Solid state fermentation was carried out to compare efficiency of acid, alkaline and urea pretreatment of sugarcane bagasse for production of citric acid using Aspergillus niger ATCC 9142. Plackett-Burman statistical design was used to evaluate significance of variables. Pretreatment of bagasse by urea was known as the most influential treatment to increase citric acid production (137.6g/kg of dry sugarcane bagasse and citric acid yield of 96% based on sugar consumed). Finally, up scaling was achieved to a 20L solid state fermentor in which humidity was constant in gas phase and urea-treated sugarcane bagasse. The produced acid concentration and yield in fermentor was 82.38g/kg of dry substrate and 26.45g/kgday, respectively.  相似文献

Heavy metals and mycotoxins in foodstuffs are one of the major concerns of our world nowadays. Food decontamination with the help of microbial biomass is a cheap, easy, efficient and green method known as bioremoval. Probiotics are able to reduce the availability of heavy metals and toxins in food products. The purpose of this review is to summarize the probiotics and potential probiotics' interesting role in food bio-decontamination. After a brief glance at the definition of potential probiotic strains with bioremoval ability, LABs (lactic acid bacteria) are described as they are the most important groups of probiotics. After that, the role of the main probiotic and potential probiotic strains (Bacillus, Lactobacillus, Lactococcus, Enterococcus, Bifidobacterium, Pediococcus, Propionibacterium, Streptococcus and Saccharomyces cerevisiae) for heavy metals and mycotoxins bioremoval are described. Additionally, the bioremoval mechanism and the effect of some factors in bioremoval efficiency are explained. Finally, the investigations about probiotic and contaminant stability are mentioned. It is worth mentioning that this review article can be exerted in different food and beverage industries to eliminate the heavy metals and mycotoxins in foodstuffs.  相似文献

Host-pathogen interactions can induce epigenetic changes in the host directly, as well as indirectly through secreted factors. Previously, uropathogenic Escherichia coli (UPEC) was shown to increase DNA methyltransferase activity and expression, which was associated with methylation-dependent alterations in the urothelial expression of CDKN2A. Here, we showed that paracrine factors from infected cells alter expression of another epigenetic writer, EZH2, coordinate with proliferation. Urothelial cells were inoculated with UPEC, UPEC derivatives, or vehicle (mock infection) at low moi, washed, then maintained in media with Gentamycin. Urothelial conditioned media (CM) and extracellular vesicles (EV) were isolated after the inoculations and used to treat naïve urothelial cells. EZH2 increased with UPEC infection, inoculation-induced CM, and inoculation-induced EV vs. parallel stimulation derived from mock-inoculated urothelial cells. We found that infection also increased proliferation at one day post-infection, which was blocked by the EZH2 inhibitor UNC1999. Inhibition of demethylation at H3K27me3 had the opposite effect and augmented proliferation. CONCLUSION: Uropathogen-induced paracrine factors act epigenetically by altering expression of EZH2, which plays a key role in early host cell proliferative responses to infection.  相似文献

Lnk, SH2-B, and APS form a conserved adaptor protein family. All of those proteins are expressed in mast cells and their possible functions in signaling through c-Kit or FcRI have been speculated. To investigate roles of Lnk, SH2-B or APS in mast cells, we established IL-3-dependent mast cells from Ink-/-, SH2-B-/-, and APS -/- mice. IL-3-dependent growth of those cells was comparable. Proliferation or adhesion mediated by c-Kit as well as degranulation induced by cross-linking FcRI were normal in the absence of Lnk or SH2-B. In contrast, APS-deficient mast cells showed augmented degranulation after cross-linking FcRI compared to wild-type cells, while c-Kit-mediated proliferation and adhesion were kept unaffected. APS-deficient mast cells showed reduced actin assembly at steady state, although their various intracellular responses induced by cross-linking FcRI were indistinguishable compared to wild-type cells. Our results suggest potential roles of APS in controlling actin cytoskeleton and magnitude of degranulation in mast cells.  相似文献
