel. The prepared ESLAN paints were studied by transmission electron microscopy (TEM), Fourier-transform infrared (FTIR) spectroscopy, scanning electron microscopy (SEM), X-ray fluorescence (XRF) analysis, and energy-dispersive X-ray spectroscopy (EDS). The transparency and coloration properties of the nanocomposite coated films were explored by CIE Lab parameters and photoluminescence spectra. The ultraviolet-induced luminescence properties of the transparent coated films demonstrated greenish phosphorescence at 518 nm upon excitation at 368 nm. Both hardness and hydrophobic activities were investigated. The anticorrosion activity of the nanocomposite films coated onto mild steel substrates immersed in aqueous sodium chloride (NaCl(aq)) (3.5%) was studied by electrochemical impedance spectroscopy (EIS). The silica-containing coatings were monitored to exhibit anticorrosion properties. Additionally, the nanocomposite films with LAN@SiO2 (25%) exhibited the optimized long-lasting luminescence properties in the dark for 90 min. The nanocomposite films showed highly reversible and durable long-lived phosphorescence.  相似文献

In this work, uranyl ion‐imprinted resin based on 2‐(((4‐hydroxyphenyl)amino)methyl)phenol was synthesized by condensation polymerization of its uranyl complex in presence of resorcinol and formaldehyde cross‐linkers. Numerous instrumental techniques including elemental analysis, Fourier transform infrared spectroscopy, ultraviolet, 1H along with 13C nuclear magnetic resonance spectroscopy have been employed for complete characterization of the synthesized ligand and its uranyl complex. Additionally, the obtained ion‐imprinted and non‐imprinted resins were investigated using scanning electron microscope and Fourier transform infrared spectroscopy. The effects of various essential parameters such as pH, temperature and contact time on removal of uranyl ions have been examined, and the results indicated that the obtained resin exhibited the optimum activity at pH 5. Furthermore, the adsorption process was spontaneous at all studied temperatures and followed the second‐order kinetics model. Also, Langmuir adsorption isotherm exhibited the best fit with the experimental results with maximum adsorption capacity 139.3 mg/g. Moreover, the selectivity studies revealed that the ion‐imprinted resin exhibited an obvious affinity toward the uranyl ions in presence of other metal ions compared with the non‐imprinted resin. Three erythraeid genera Balaustium von Heyden, Charletonia Oudemans, and Erythraeus Latreille (Trombidiformes: Prostigmata) are reported for first time from Saudi Arabia based on three new larval species, Balaustium yousifi sp. n., Charletonia bahaensis sp. n., and Erythraeus (Erythraeus) uhadi sp. n. and one new record Erythraeus (Zaracarus) lancifer Southcott. A high degree of endemism has been recorded for several plant groups collectively in Saint Katherine Protectorate (SKP) in the Sinai Peninsula. Nine endangered endemic plant species in SKP were selected to test the variable abilities of three different DNA barcodes; Riboluse-1,5- Biphosphate Carboxylase/Oxygenase Large subunit (rbcL), Internal Transcribed Spacer (ITS), and the two regions of the plastid gene (ycf1) as well as Start Codon Targeted (SCoT) Polymorphism to find the phylogenetic relationships among them. The three barcodes were generally more capable of finding the genetic relationships among the plant species under study, new barcodes were introduced to the National Centre for Biotechnology Information (NCBI) for the first time through our work. The barcode sequences were efficient in finding the genetic relationships between the nine species. However, SCoT polymorphism could only cluster plant species belonging to the same genus together in one group, but it could not cluster plant species belonging to the same families except for some primers solely. RbcL was the most easily amplified and identified barcode in eight out of the nine species at the species level and the ninth barcode to the genus level. ITS identified all the species to the genus level. Finally, ycf1 identified six out of the eight species, but it could not identify two of the eight species to the genus level.  相似文献
