Disease resistance, stress response and effects of triploidy in growth hormone transgenic coho salmon 总被引:2,自引:0,他引:2
Diploid and triploid coho salmon Oncorhynchus kisutch transgenic for growth hormone (GH) and control coho salmon were compared for differences in disease resistance and stress response. Resistance to the bacterial pathogen Vibrio anguillarum was not affected in transgenic fish relative to their non‐transgenic counterparts when they were infected at the fry stage, but was lower in transgenic fish when infected near smolting. Vaccination against vibriosis provided equal protection to both transgenic and non‐transgenic fish. Triploid fish showed a lower resistance to vibriosis than their diploid counterparts. Diploid transgenic fish and non‐transgenic fish appeared to show similar physiological and cellular stress responses to a heat shock. These studies provide information useful for both performance and ecological risk assessments of growth‐accelerated coho salmon. 相似文献

Triploid intersexes homozygous for a mutant (msl-2) known to impede the hyperactivation of the X chromosome in diploid males differentiate into adults, sexually indistinguishable from their heterozygous sibs. A shift toward female sexual differentiation mediated by manipulating the rearing temperature is accompanied by an apparent increase in the level of an X-linked gene product. This unexpected result is rationalized in terms of differential lethality of individuals at the two extremities of the distribution of X-activity levels in intersexes raised at a particular temperature. No evidence of a mosaicism comparable to the sexual mosaicism exhibited could be found with respect to an X-linked gene product in triploid intersexes. 相似文献

Comparative morphological, karyological and chorological studies prove the distinctness of the SE European and Asiatic populations ofPulmonaria mollis s.l., to be treated asP. dacica (Figs. 4, 5).P. mollis s.str. is interpreted as a relatively young taxon, which probably has evolved from an older stock ofP. dacica ancestors during the Pleistocene (Figs. 6, 8) and might have migrated, perhaps with oak woodland communities, from SE to C. Europe. A similar evolution can be postulated for S—SE European dysploidP. rubra-carnica-stiriaca-vallarsae group; this may have involved still unknown tetraploids (P. carnica?, Fig. 7). This group is linked via the hypertriploid speciesP. vallarsae with theP. saccharata group and at least the eastern species of theP. australis group. The monotypic genusParaskevia apparently marks the earliest divergence from the common Tertiary ancestral stock (Fig. 8). We manufactured a new fermented tea by tea-rolling processing of third-crop green tea (Camellia sinensis) leaves and loquat (Eriobotrya japonica) leaves. The mixed fermented tea extract inhibited pancreatic lipase activity in vitro, and effectively suppressed postprandial hypertriacylglycerolemia in rats. Rats fed a diet containing 1% freeze-dried fermented tea extract for 4 weeks had a significantly lower liver triacylglycerol concentration and white adipose tissue weight than those fed the control diet lacking fermented tea extract. The activity of fatty acid synthase in hepatic cytosol markedly decreased in the fermented tea extract group as compared to the control group. The serum and liver triacylglycerol- and body fat-lowering effects of the mixed fermented tea extract were strong relative to the level of dietary supplementation. These results suggest that the new fermented tea product exhibited hypotriacylglycerolemic and antiobesity properties through suppression of both liver fatty acid synthesis and postprandial hypertriacylglycerolemia by inhibition of pancreatic lipase. 相似文献

江苏东山山地枇杷果实品质与果园土壤、叶片和果实矿质元素的关系 总被引:1,自引:0,他引:1
以江苏省苏州吴中区山地种植的24个枇杷果园的‘白玉’枇杷为研究对象,采用典型相关分析法研究土壤、叶片和果实矿质元素间及其与果实品质指标的相关性,应用多元线性回归分析法筛选影响枇杷果实品质的土壤、叶片和果实的主要矿质元素和影响叶片矿质元素的主要土壤矿质养分因子,为改善枇杷果实品质的果园营养管理技术提供理论参考依据。结果表明:(1)枇杷果实品质指标在不同果园间存在较大差异。(2)枇杷果实品质主要受其叶片和果实矿质元素含量影响,其中:枇杷单果质量与叶片中N含量呈显著正相关,与果实中N含量呈显著负相关;可溶性固形物含量与叶片中N和Mg元素含量呈极显著正相关,与果实中P、Mn元素含量呈极显著正相关,与果实中Ca、Mg元素含量呈极显著负相关;可滴定酸含量与叶片中N和Zn元素含量呈显著负相关,与果实中N、Mn元素含量呈显著负相关,与果实中Ca元素含量呈显著正相关。(3)果实品质指标与土壤矿质养分含量的相关性较弱,土壤矿质养分主要通过影响叶片矿质元素含量,进而间接影响果实品质,其中土壤中P、K、Ca、Mn、Cu元素对叶片中矿质元素含量影响较大。研究认为,提高土壤中P、K、Zn含量并减少土壤Ca含量是东山山地枇杷果园土壤管理的关键技术途径;提高叶片中N含量以及果实中N、P、Mn含量和减少果实中Ca、Mg含量是改善东山山地枇杷果实品质的关键措施。 相似文献

