Abstract. Epidermal (non-stomatally-controlled) conductance from the fourth leaf, first node leaf, flag leaf and ear of durum wheat (Triticum turgidum var durum L.) grown under Mediterranean field conditions has been measured, along with leaf stomatal frequency and the amount and distribution of epicuticular waxes. Measurements were carried out on varieties and land-races from the Middle East, North Africa, ‘Institut National de la Recherche Agricole’ (INRA) and ‘Centra Internacional de Mejora de Maiz y Trigo’ (CIMMYT). Significant differences were observed among genotypes in the epidermal conductances (ge) of the four organs. For each of the four organs tested, genotypes from the Middle East and CIMMYT showed higher ge. values than those from North Africa and INRA. Ears showed epidermal conductances that were more than four times higher than those of leaves when ge. values were expressed per unit dry weight. The amount of epicuticular waxes was higher in the fourth leaves, intermediate in the first node and flag leaves and lower in the ears. For each organ, ge differences among genotypes were unrelated with the amount of epicuticular waxes. Removal of epicuticular waxes by dipping the organs into chloroform significantly increased the epidermal conductance for the fourth and first node leaves and the ear. However, this did not occur for the flag leaf. For the fourth leaf, ge of intact leaves and ge of leaves in which epicuticular waxes were removed were unrelated (r = -0.265). The regression coefficient of this relation for the first node and flag leaves showed values of 0.666 and 0.650 (P > 0.05), respectively, and values were even higher in the ear (r > m 0.892, P > 0.01). Scanning electron microscope analysis showed that wax bloom decreased from the fourth leaf to the flag leaf, whereas the extent of amorphous wax increased. Wax bloom in leaves consisted mainly of deposits of thin wax plates. In the ears and the adaxial surface of flag leaves, fibrillar waxes predominated. In the first node and flag leaves, the wax deposits on the adaxial side cover the surface of the leaf more densely and uniformly than those on the abaxial side. There was no significant correlation between ge and total stomatal density, or between ge and either adaxial or abaxial stomatal density for any sample of the three different leaves. The contribution of epicuticular waxes plus total stomatal frequency only explained 42.4, 11.8, 28.3 and 16% of ge (per unit leaf area) variations for the fourth leaf, first node leaf, flag leaf and the combined variation of the three leaves together, respectively. From these results, it is concluded that complex interrelationship between different morphophysiological characteristics probably control ge differences among genotypes and that these interrelationships differ for each different plant part.  相似文献

Net photosynthesis rate (Pn), stomatal conductance to CO2 andresidual conductance to CO2 were measured in the last six leaves(the sixth or flag leaf and the preceding five leaves) of Triticumaestivum L. cv. Kolibri plants grown in Mediterranean conditions.Recently fully expanded leaves of well-watered plants were alwaysused. Measurements were made at saturating photosynthetic photonflux density, and at ambient CO2 and O2 levels. The specificleaf area, total organic nitrogen content, some anatomical characteristics,and other parameters, were measured on the same leaves usedfor gas exchange experiments. A progressive xeromorphic adaptation in the leaf structure wasobserved with increasing leaf insertion levels. Furthermore,mesophyll cell volume per unit leaf area (Vmes/A) decreasedby 52·6% from the first leaf to the flag leaf. Mesophyllcell area per unit leaf area also decreased, but only by 24·5%.However, nitrogen content per unit mesophyll cell volume increasedby 50·6% from the first leaf to the flag leaf. This increasecould be associated to an observed higher number of chloroplastcross-sections per mm2 of mesophyll cell cross-sectional areain the flag leaf: values of 23000 in the first leaf and 48000in the flag leaf were obtained. Pn per unit leaf area remainedfairly constant at the different insertion levels: values of33·83±0·93 mg dm–2 h–1 and32·32±1·61 mg dm–2 h–1 wereobtained for the first leaf and the flag leaf, respectively.Residual conductance, however, decreased by 18·2% fromthe first leaf to the flag leaf. Stomatal conductance increasedby 41·7%. The steadiness in Pn per unit leaf area across the leaf insertionlevels could be mainly accounted for by an opposing effect betweena decrease in Vmes/A and a more closely packed arrangement ofphotosynthetic apparatus. There is continuing controversy over whether a degree of C4 photosynthetic metabolism exists in ears of C3 cereals. In this context, CO2 exchange and the initial products of photosynthesis were examined in flag leaf blades and various ear parts of two durum wheat (Triticum durum Desf.) and two six-rowed barley (Hordeum vulgare L.) cultivars. Three weeks after anthesis, the CO2 compensation concentration at 210 mmol mol?1 O2 in durum wheat and barley ear parts was similar to or greater than that in flag leaves. The O2 dependence of the CO2 compensation concentration in durum wheat ear parts, as well as in the flag leaf blade, was linear, as expected for C3 photosynthesis. In a complementary experiment, intact and attached ears and flag leaf blades of barley and durum wheat were radio-labelled with 14CO2 during a 10s pulse, and the initial products of fixation were studied in various parts of the ears (awns, glumes, inner bracts and grains) and in the flag leaf blade. All tissues assimilated CO2 mainly by the Calvin (C3) cycle, with little fixation of 14CO2 into the C4 acids malate and aspartate (about 10% or less). These collective data support the conclusion that in the ear parts of these C3 cereals C4 photosynthetic metabolism is nil.  相似文献

