Cultured mouse Cloudman melanoma cells, EMT6 breast carcinoma cells, and 3T3 fibroblasts all accumulated in the G2/M phase of the cell cycle when exposed to UVB radiation. The effects of UVB were maximal at 20–30 mJ/cm2 for all three cell lines, and could be observed by flow cytometry as early as 12 hr post irradiation. It has been known since the mid-1970s that MSH receptor binding activity is highest on Cloudman melanoma cells when they are in the G2/M phase of their cycle. Here we show that either UVB irradiation or synchronization of Cloudman cells with colchicine results in a stimulation of MSH binding within 24 hr following treatment, a time when both treatments have resulted in accumulation of cells in the G2/M phase of the cycle. Furthermore, the two treatments performed together on the melanoma cells stimulated MSH receptor activity to the same extent as either treatment performed separately, suggesting that each may be influencing MSH receptor activity solely through a G2/M accumulation of cells. Together, these results raise the possibility that an increase in the number of cells in the G2 phase of the cell cycle is a generalized cellular response to injury, such as UV irradiation. However, in the case of pigment cells this response includes a mechanism for increasing melanin formation, i.e., increased MSH receptor activity. Should this be the case, similar G2/M “injury responses” of other cell types might be expected, consistent with their differentiated phenotypes.  相似文献

Twenty-two genotypes of five Brassica species (oilseed rapeand mustard), consisting of two different maturity groups anddiffering in plant morphology, were grown on sandy soil withoutpost-sowing irrigation during 1982–83 winter season. Canopytemperature (Tc minus air temperature (Ta, stomatal conductance($) and osmotic adjustment were studied on two different deardays corresponding to the siliqua formation stage in early duration(137–150 d) and late maturing genotypes (151–165d) of oilseed Brassicas. The absolute mid-day values of transpirationalcooling (Tc–Ta) were relatively higher in B.juncea andB. carinata than in B. tomifortii, B. napus and B. campestris.Tc-Tm showed a genetic variability ranging from 0-5 to 4 Cfor early maturing genotypes, and from 0–5 to 5 C forlate maturing genotypes. Mid-day Tc–Ta showed significantpositive correlation with Kh osmotic adjustment and seed yieldin Brassica cultivars. Early genotypes out-yielded the latematuring ones, despite the lower values of AT, and Tc–Taat siliqua formation in the former rather than later group ofcultivars. It is suggested that average mid-day Tc–Taas observed with an infrared thermometer, could effectivelybe used as a technique to screen Brassica genotypes for droughttolerance under receding soil moisture conditions.  相似文献

When human skin is exposed to ultraviolet (UV) light, a highly complex cascade of events ensues that culminates, among other things, in increased skin melanin content. From analyses at the tissue and cellular level, it has been shown that following exposure to UV light there is an increase in the number of active melanocytes in the basal layer of the epidermis, and individual melanocytes are stimulated to produce more melanin. In addition, the rate of transfer of melanosomes from melanocytes to keratinocytes is apparently increased, although the role of UV light in this process remains to be demonstrated. Recent biochemical evidence is reviewed on factors that regulate these processes. A plausible explanation for the effects of UV on pigmentation is that there are mechanisms in the skin for the orderly, regulated reception of UV signals that are then transduced to initiate the cascade. The signals involve both melanocytes and keratinocytes, and avail-able evidence supports a model in which melanotropins and their receptors play a central role in the process.  相似文献

The inhibitory effect of arbutin, a naturally occurring β-D-glucopyranoside derivative of hydroquinone, on melanogenesis was studied biochemically by using human melano-cytes in culture. Cells were cultured in the presence of different concentrations of arbutin. The maximum concentration of arbutin that was not inhibitory to growth of the cells was 100 ug/ml. At that concentration, melanin synthesis was inhibited significantly by ~20% after 5 days, compared with untreated cells. This phenotypic change was associated with the inhibition of tyrosinase and DHICA polymerase activities, and the degree of inhibition was dose dependent. No significant difference in DOPAchrome tautomerase (DT) activity was observed before or after arbutin treatment. Western blotting experiments revealed there were no changes in protein content or in molecular size of tyrosinase, TRP-1 or TRP-2, indicating that inhibition of tyrosinase activity by arbutin might be due to effects at the post-translational level.  相似文献

