根霉葡萄糖淀粉酶的简易纯化法及部分理化性质 总被引:6,自引:0,他引:6
从汾酒大曲中分离到一株根霉,经诱变处理后获得了具有高产葡萄糖淀粉酶的变异株。将此菌株用固体麸曲培养,水抽提得原酶液。经DEAE-Sephadex A—50、Sephadex G—100和CM—Cellulosc C52三步柱层析后,得到了紫外光谱和SDS-聚丙烯酰胺凝胶电泳皆为均一的葡萄糖淀粉酶。总活力回收达70%左右,比活力提高23.70倍。该酶的A1%280nm=14.70,经SDS—聚丙烯酰胺凝胶电泳和凝胶过滤层析表明其分子量约为78400,酶分子中赖氨酸含量较高,属典型的根霉型葡萄糖淀粉酶。 相似文献

本文研究了食用菌深层发酵金针菇菌丝体多糖的提取条件。实验表明,在加水比为1:6,沸水提取4小时,进行三次抽提后,用4倍乙醇沉淀所获粗多糖得率最高。粗多糖经脱色去蛋白后以70%(V/V)乙醇分级可获得最高级分精制多糖。 相似文献

The meridian phenomena are closely associated with the neuro-muscular function. Our experiments show that meridian phenomenon,
acupoint stimulation-induced generating-sensations along meridians (GSM), does exist in some volunteers, and the subjects
can describe the characteristics of the feelings and the route of GSM. Simultaneously, GSM is often accompanied with progressive
myoelectric activity along the meridian which is able to be recorded objectively. Brachial plexus anesthesia and regional
neuro-muscular block may arrest these responses while the neuro-muscular blocking agent may stop neuromuscular transmission
at the motor end-plate rather than check GSM. The facts reveal that GSM probably depends on muscular activity and is involved
in the secondary neuro-muscular excitation propagating along the longitudinal axis of muscles. These findings suggest that
the myoelectric activities constitute the foundation of secondarily generating-sensation propagating along the longitudinal
orientation of limbs (muscles), or along the meridian. 相似文献

建立云南与四川僵蚕药材的总蛋白质指纹图谱及分级指纹图谱,对比分析以发现其中的差异表达蛋白质分子并进行鉴定及生物信息学分析,探索其用于产地鉴定的可行性。提取两种僵蚕的酸溶、碱溶、水溶及醇溶性总蛋白质,对其进行丙酮分级沉淀,对所得样品进行SDS-PAGE指纹图谱对比分析,对所得差异条带进行LC-MS/MS分析,数据库检索后进行生物信息学分析。两种僵蚕的总蛋白质指纹图谱相似度很高,难以据此进行产地鉴定;两种僵蚕的分级指纹图谱则有较显著差异,有望用于产地鉴定;依据分级指纹图谱差异共鉴定出β-葡萄糖苷酶、抗胰凝乳蛋白酶、抗胰蛋白酶、含脂肪酶结构域蛋白等273种蛋白质。云南与四川僵蚕的蛋白质组成及其分级指纹图谱存在显著差异,有望用作产地鉴定的分子依据;醇溶蛋白质构建分级指纹图谱可以提供更加丰富的信息及更好的分辨率;分级指纹图谱技术具有简便易行、分辨率较高的优点,为其他中药材的产地鉴定研究提供了方法学思路。 相似文献

目的研究临床多重耐药铜绿假单胞菌群体感应(QS)系统与主动外排泵MexAB-OprM系统基因表达水平与抗生素耐药关系。方法收集苏州市立医院和上海市江湾医院2011年2月至6月间临床标本中分离的铜绿假单胞菌,定量分析细菌生物被膜形成能力;MIC法检测细菌抗生素耐药性,用多重聚合酶链反应(PCR)扩增群体感应系统lasI、lasR及主动外排泵系统mexA基因,实时定量逆转录RT-PCR检测lasI、lasR和mexA基因的相对表达量。结果临床样本分离出84株铜绿假单胞菌,其中产生物被膜菌58株,占比69%;多重耐药菌共24株,占比28.6%;多重耐药菌株中产生物被膜有11株,占45.8%;多重耐药菌中mexA基因表达上调有18株,占75%;lasI基因表达上调有8株,占33.3%。结论多重耐药菌株的生物被膜形成率显著低于非多重耐药组,多重耐药铜绿假单胞菌的主动外排泵MexAB-OprM系统基因表达出现显著上调,生物被膜菌的lasI基因表达显著上调而lasR基因的表达无明显变化。 相似文献