The relationships between leaf and kernel carbon isotope discriminations (Δ) and several leaf structural parameters that are indicators of photosynthetic capacity were studied in durum wheat grown in the field under three water regimens. A set of 144 genotypes were cultivated in two rain-fed trials, and 125 of these were grown under supplementary irrigation before heading. Total chlorophyll and nitrogen (N) contents, the dry mass per unit leaf area (LDM, the reciprocal of specific leaf area) and carbon isotope discrimination (Δ) were measured in penultimate leaves and Δ of mature kernels was also analysed. Both LDM and N per unit area showed significant (P≤ 0.001) negative correlation (r=–0.60 and r=–0.36, respectively) with leaf Δ in the wettest trial. Little or no correlation was found for any structural parameter and leaf Δ in the rain-fed trials. In contrast, in the two rain-fed trials LDM was the parameter with the strongest positive correlation (P≤ 0.001) with kernel Δ (r= 0.47 and 0.30) and grain yield (r= 0.43 and 0.29), whereas no correlation was found in the irrigation trial. These correlations, rather than representing a causal link between the amount of photosynthetic tissue and Δ, were probably indirect associations caused by a parallel effect of water status and phenology on leaf structure, grain Δ and yield. Correlations across trials (i.e. environments) between leaf structure and either Δ and yield were very high, although also spurious. Our results suggest that LDM should be used to cull segregating population differences in leaf Δ based on the internal photosynthetic capacity only in the absence of drought. Selecting for kernel Δ and grain yield on the basis of LDM is worthwhile for rain-fed trials.  相似文献

Carbon isotope discrimination (Δ) was determined for kernels of six-row barley and durum wheat cultivated in the western Mediterranean basin during the last seven millennia. Samples came from different archaeological sites in Catalonia (north-east Spain) and in the south-east of Spain (mainly eastern Andalusia). Samples from the present were also analysed. Mean values of Δ for barley and durum wheat grains decreased slightly from Neolithic (7000–5000 BP) to Chalcolithic-Bronze (5000–3000 BP) and Iron ages (3000–2200 BP) both in Catalonia and in south-east (SE) Spain. Values were consistently lower in SE Spain than in Catalonia throughout these five millennia, which suggests that Catalonia was less arid than SE Spain in this period. Within a given region, current discrimination values for kernels of the same cereal species cultivated under rainfed conditions were lower than those of archaeological grains, which implies more arid conditions at present. Furthermore, an empirical relationship between Δ of mature kernels and total precipitation (plus irrigation where applicable) during grain filling (r2 = 0.73, N = 25) was established for barley, currently cultivated at different locations in the western Mediterranean basin in Spain. The resulting relationship was applied to the Δ data for barley kernels from 10 archaeological sites in Catalonia and 10 sites in SE Spain, to estimate the precipitation during grain filling at the time the kernels were produced. For both regions, current climatic conditions are consistently more arid than those inferred for the Neolithic, Bronze and Iron ages. In addition, although Catalonia was estimated to have had consistently wetter conditions (about 20% more precipitation) than SE Spain throughout these millennia, differences in precipitation between these two regions have recently increased, with 79% more precipitation in Catalonia. Results indicate a more rapid increase in aridity in SE Spain than in Catalonia, probably produced during the last few centuries, and due to anthropogenic causes.  相似文献

