  1. Respiratory activity of sweet potato root tissues, stored at0° (chilled) and 20° (control), was measured. The activityof the control sample remained approximately constant, whilethat of the chilled one declined sharply after about ten daysof the treatment. Just before this time, DNP-stimulated respiratoryactivity began to drop in the chilled sample, but remained constantin the control.

Oxidative and phosphorylative activities ofmitochondria fromthe chilled tissues were compared with thosefrom the control.Exactly as in respiration of the intact roottissues, a sharpdecline in mitochondrial oxidative activitywas found in thechilled tissue after about ten days of thetreatment, but notin the control. In spite of the decreasein oxidative activityof mitochondria, the P/O ratio remainedfairly constant throughoutthe storage. Protein nitrogen ofmitochondria in root tissuesalso remained constant.
  3. Contentof polyphenolic substances did not show any significantdifferencebetween the chilled and control tissues.

The DEAE-cellulose column chromatography has shown two differentforms of starch synthetase, which are referred to as fractionsI and II in extracts of rice seeds (non-waxy and waxy varieties)harvested at the milky stage. Similarly treated leaf extractsof rice (non-waxy) and maize (non-waxy) also demonstrate dieexistence of two major isozyme fractions. In all enzyme preparationstested, ADP-glucose was the sole glucosyl donor and UDP-glucosewas totally inactive. Rechromatography, on a DEAE-cellulosecolumn, of two enzyme fractions (I and II) separated from non-waxyrice seed extracts did not alter their elution patterns. Someof their enzymic properties were compared, in particular, theirglucosyl-acceptor (primer) specificities. Regardless of potentamylase activities in the two fractions, notable differenceswere observed in that fraction I utilized the long-chain oligosaccharides[maltododecaose] and various types of high molecular -glucansmore readily than fraction II. However, short-chain oligosaccharides[maltose, maltotriose and maltotetraose] were utilized morereadily by fraction II than by fraction I. A possible role forthe two starch synthetase isozymes in starch synthesis in plantcells is discussed. (Received January 5, 1971; )  相似文献

RuDP carboxylase isolated from autotrophically grown cells ofphotosynthetic sulfur bacterium, Chromatium strain D, was partiallypurified by (NH4)2SO4 precipitation and Sephadex G-200 gel filtration.The molecular size of the bacterial RuDP carboxylase was foundto be large, analogous to that of the plant enzyme, in agreementwith results of previous workers. Sucrose density gradient centrifugationshowed the Srel to be approximately 18; the omission of Mg++caused no dissociation of the enzyme molecule in its subunits.Chromatium RuDP carboxylase showed similarities to the plantenzyme in some of its kinetic properties; (a) a shift of pHoptimum to the neutral side from the alkaline side on the additionof Mg++, (b) deviation of the substrate concentration (NaHCO3)-activityrelationship from the MICHAELIS formula and (c) a marked stimulativeeffect of Mg++. A unique sigmoidal saturation curve of the enzymeto RuDP, which had been detected in Rhodospirillum rubrum andRhodopseudomonas spheroides RuDP carboxylase in the absenceof Mg++, was not found. Another characteristic feature of ChromatiumRuDP carboxylase is its partial immunological response to therabbit anti-spinach RuDP carboxylase serum as detected by theinhibition of the carboxylation reaction due to the antibody-antigenreaction. 1Part X, Structure and Function of Chloroplast Proteins. Some enzymic properties of RuDP carboxylase isolated from Rhodopseudomonasspheroides and Rhodospirillum rubrum, grown under autotrophic,semi-autotrophic and heterotrophic conditions were studied.Regardless of the growing condition, the nature of the enzymefrom each respective bacterium remained unchanged. The molecularweight of R. spheroides and R. rubrum RuDP carboxylase was estimatedto be approximately 2.4x105 and 8.3x104, respectively. Bothenzymes require Mg++. The stimulative effect of K+ on the activity of granule-boundstarch synthetase was uniformly observed in the enzyme preparationsobtained from the root, leaf and stem of sweet potato plants.The stimulation of K+ was most marked with the sweet potatoenzyme, while the broad bean enzyme was also appreciably stimulated.The K+-effect was very small in the enzyme from rice, barleyand taro plants.
