第十四届国际微生物学大会的情况介绍

国际微生物学联合会(IUMS)于1986年9月7日至13日在英国Manchester召开第十四届国际微生物学大会,我国派有8人代表团参加。现将会议概况,学术活动等扼要汇报如下: (一)会议概况国际微生物学大会以细菌学和真菌学为主,病毒学为副,因国际病毒学大会将另行召开。本届大会规模颇大,约有3,500人参加。学术活

新疆六个民族指纹白线的调查研究The hand Print of Six Nationalities in Xinjiang

本文首次报道了新家6各民族6134人（南3181人、女2953人）的指纹白线出现频率。结果表明,白线频率女高于男,民族间亦有差异,其中维吾尔、哈萨克、鸟孜别克族频率接近。塔吉克族居住高寒高原地区,属白色人种,频率明显低于以上3个民族,锡伯族和汉族生活习惯近似,且高于以上4个民族,作者认为,白线出现频率的高低与种族、性别、年龄和群体生长环境及生活习惯有关。We’ve studied 6134 individuals (male 3181, fimale 2953) of six nationalities in Xinjiang.The result is that the white line of hand print is associated with rae,sex,age,grow environments of groups and life habbits.The ifference of races is:The appearant rates of white line of Weivuerite,Hasakese and Wuzibese are near,Tajike’s rate is lower than xiboman’s and han are higher than others.     

原生质体诱变选育无孢平菇

用紫外线照射紫孢侧耳(Pleurotus sapidus)双核菌丝原生质体,再生后筛选生长势好的菌株, 通过出菇试验得到了生产性状与紫孢侧耳一致的无孢和少孢平菇新品种。Abstract: This paper reported isolation and regeneration of the dikaryocyte protoplasts from Pleurotus sapidus, and the protoplasts were treated by U.V-irradiation for selection spore less mutants.     

Distribution and Significance of Nitric Oxide Synthase in Brain Tissues of Rats Exposed to Deltamethrin Insecticide

The distribution of nitric oxide synthase(NOS)in brain tissues of rats exposed to deltamethrininsecticide has been examined by histochemical NADPH-diaphorase staining techniques on frozen sec-tions.After injection of deltamethrin(12.5mg/kg,i.p.),a reproducible sequence of toxic signs ofhyperexcitability were elicited.The observation and image analysis showed that,within brain sec-tions of rats exposed to deltamethrin,the numbers and the total staining areas of the NOS positiveneurons were greatly increased,especially in cerebral cortex,hippocampal formation and paraventric-ular nucleus.In addition,the density of single neuron and the processes were also increased.The re-sults suggested that deltamethrin may induce the NOS expression or activate the NOS activity.TheNOS activation may involve in the chains responsible for the excitatory neurotoxicities induced bydeltamethrin.

Effect of Human WEE1 and Stem Cell Factor on Human CD34^＋ Umbilical Cord Blood Cell Damage Induced by Chemotherapeutic Agents

Myelosuppression is one of the major side-effects of most anticancer drugs. To achieve myeloprotection, one bicistronic vector encoding anti-apoptotic protein human WEE l （WEElHu） and proliferation-stimulating stem cell factor （SCF） was generated. In this study, we selected human umbilical cord blood CD34^＋ cells as the in vitro model in an attempt to investigate whether WEEIHu, rather than conventional drug-resistant genes, can be introduced to rescue cells from the damage by chemotherapeutic agents such as cisplatin, adriamycin, mitomycin-c and 5-fluorouracil. Cell viability and cytotoxicity assay, colony-forming units in culture assay and externalization of phospholipid phosphatidylserine analysis showed that the expression of WEElHu and SCF in CD34^＋ cells provided the cells with some protection. These findings suggest that the expression of WEElHu and SCF might rescue CD34^＋ cells from chemotherapyinduced myelosuppression.     

PCR-RFLP检测LDL受体基因TaqⅠ多态性位点的研究

应用聚合酶链反应(PCR)扩增人类LDL受体基因外显子4-内含子4-外显子5片段,PCR产物为1.55kb,DNA片段经序列鉴定后,进行TaqI酶切位点的RFLP分析。结果显示:中国汉族人群LDL受体基因中存在着TaqⅠ酶切位点多态性; 200个LDL受体等位基因中TaqⅠ酶切位点出现的频率为0.515,该点频率较为适中, 可作为中国汉族人群LDL受体基 因的遗传标志来进行家族性高胆固醇血症(FH)的基因诊断。所建立起的LDL受体基因TaqⅠ位点的PCR -RFLP方法具有快速、简便的特点,在FH的基因诊断上有应用价值。 Abstract:To develop rapid and sensitive technique for detectin the TaqI polymorphism at the human LDL receptor gene in Chinese,the exon4-intron4-exon5 of the human LDL receptor gene was amplified by polymerase chain reaction(PCR).The PCR products were directly analysed by restriction fragment length polymorphisms(RFLP).The results showed that the TaqI polymorphism is associated with the LDL receptor gene in Chinese of Han nationality;The frequency of T= allele (presence of TaqI cutting site)is 0.515 in 200 LDL receptor alleles.This technique may be used for rapid and sensitive screening of the LDL receptor gene for the TaqI polymorphism.     

A Primary Study of the Subgroups of T Lymphocytes in MHV-3 Induced Chronic Viral Hepatitis

To study the contribution of T cell subsets in the pathogenesis of Murine hepatitis virus Type 3(MHV-3) induced chronic viral hepatitis in C3H/Hej mice,ninety C3H/Hej mice were chosen to individually receive 10 plaque forming units(PFU)of MHV-3 intraperitoneally.The changes of virus titer and pathology in liver tissue were examined by standard plaque assay and by the hematoxylin/eosin(HE) staining method from 2 days post MHV-3 infection.The ratios of T cell subsets including CD3 CD4 CD8-,CD3 CD4-CD8 ,CD3 CD4-CD8-,CD3 CD4 CD25 ,CD3 CD4 CD25-and CD3 CD4-CD25 T lymphocyte of total T lymphocytes in blood,spleen and liver were examined at 0,2,4,6,8,10,12,15,20,25,30,40 days post MHV-3 infection by flow cytosorting.We observed that the virus titer raised and showed persistent virus duplications and inflammatory changes in the livers of C3H/Hej mice from 2 days post MHV-3 infection.The double negative T cell(DN Treg cell) and CD4 CD25 T cell ratios increased significantly from 2 days post MHV-3 infection in C3H/Hej mice,and CD3 CD4 CD8-,CD3 CD4-CD8 ,CD3 CD4 CD25-and CD3 CD4-CD25 T cell ratios decreased accordingly.In conclusion,the changes of virus titer and pathology in the livers of C3H/Hej mice post MHV-3 suggest their contribution to viral persistence.Further characterizations of DN Treg cells are that infection indicates that MHV-3 could induce the chronic inflammation in livers of C3H/Hej mice.The increase of the DN Treg cell and CD4 CD25 T cell ratios in C3H/Hej mice post MHV-3 infection suggests that DN Treg cells and CD4 CD25 T cells may both have important suppressive immunomodulation functions in the development of chronic viral hepatitis and have important roles in the virus persistent infection.Further characterizations of DNT cell and CD4 CD25 T cell are under investigation.