Measurement of stable isotopes in plant dry matter is a useful phenotypic tool for speeding up breeding advance in C3 crops exposed to different water regimes. However, the situation in C4 crops is far from resolved, since their photosynthetic metabolism precludes (at least in maize) the use of carbon isotope discrimination. This paper investigates the use of oxygen isotope enrichment (Δ18O) as a new secondary trait for yield potential and drought resistance in maize ( Zea mays L). A set of tropical maize hybrids developed by the International Maize and Wheat Improvement Center was grown under three contrasting water regimes in field conditions. Water regimes clearly affected plant growth and yield. In accordance with the current theory, a decrease in water input was translated into large decreases in stomatal conductance and increases in leaf temperature together with concomitant 18O enrichment of plant matter (leaves and kernels). In addition, kernel Δ18O correlated negatively with grain yield under well-watered and intermediate water stress conditions, while it correlated positively under severe water stress conditions. Therefore, genotypes showing lower kernel Δ18O under well-watered and intermediate water stress had higher yields in these environments, while the opposite trend was found under severe water stress conditions. This illustrates the usefulness of Δ18O for selecting the genotypes best suited to differing water conditions.  相似文献

This study compared the performance of the stable isotope composition of carbon (δ13C), oxygen (δ18O) and nitrogen (δ15N) by tracking plant response and genotypic variability of durum wheat to different salinity conditions. To that end, δ13C, δ18O and δ15N were analysed in dry matter (dm) and the water‐soluble fraction (wsf) of leaves from plants exposed to salinity, either soon after plant emergence or at anthesis. The δ13C and δ18O of the wsf recorded the recent growing conditions, including changes in evaporative conditions. Regardless of the plant part (dm or wsf), δ13C and δ18O increased and δ15N decreased in response to stress. When the stress conditions were established just after emergence, δ15N and δ13C correlated positively with genotypic differences in biomass, whereas δ18O correlated negatively in the most severe treatment. When the stress conditions were imposed at anthesis, relationships between the three isotope signatures and biomass were only significant and positive within the most severe treatments. The results show that nitrogen metabolism, together with stomatal limitation, is involved in the genotypic response to salinity, with the relative importance of each factor depending on the severity and duration of the stress as well as the phenological stage that the stress occurs.  相似文献

Breeding for improved productivity has been tremendously successful in the last half‐century, but needs to be even more efficient in the future. Hope based on contributions from molecular biology for improved yield potential seems to depend upon an improved knowledge of yield physiology. This knowledge may assist breeding either directly, recommending selection criteria, or indirectly identifying simpler traits that could be reliably mapped and selected for through marker‐assisted selection. Physiological traits associated with improved performance under water‐limited conditions, include phenology (that allows the crop to escape stresses) and those associated with improved water use, water use efficiency and partitioning. Undoubtedly, earliness has been the predominant trait improved for under Mediterranean conditions, and may not be a prospective trait for future breeding. Different traits that may confer the ability to the crop for capturing more water, such as deeper root systems or osmotic adjustment, may be unworkable in terms of their direct use in selection and surrogates would be needed. For instance, canopy temperature depression and discrimination against 13C may be used to assess improved ability to capture water (in these cases yield is positively related to discrimination against 13C in grains). Early vigour, which allows faster ground coverage, also increases the amount of water actually transpired by the canopy by reducing direct evaporation and presents substantial intraspecific variation, and selection for this trait may be successfully carried out either directly or through the use of vegetation indexes. Improved water use efficiency based on transpiration efficiency is largely restricted to conditions where additional water is not available. A constitutively low stomatal conductance or a high stomatal sensitivity may optimise the transpiration efficiency. In this context, discrimination against 13C is also a simple and reliable measure of water use efficiency, and in cases in which no major differences in capturing water is possible discrimination against 13C correlates negatively with yield. Substantial further improvements in partitioning may be limited in most cereals.  相似文献