以'青种'、'霸红'和'鸡蛋白'3个枇杷品种为材料,测定不同果实发育时期果实中蔗糖、葡萄糖和果糖含量以及蔗糖代谢相关酶即酸性转化酶(AI)、中性转化酶(NI)、蔗糖合成酶(SS)和蔗糖磷酸合成酶(SPS)的活性,并对果实中糖积累与酶活性的关系进行了分析.结果表明:在果实膨大期(5月3日)之前,3种枇杷果实的蔗糖、葡萄糖和果糖积累缓慢,之后则迅速积累,存在着明显的转折点;果实成熟(5月23日)之后糖分积累速度趋于平稳.3种枇杷果实在发育过程中转化酶、蔗糖合成酶和蔗糖磷酸合成酶的活性变化与3种糖积累的动态变化趋势相一致.NI和AI活性在果实膨大期之前都较低且没有明显的变化,之后均快速上升;SS和SPS的活性在果实膨大期之前都很低且几乎无变化,随后'鸡蛋白'的活性迅速上升至果实成熟之后便缓慢上升,而'青种'和'霸红'随果实成熟度的增加而升高,但均低于'鸡蛋白'.可见,枇杷果实膨大期是糖分积累代谢活跃期,其糖积累受蔗糖代谢相关酶综合调控. 相似文献

Summary Direct shoot and cormlet regeneration from leaf explants were obtained in triploid dessert banana cultivar Nanjanagud Rasabale
(NR) that is classified under the group ‘Silk’ and has the genotype AAB. The response for both cormlet and direct shool formation
was observed only in leaf explants obtained from shoots cultured in liquid medium but not in similar explants obtained from
shoots grown on gelled medium. Shoot initiation occurred after a sequential culture of leaf (sheath) explants on modified
Murashige and Skoog (MS) medium supplemented with different growth regulators. In the sequence, the leaf explants were cultured
first on medium with a high level (22.4 μM) of benzyladenine (BA), second on indolc-3-butyric acid (IBA) supplemented medium, and third on reduced BA medium under incubation
in the dark. The highest adventitious shoot regeneration in 24% of the explants, with the number of shoots ranging from 2
to 3 per explant, occurred in the explants incubated at the first step in medium with 22.4 and 0.198 μM IBA. Further growth and complete shoot formation occurred under incubation in a 16-h photoperiod. While keeping the culture
conditions constant and replacing BA with picloram (0.83–20.71 μM) in the initial step, adventious origin of cormlets occurred in 12% of the explants. However, when rhizome explants (also
obtained from shoots grown in liquid medium) were cultured with various growth regulators in the first step, medium containing
2,4,5-trichlorophenoxyacctic acid (7.82 μM) produced friable callus that re-differentiated into roots only. Physical forms of the medium, ie.e. agar-gelled or liquid,
imparted specific effects on the extent of multiplication of leaf-regenerated shoots with no differences in morphology and

The histological and ultra-structure of the pituitary in diploid red crucian carp(Carassius auratus red var.),triploid crucian carp and allotetraploid hybrids within and after the breeding season were comparatively studied.The result showed that there were six endocrine cell types in the pituitary of these three kinds of fishes,and there was an obvious difference in cell size among different ploidy level fishes.As for the same type of pituitary cells,the cell size was increased gradually with the in- creasing ploidy level.In the breeding season,the allotetraploid hybrids had higher proportion of go- nadotropin cells(GTH)than triploids,and the triploids had higher proportion of GTH than diploids.The results were related to the earlier sexual maturity of allotetraploid hybrids and sterility of triploid cru- cian carp.On the other hand,among the three kinds of fishes,the proportion of somatotropin(STH) cells in triploids crucian carp was the highest,whereas that in allotetraploid hybrids was the lowest. The results might be connected with the faster growth rate of triploids and slower growth rate of al- lotetraploid hybrids.In addition,in GTH cells of meso-adenohypophysis after the breeding season, there were many endocrine particles in triploids,while those endocrine particles were released from the cells in allotetraploids and diploids.This result showed that the sterility of triploid crucian carp might be related to the hormone which was not released from the GTH cells.In a word,the present study indicated that the differences in the structure of pituitary among different ploidy level fishes contributed to their difference in the growth rate and gonadal development. 相似文献

Survival and escape of Agrobacterium tumefaciens in triploid hybrid lines of Chinese white poplar transformed with two insect-resistant genes 下载免费PDF全文
Two partly modified insect-resistant genes (BtCryI Ac gene [Bt gene toxin against Lepidopterean insects] and API gene [arrowhead proteinase inhibitor]) were transferred to the triploid hybrid of Chinese white poplar ((Populus tomentosa Carr. × Populus bolleana Louche) × Populus tomentosa Carr.) mediated by A. tumefaciens. The survival of Agrobacterium in transgenic plants was examined during the processes of transplanting and subculturing on the nutrient medium. The results suggested that 80% of the plants, which were obtained by repeated selection on media added with 50 mg/L kanamycin and 300 mg/l carbenicillin, showed positive reactions after examination using molecular methods. The ELISA test indicated that the Bt toxoprotein was expressed in seven of the transgenic sub-clones. Leaves, stems, and roots of all the 28 transgenic plants were cultured on the YEB medium added with 50 mg/L kanamycin, and it was found that Agrobacterium survived in three sub-clones (33, 37, 5) and could have existed for 24 months in the bottle. These three transgenic sub-clones were transplanted and cultivated for one month in the room, and then the target Agrobacterium was found in rhizosphere of the sub-clone 33. 相似文献