The genus Prosopis (Leguminosae, Mimosoideae), comprises 44 species widely distributed in arid and semi-arid zones. Prosopis pallida (Humb. & Bonpl. ex Willd.) Kunth and P. juliflora (Sw.) DC. are the two species that are truly tropical apart from P. africana, which is native to tropical Africa (Pasiecznik et al. 2004), and they have been introduced widely beyond their native ranges. However, taxonomic confusion within the genus has hampered exploitation and better management of the species. The present study focusses primarily on evaluating the genetic relationship between Prosopis species from the section Algarobia, containing most species of economic importance, though P. tamarugo from section Strombocarpa is also included for comparison. In total, 12 Prosopis species and a putative P. pallida × P. chilensis hybrid were assessed for their genetic relationships based on RAPD markers and microsatellite transferability. The results show that P. pallida and P. juliflora are not closely related despite some morphological similarity. Evidence also agrees with previous studies which suggest that the grouping of series in section Algarobia is artificial.  相似文献

Dwarf plant height and tillering ability are two of the most important agronomic traits that determine the plant architecture, and have profound influence on grain yield in rice. To understand the molecular mechanism controlling these two traits, an EMS-induced recessive dwarf and increased tillering1 (dit1) mutant was characterized. The mutant showed proportionate reduction in each internode as compared to wild type revealing that it belonged to the category of dn-type of dwarf mutants. Besides, exogenous application of GA3 and 24-epibrassinolide, did not have any effect on the phenotype of the mutant. The gene was mapped on the long arm of chromosome 4, identified through positional candidate approach and verified by cosegregation analysis. It was found to encode carotenoid cleavage dioxygenase7 (CCD7) and identified as an allele of htd1. The mutant carried substitution of two nucleotides CC to AA in the sixth exon of the gene that resulted in substitution of serine by a stop codon in the mutant, and thus formation of a truncated protein, unlike amino acid substitution event in htd1. The new allele will facilitate further functional characterization of this gene, which may lead to unfolding of newer signalling pathways involving plant development and architecture.  相似文献

Indian geographical populations of Drosophila melanogaster, collected at latitude 10–32.3 oN, longitude 76.1–80.1 oE and altitude 16–2050 m, were analysed for changes in Adh 1 frequency and utilization potential of diverse alcoholic resources. Until now, there have been limited data on climatic association of correlated changes on these aspects in continental populations of D. melanogaster. Indian populations demonstrate an increase in Adh F frequency by 0.04 per degree latitude and significantly higher utilization of ethanol (9.0–15.2%), acetic acid (8.1–12.5%), n-propanol (2.55-4.25%), 2-propanol (1.5-3.5%), n-butanol (2.15 3.5%) and 2-butanol (0.68-1.2%). Data also revealed significant correlation with latitude as well as altitude. Regression analysis of climatic data from collection sites confirmed the observed clinal variation of Adh F as well as alcoholic utilization in D. melanogaster. On the Indian subcontinent, variation in Adh F frequency and alcoholic resource utilization along increasing latitude as well as altitude are due to climatic selection factors such as temperature and rainfall.  相似文献

Elucidation of the genetic diversity and relatedness of the subpopulations of India may provide a unique resource for future analysis of genetic association of several critical community-specific complex diseases. We performed a comprehensive exploration of single nucleotide polymorphisms (SNPs) within the gene DNA ligase 1 (LIG1) among a multiethnic panel of Indian subpopulations representative of the ethnic, linguistic and geographical diversity of India using a two-stage design involving DNA resequencing-based SNP discovery followed by SNP validation using sequenom-based genotyping. Thirty SNPs were identified in LIG1 gene using DNA resequencing including three promoter SNPs and one coding SNP. Following SNP validation, the SNPs rs20580/C19008A and rs3730862/C8804T were found to have the most widespread prevalence with noticeable variations in minor allele frequencies both between the Indian subpopulation groups and also from those reported on other major world populations. Subsequently, SNPs found in Indian subpopulations were analysed using bioinformatics-based approaches and compared with SNP data available on major world populations. Further, we also performed genotype–phenotype association analysis of LIG1 SNPs with publicly available data on LIG1 mRNA expression in HapMap samples. Results showed polymorphisms in LIG1 affect its expression and may therefore change its function. Our results stress upon the uniqueness of the Indian population with respect to the worldwide scenario and suggest that any epidemiological study undertaken on the global population should take this distinctiveness in consideration and avoid making generalized conclusions.  相似文献