The role of ear photosynthesis in grain filling was studied in a number of durum wheat (Triticum turgidum var durum L.) landraces and varieties from the Middle East, North Africa, and from the collections of ‘Institut National de la Recherche Agronomique’ (INRA, France) and ‘Centro International de Mejora de Maiz y Trigo’ (CIMMYT, Mexico). Plants were grown in the field in a Mediterranean climate. Flag leaves (blade plus sheath) and ears were kept in the dark from 1 week after anthesis to maturity which reduced grain weight by 22.4% and 59.0%, respectively. In a further experiment, the carbon isotope discrimination ratio (Δ) of ear bracts, awns and flag leaves was measured on samples taken at anthesis and on mature kernels. The mean value of Δ for the water soluble fraction of bracts (17.0‰) and awns (17.7‰) were lower than those of leaves (19.5‰) and fairly similar to those of kernels (17.4‰) averaged across all genotypes. Data indicate that most of the photosynthates in the grain come from ear parts and not from flag leaves. In addition, a higher water use efficiency (WUE) of ear parts than of the flag leaf is suggested by their lower Δ values. Gas exchange in ears and flag leaves was measured during grain filling. Averaged over all genotypes, CO2 diffusive conductance was about five times higher in the flag leaf than in the spike (with distal portions of awns outside the photosynthetic chamber) 2 weeks after anthesis. In absolute terms, the dark respiration rate (Rd) was greater than the net photosynthesis rate (Pn) by a factor of 1.74 in the spike, whereas Rd was much smaller, only 22.1, 65.7 and 24.8% of Pn in blade, sheath and awns, respectively. Data indicate that photosynthesis, and hence the water use efficiency (photosynthesis/transpiration), is greatly underestimated in ears because of the high rates of respiration which diminish the measured rates of net CO2 exchange. Results of 13C discrimination and gas exchange show that genotypes from North Africa have higher WUE than those from the Middle East. The high Rd values of ears as well as their low diffusive conductance suggest that CO2 from respiration may be used as source of carbon for ear photosynthesis. In the same way, the anatomy of glumes, for example, supports the role of bracts using internal CO2 as source of photosynthesis. In the first experiment, the Δ in mature grains from culms with darkened ears compared with control culms provided further evidence in support of this hypothesis. Thus, the Δ from kernels of control plants was 0.40 higher than that from ear-darkened plants, probably because of some degree of refixation (recycling) of respired CO2 in the grains.  相似文献

Comparisons of leaf structure and gas exchange characteristicshave been made between flag leaves of four old genotypes ofcultivated tetraploid wheats and three current varieties ofhexaploid Triticum aestivum grown under Mediterranean climateconditions. For some genotypes the effect of varying the sowingdate was investigated. In the hexaploid wheat Kolibri the effectof sowing date on leaf structure and gas exchange of the penultimateleaf was also studied. Flag leaves differed significantly in photosynthetic capacityand leaf structure characteristics between genotypes, withineach ploidy level. When the mean values for each ploidy levelwere considered, there were no significant differences in valuesfor photosynthesis per unit leaf area, stomatal conductance,intercellular CO2 concentration, residual CO2 conductance andwater-use efficiency between the tetraploid and hexaploid wheatssown on the same date. When comparisons were made of leaf structurethe only significant differences observed were in adaxial andabaxial stomatal frequencies and leaf area: mean values of theseparameters were higher in tetraploid than in hexaploid wheats. The changes in leaf structure in response to varying sowingdate were significant and followed the same pattern in all thegenotypes studied: a xeromorphic adaptation was observed inlater sowings in response to warmer climate. Such structuralchanges affected some gas exchange characteristics. For example,the area of flag leaves decreased by 60% in the hexaploid wheatKolibri from first to last sowing, which led to a transpirativeloss of 49% per single leaf, in spite of the fact that transpirationrate per unit leaf area increased by 26%. Penultimate leavesof cv. Kolibri followed a fairly similar adaptive pattern inlater sowings as compared with flag leaves. The adaptive significanceof changes in leaf structure and gas exchange characteristicswith varying sowing date under Mediterranean climate conditionsis discussed.
